Identification of Virus-Host Interactions.

No protein acts on its own to carry out its designated function. Proteins interact with other biomolecules to form complexes that perform a coordinated function. While proteins most often form complexes involving other proteins, they also interact with nucleic acids, lipids, carbohydrates, ions, and other small molecules and metabolites (Wang et al., Proteomics 22(15-16):e2100190, 2022; Veenstra, Proteomics 21:e2000235, 2021). While the early days of proteomics were focused on identifying and quantifying as many proteins as possible, scientists quickly recognized the power of proteomics to rap

Identification of interbacterial adhesion between oral Schaalia species and streptococcal serine-rich repeat glycoproteins.

Interbacterial adhesion is central to multispecies biofilm formation. Streptococcal serine-rich repeat glycoproteins (SRRPs) play a well-defined role in oral colonization, yet SRRP involvement in interbacterial adhesion remains understudied. Here, we aimed to isolate SRRP-dependent adhesion partners of Streptococcus gordonii Challis (DL1) from human oral microbiota. Of 179 isolates screened, 23 coaggregated with DL1 but not with an isogenic SRRP-encoding gene deletion mutant. Whole-genome sequencing identified isolates as 19 Schaalia odontolytica, 3 Schaalia sp. HMT-172, and 1 non-Schaalia mix

Generation and Characterization of HDV-Specific Antisera with Respect to Their Application as Specific and Sensitive Research and Diagnostic Tools.

The hepatitis D virus (HDV) is a small, defective RNA virus that induces the most severe form of viral hepatitis. Despite its severity, HDV infections are under-diagnosed due to non-standardized and costly diagnostic screening methods. However, limited research has been conducted on characterizing HDV-specific antibodies as alternative tools for diagnosis. Thus, we generated HDV-specific, polyclonal antibodies by immunizing rabbits with the HDV protein, small hepatitis delta antigen (SHDAg), in its oligomeric or denatured form. We identified SHDAg-specific linear epitopes by peptide array anal

In vitro monitoring of amyloid β-peptide oligomerization by Electrospray differential mobility analysis: An alternative tool to evaluate Alzheimer's disease drug candidates.

Alzheimer's disease (AD) is a neurodegenerative disorder linked to protein aggregation, like more than twenty other human pathologies. One major protein incriminated in AD is the 42-residue amyloid-β peptide (Aβ1-42) which aggregates to form neurotoxic oligomers and fibrils. While, low molecular weight oligomers have been evidenced as neurotoxic species, only scarce methods raise the challenge to monitor the beginning of the aggregation process, called oligomerization. We propose here an innovative and fast monitoring of the time-dependent Aβ1-42 oligomerization pattern by electrospray differe

Novel plasminogen gene mutations in Turkish patients with type I plasminogen deficiency.

The plasminogen (Plg) protein is the inactive proenzyme form of plasmin that dissolves fibrin thrombi by a process called fibrinolysis. It has been shown that homozygous or compound-heterozygous deficiency of this protein is a major cause of a rare inflammatory disease affecting mainly mucous membranes found in different body sites. In this study, five individual Turkish patients and nine Turkish families with type 1 Plg deficiency were investigated for PLG gene mutations. All of the coding regions of the PLG gene mutations were screened for mutations using denaturing high-pressure liquid chro

Expression of CTB-10×rolGLP-1 in E. coli and its therapeutic effect on type 2 diabetes.

Glucagon-like peptide-1 (GLP-1) is a short peptide that can significantly reduce blood glucose level. Recombination oral long-acting glucagon-like peptide-1 (rolGLP-1), is a GLP-1 analog generated from site-specific mutation of GLP-1. CTB is a non-toxic portion of the cholera toxin and an ideal protein antigen carrier. In this study, we firstly constructed a vector pET-22b (+)-CTB-10×rolGLP-1 to express a fusion protein composed of CTB and ten tandem repeated rolGLP-1 in BL21 (DE3) line of E. coli. The CTB-10×rolGLP-1 was expressed efficiently in the inclusion bodies. The expression product wa

Impact of trichloroethylene exposure on the microbial diversity and protein expression in anaerobic granular biomass at 37°C and 15°C.

