Primary lymphoma of the nasopharynx is uncommon and is more commonly seen as an association with other primary pathologies. Nasopharyngeal lymphoma is an aggressive tumor and can affect all age groups but is more commonly seen in middle-aged and older individuals. Majority of these tumors belong to the non-Hodgkin's group. Cavernous sinus syndrome (CSS) is a rare but severe complication indicating advanced aggressive stage of disease with direct extension of tumor into the cavernous sinus. Our patient is a young teenage 18-years old presented to emergency department with the symptoms of left j

Early relapse of diffuse large B-cell lymphoma (DLBCL) within 12 months of diagnosis confers a poor prognosis, underscoring the critical need for accurate pretreatment prediction. Current approaches relying on radiomics and clinical indicators are limited by scarce medical imaging data, time-consuming manual feature extraction (susceptible to human subjectivity), and insufficient use of multimodal deep learning frameworks. To address these challenges, we propose MM-DLBCL, a novel multimodal transfer learning framework for pretreatment 12-month disease-free survival (DFS12) prediction. It lever

BACKGROUND: Extranodal (EN) involvement is common in diffuse large B-cell lymphoma (DLBCL), but current prognostic models inadequately capture the anatomical complexity of EN dissemination. We evaluated primary multifocal extranodal DLBCL (PMEN-DLBCL) as a distinct clinical and anatomical phenotype using a standardized lesion-level classification framework.
METHODS: We retrospectively analyzed 393 adults with newly diagnosed DLBCL treated with rituximab-based chemoimmunotherapy between 2003 and 2025. Patients were classified as primary nodal (PN; n = 231), primary focal extranodal (PFEN; n =

Primary central nervous system diffuse large B-cell lymphoma (PCNS-DLBCL) is a rare form of non-Hodgkin lymphoma with a notably poor prognosis. This study aimed to identify new prognostic factors for improved prognostic stratification in PCNS-DLBCL. A total of 85 PCNS-DLBCL cases from three hospitals were retrospectively analyzed. Disease progression within 24 months (POD24) and its risk factors were investigated, and a prognostic model for predicting POD24 was constructed. The median overall survival (mOS) and median progression-free survival (mPFS) for the entire cohort were 48.54 months and

Background/Objectives: Diffuse large B-cell lymphoma (DLBCL) is molecularly heterogeneous, and approximately 30-50% of patients fail to achieve cure with standard R-CHOP. Genotype-directed first-line therapy may improve outcomes by targeting subtype-specific oncogenic pathways. This study evaluated the feasibility, efficacy, and safety of a molecularly adapted R-CHOP-X strategy with two-year follow-up. Methods: In this single-center, prospective, non-randomized study conducted between September 2023 and the data cut-off (16 September 2025), 43 adults with newly diagnosed DLBCL (excluding high-

We conducted a retrospective, multi-center, real-world study to evaluate efficacy and safety of zanubrutinib combination therapy as first-line treatment for diffuse large B-cell lymphoma (DLBCL) between August 2020 and August 2025. Patients received RCHOP as standard therapy, while those with central nervous system (CNS) involvement were treated with R plus high-dose methotrexate (R + HD-MTX). Seventy patients were enrolled: 53 (75.7%) received RCHOP and17 (24.3%) received R + HD-MTX. The complete response rate (CRR) was 88.6% and the overall response rate (ORR) was 92.9%. Salvage chemotherapy and autologous stem cell transplantation (ASCT) offer the opportunity to cure eligible patients with relapsed diffuse large B-cell lymphoma (DLBCL). Epigenetic alterations such as aberrant DNA methylation patterns have been linked to chemotherapy resistance in DLBCL. Oral Azacitadine (AZA) is a hypomethylating agent that inhibits DNA methyltransferase and has provided evidence of chemotherapy sensitization in DLBCL. In this phase I trial the safety and feasibility of two dose levels of AZA were investigated in combination with standard cytotoxic chemotherapy rituximab, ifo

Rituximab, an anti-CD20 monoclonal antibody, is a cornerstone of therapy for diffuse large B-cell lymphoma (DLBCL), yet a substantial proportion of patients develop treatment failure. While loss of CD20 expression has been considered a major mechanism of resistance, additional mechanisms limiting effective antibody binding remain incompletely understood. We hypothesized that circulating CD20-positive large extracellular vesicles (EVlarge) act as antigen sinks that sequester rituximab and attenuate its therapeutic activity. Circulating CD20-positive EVlarge were quantified from archived serum s

Intussusception is predominantly a pediatric condition, with only about 5 % of all intussusception cases occurring in adults with primary intestinal lymphomas comprising 1 %-4 % of gastrointestinal malignancies, 90 % of which are B-cell non-Hodgkin lymphomas (NHL). Diffuse large B-cell lymphoma (DLBCL), the most common subtype of NHL, often leads to intussusception without classical B symptoms. We present the case of a 33-year-old male with ileocolonic intussusception caused by a 4.8 × 5.3 × 5.5 cm enhancing mass. Histopathology and FISH confirmed DLBCL without MYC, BCL2, or BCL6 rearrangement

