Ameba-inspired strategy enhances probiotic efficacy via prebound nutrient supply.

Nutrient competition with indigenous microbes or pathogens presents a significant challenge for oral probiotic efficacy. To address this issue, we develop an ameba-inspired food-carrying strategy (AIFS) by prebinding ginger-derived exosome-like nanoparticles (GELNs) onto probiotics as food depots. AIFS enables probiotics to efficiently and exclusively consume GELNs in situ, even in the presence of competing bacteria. This results in up to 21 times higher uptake efficiency compared to unengineered probiotics, dramatically accelerating probiotic proliferation. Meanwhile, AIFS potentiates probiot…

Engineered Probiotic-Powered Micro-Rod Robot Bomb for Thrombus-Penetrating Explosion.

Conventional thrombolytic agents demonstrated limited efficacy in treating thrombotic disorders characterized by narrow therapeutic windows and progressive vascular injury, lacking the required precision, timeliness, and treatment durability. Here an engineered probiotic powered micro-rod robot for targeted and penetrative treatment for thrombus is developed. This micro-rod robot (SrEcNPL) using natural probiotics as bio-carriers, functionalized with thrombolytic carbon nanotubes and platelet membrane-coated nanoparticles loaded with targeted vasodilators. The robot exploits the rapid movement…

The effect of probiotic supplementation on constipation in patients with Parkinson's disease: A randomized controlled trial.

Background: Constipation is a common non-motor symptom (NMS) in Parkinson's disease (PD), affecting up to 70% of patients and reducing quality of life (QOL). Probiotics may improve bowel function via gut microbiota modulation and gut-brain axis regulation. This randomized, double-blind, placebo-controlled trial evaluated the efficacy of six-week multi-strain probiotic supplementation on constipation in Iranian patients with PD. Methods: Seventy-two patients with PD (aged 50-80 years, functional constipation per Rome IV criteria) were randomized 1:1 to probiotics or placebo. The probiotic capsu…

Alkalihalobacillus clausii bacteremia after probiotic use for acute diarrhea: a case report.

Alkalihalobacillus clausii (previously known as Bacillus clausii) is a spore-forming bacterium used as a probiotic due to its resistance to gastric acidity and intestinal conditions. It is marketed for managing acute diarrhea, though evidence of its efficacy and safety is limited. We present the case of an 87-year-old man with acute viral diarrhea treated with Alkalihalobacillus clausii, who developed septic shock with blood cultures confirming Alkalihalobacillus clausii bacteremia. Intensive care and daptomycin therapy led to clinical resolution.

Engineered Probiotic Hydrogel Alleviates Vaginitis via Antimicrobial and Anti-inflammatory Pathways.

Vulvovaginal candidiasis (VVC), primarily caused by Candida albicans, is a prevalent infection that presents significant clinical challenges. Current antifungal treatments have limitations, including a narrow antimicrobial spectrum and a lack of anti-inflammatory effects. To address these limitations, we genetically modified Bacillus subtilis to produce high-molecular-weight hyaluronic acid (HMHA) for sustained local delivery of antimicrobial peptides (AMP) and HMHA at the infection site. The engineered Bacillus subtilis (B. sub@HMHA) hydrogel exhibited both antifungal and vaginal microbiome-m