Granular biomass from a laboratory-scale anaerobic bioreactor trial was analysed to identify changes in microbial community structure and function in response to temperature and trichloroethylene (TCE). Two bioreactors were operated at 37°C, while two were operated at 15°C. At the time of sampling, one of each temperature pair of bioreactors was exposed to process failure-inducing concentrations of TCE (60 mg L(-1)) while the other served as a TCE-free control. Bacterial community structure was investigated using denaturing gradient gel electrophoresis (DGGE) and 16S rRNA gene clone library an

A parasite vector-host epidemic model for TSE propagation.

BACKGROUND: Transmissible spongiform encephalopathies (TSEs) are a family of diseases that infect mammals. They are explained by cross-contamination through an unknown route or from infection of food contaminated with prion proteins (PrPs), natural proteins that take an infectious form contributing to the slow destruction of the animal brain. While the extreme resistance of PrPs to denaturation and proteolysis accounts for a route from the mouth to the brain, the possible role of another route of contamination is explored here. Many diseases are spread by vectors, as seen in plague, typhus, ma

Papaya glutamine cyclotransferase shows a singular five-fold beta-propeller architecture that suggests a novel reaction mechanism.

Cyclisation of N-terminal glutamine and/or glutamate to yield pyroglutamate is an essential posttranslational event affecting a plethora of bioactive peptides and proteins. It is directly linked with pathologies ranging from neurodegenerative diseases to inflammation and several types of cancers. The reaction is catalysed by ubiquitous glutaminyl cyclotransferases (QCs), which present two distinct prototypes. Mammalian QCs are zinc-dependent enzymes with an alpha/beta-hydrolase fold. Here we present the 1.6-A-resolution structure of the other prototype, the plant analogue from Carica papaya (P

Activation of deoxycytidine kinase in lymphocytes is calcium dependent and involves a conformational change detectable by native immunostaining.

Deoxycytidine kinase (dCK), the principal deoxynucleoside salvage enzyme, plays a seminal role in the bioactivation of a wide array of cytotoxic nucleoside analogues. Recently, activation of dCK has been considered as a protective cellular response to a number of DNA-damaging agents in lymphocytes. Regarding the molecular mechanism of the enzyme activation, a post-translational modification by protein phosphorylation has been suggested. Here we provide evidence that both the activation process and the maintenance of the activated state require free cytosolic calcium. BAPTA-AM, a cell-permeable

Nonprecipitating anti-La(SS-B) autoantibodies in primary Sjögren's syndrome.

Anti-La(SS-B) precipitin-negative sera show a restricted epitope recognition and can be easily overlooked in routine laboratory testing. We have therefore determined the prevalence of nonprecipitating anti-La(SS-B) antibodies in patients with primary Sjögren's syndrome and studied their clinical and immunological associations. Clinical details were obtained from 68 patients with primary Sjögren's syndrome, and serum samples were examined by enzyme-linked immunosorbent assay using purified recombinant La, 60-kDa Ro, and 52-kDa Ro proteins and by counterimmunoelectrophoresis. Thirteen patients (

Studies of the metabolism of asialotransferrins: the metabolic heterogeneity of human asialotransferrin.

Catabolism of human transferrin and human asialotransferrin was simultaneously studied in guinea pigs by means of total body radioactivity measurements. Total body activity representing transferrin decreased at a constant rate with an average half-life of 88 h. Decrease of the total body activity representing asialotransferrin exhibited at least two rates; the half-life of the fast initial component averaged at 25 h, whereas the half-life of the slower component averaged at 55 h. Transition occurred between the 50th and 80th hours of the experiments. The complex character of the elimination cu

Class sigma glutathione transferase unfolds via a dimeric and a monomeric intermediate: impact of subunit interface on conformational stability in the superfamily.

Solvent-induced equilibrium unfolding of a homodimeric class sigma glutathione transferase (GSTS1-1, EC 2.5.1.18) was characterized by tryptophan fluorescence, anisotropy, enzyme activity, 8-anilino-1-naphthalenesulfonate (ANS) binding, and circular dichroism. Urea induces a triphasic unfolding transition with evidence for two well-populated thermodynamically stable intermediate states of GSTS1-1. The first unfolding transition is protein concentration independent and involves a change in the subunit tertiary structure yielding a partially active dimeric intermediate (i.e., N2 left and right a

Characterization of an immuno-dominant antigen in Brucella ovis and evaluation of its use in an enzyme-linked immunosorbent assay.