Primary non-Hodgkin's lymphoma is a highly prevalent haematologic malignancy but contrarily, primary lymphoma of the female genital tract (PLFGT) is extremely rare. The most common histological subtype of PLFGT is diffuse large B-cell lymphoma (DLBCL), which most commonly arises from the ovary, cervix, uterus or vagina. It is rarer still for DLBCL to arise from the fallopian tube, and therefore, we present the second case ever reported. A woman in her 30s presented with abdominal pain and a pelvic mass on CT. The mass was laparoscopically excised, which allowed the histological diagnosis to be

B-cell lymphoma 6 (BCL6) is an attractive drug target for diffuse large B-cell lymphoma (DLBCL). This study aimed to create a lipid nanoparticle (LNP)-based peptide-proteolysis-targeting chimera (PROTAC), specifically BCL6-PROTAC, to develop effective strategies for treating DLBCL via targeted degradation of the BCL6 protein. Molecular docking, SPR and cellular thermal shift assays revealed that F1324 (Ac-LWYTDIRMSWRVP-OH) is a high-affinity BCL6-binding peptide. PROTAC LNPs were synthesized by modifying F1324 and pomalidomide aptamers onto LNPs via a covalent chemical reaction in a certain pr

Diffuse large B-cell lymphoma (DLBCL) frequently involves extranodal sites, most commonly the gastrointestinal tract; however, predominant colorectal involvement and very late relapse after long-term remission are uncommon. A 70-year-old woman with a history of DLBCL in complete remission for more than 10 years after 6 cycles of R-CHOP presented with progressive weight loss, early satiety, lower abdominal pain, and new-onset urinary urgency and frequency. Cross-sectional imaging revealed a large cecal mass with retroperitoneal extension causing right-sided hydronephrosis. Colonoscopy demonstra

R-GDP has been established as an effective salvage treatment for Rel/Ref DLBCL. We aimed to clarify the efficacy of R-GDP therapy. We included 41 consecutive patients with Rel/Ref DLBCL, who received R-GDP therapy as salvage chemotherapy at our hospital between January 2014 and August 2024. Thirty-three patients received R-GDP therapy as a 2nd-line regimen, whereas 8 received R-GDP as a 3rd or later-line regimen. The ORR was 70.7%. 23 out of 25 relapsed patients (92%) responded to R-GDP therapy, whereas only 6 out of 16 refractory patients responded (37.5%). With regard to the duration of resp

BACKGROUND: Late-onset neutropenia (LON) and hypogammaglobulinemia are recognized sequelae of rituximab therapy in B-cell non-Hodgkin lymphoma, and both may leave patients vulnerable to infection once treatment has ended.
METHODS: Fifty-three patients with newly diagnosed, unfavorable diffuse large B-cell lymphoma (DLBCL) who entered remission on dose-adjusted (DA) R-EPOCH immunochemotherapy were retrospectively evaluated. Neutropenia (absolute neutrophil count < 1.5 × 109/L, CTCAE-graded), hypogammaglobulinemia and infections were registered at treatment completion and 6 and 12 months later.

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is the most prevalent type of aggressive lymphoma, commonly spreading to sites such as the lymph nodes, spleen, bone marrow, liver, lungs, and central nervous system. However, metastasis to the heart and ovaries is relatively uncommon.
CASE DESCRIPTION: A 63-year-old woman visited the hospital with abdominal pain and bloating, but showed none of the typical signs of lymphoma. Imaging scans revealed abnormal masses in both the pericardium and ovaries. A biopsy confirmed it was DLBCL, presenting in the rare form of simultaneous spread to the hea

Diffuse large B-cell lymphoma is a prevalent subtype of adult non-Hodgkin lymphoma; noted for its biological and clinical variability. Background and Objectives: This study seeks to assess the expression and prognostic implications of Ki-67, MYC, and BCL2 utilising immunohistochemistry on a cohort of Romanian patients diagnosed with DLBCL while also addressing the limitations imposed by the absence of fluorescence in situ hybridisation testing in resource-constrained settings. Materials and Methods: A single-centre, retrospective study involved 66 cases of formalin-fixed, paraffin-embedded tis

Diffuse large B-cell lymphoma (DLBCL) is an aggressive malignancy characterized by a poor prognosis, mainly caused by late diagnosis and the absence of effective treatment options. Malignant transformation of B-cells emphasizes the necessity of novel biomarkers for early diagnosis and prognosis. Dysregulation of long non-coding RNAs (lncRNAs), fundamental regulators in biological pathways, is linked to the development of different cancer types, including DLBCL. In this study, we assessed lncRNA HOTAIR expression in DLBCL and its association with clinicopathological features while investigating

Diffuse large B-cell lymphoma (DLBCL) is a clinically and molecularly heterogeneous disease. The increasing recognition and targeting of genetically defined DLBCLs highlight the need for robust classification algorithms. We previously characterized recurrent genetic alterations in DLBCL and identified 5 discrete subtypes, clusters 1 to 5 (C1-C5), with unique mechanisms of transformation, immune evasion, candidate treatment targets, and different outcomes after standard first-line therapy. Herein, we validate the C1 to C5 DLBCL taxonomy in an independent data set and use the expanded series of