A panel of 45 Brucella ovis serologically positive sera were tested in immunoblots against B. ovis outer membrane proteins Omp31 and Omp25, purified by preparative SDS-gel electrophoresis. Forty-three sera reacted with Omp31, while only 11 reacted with Omp25, suggesting that Omp31 is identical to the previously reported immuno-dominant 29-kDa protein. Attempts to purify Omp31 on a larger scale by using procedures such as ion exchange-, reversed phase-, affinity- and gel filtration chromatography suggested that the outer membrane proteins were aggregated with rough lipopolysaccharide. Allergic sensitization to native and heated celery root in pollen-sensitive patients investigated by skin test and IgE binding.

The rates of sensitization and allergy to four birch pollen related plant foods were investigated in a group of 167 patients who were sensitive to at least one kind of pollen and one particular food. Sensitivity was concluded from a positive skin prick test or the determination of specific IgE, whereas allergy was based on anamnestic data. The positivity rates for sensitization and allergy, respectively, were: apple, 93 and 84%; hazelnut, 90 and 78%; celery, 70 and 14%; carrot, 60 and 37%. Context-dependent dysregulation of transcription by mutant huntingtin.

Huntington disease (HD) is an adult-onset neurodegenerative disease caused by expansion of a polyglutamine (poly(Q) tract in the N-terminal region of huntingtin (htt). Although the precise mechanisms leading to neurodegeneration in HD have not been fully elucidated, transcriptional dysregulation has been implicated in disease pathogenesis. In HD, multiple N-terminal mutant htt fragments smaller than the first 500 amino acids have been found to accumulate in the nucleus and adversely affect gene transcription. It is unknown whether different htt fragments in the nucleus can differentially bind

Native gel electrophoresis and isoelectric focusing of a 64-kilodalton eye muscle protein shows that it is an important target for serum autoantibodies in patients with thyroid-associated ophthalmopathy and not expressed in other skeletal muscle.

Although sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blotting are widely used to detect serum antibodies in patients with autoimmune disorders, this procedure unfolds and denatures proteins and may alter antibody-binding sites. We have used a gentle protocol for the preparation and purification of a 64-kilodalton (kDa) eye muscle (EM) membrane antigen associated with thyroid-associated ophthalmopathy (TAO) for use as antigen in immunoblotting. Pig EM membrane proteins were prepared from crude homogenates by high speed centrifugation and solubilized by hand

The role of the C-terminus of human α-synuclein: intra-disulfide bonds between the C-terminus and other regions stabilize non-fibrillar monomeric isomers.

Substantial evidence implicates that the aggregation of α-synuclein (αSyn) is a critical factor in the pathogenesis of Parkinson's disease. This study focuses on the role of αSyn C-terminus. We introduced two additional cysteine residues at positions 107 and 124 (A107C and A124C) to our previous construct. Five X-isomers of oxidative-folded mutation of α-synuclein with three disulfides were isolated and their secondary structures and aggregating features were analyzed. All isomers showed similar random coil structures as wild-type α-synuclein. However, these isomers did not form aggregates or

Dengue protease-mediated activation and polarization of macrophages.

Structural proteins of viruses are well recognized as major determinants of host immune responses. However, increasing evidence indicates that viral non-structural proteins can also be released from infected cells under inflammatory conditions, including via extracellular vesicles and inflammasome-associated cell death. The immunological consequences of such extracellular exposure of viral non-structural proteins remain poorly understood. Here, we investigated a non-canonical extracellular function of the dengue virus NS2B/NS3 protease and examined its ability to modulate macrophage activation

Rapid and Label-Free Structural Proteomics Using One-Step Swift Trypsin LiP-MS.

Limited proteolysis mass spectrometry (LiP-MS) is a powerful approach for probing protein conformational changes on a proteome-wide scale. However, conventional workflows rely on a two-step digestion with proteinase K and trypsin, which increases complexity and reduces reproducibility and sensitivity. This study aimed to develop a simplified one-step protocol, termed Swift Trypsin LiP-MS (STLiP-MS), which uses a trypsin-immobilized spin column and high-speed centrifugation to achieve rapid and reproducible surface-limited proteolysis. Using HEK293 cell extracts, STLiP-MS identified 286 protein
