Qiju Dihuang Pill modulates complement system in age-related dry eye disease: Tripartite validation through clinical-omics-animal integration.

ETHNOPHARMACOLOGICAL RELEVANCE: Qiju Dihuang Pill (QJDHW) is a traditional Chinese medicine (TCM) formulation used for treating ocular diseases, and clinical evidence has demonstrated its efficacy in managing dry eye disease (DED). Age-related DED is characterized by tear film instability and ocular surface dysbiosis. Current therapeutic agents face challenges in achieving sustained efficacy. TCM offers a multi-targeted intervention strategy for age-related DED. However, the molecular mechanisms underlying QJDHW, a classical TCM formulation, remain to be systematically elucidated. AIM OF THE

Integrative Systems Genomics and Pharmacology Define Shared Immune-Vascular Programs and Prioritize an Experimentally Testable Candidate Linking Sleep Loss to Human Atherosclerosis.

Background: Sleep loss is epidemiologically linked to atherosclerotic cardiovascular disease, yet shared-transcriptome studies typically intersect differentially expressed gene lists and treat the overlap as a shared response, assuming rather than testing directional agreement. Methods: Controlled human sleep restriction (GSE39445; 438 whole-blood samples from 26 participants) and paired carotid plaque versus patient-matched intact artery (GSE43292; 32 patients) were compared across biological compartments at three resolutions: individual genes, annotated gene sets, and redundancy-collapsed pr

Selective and green conversion of 5-HMF to FDCA via enzymatic (laccase) and transition metal (MnO2 and Co-Mn/AC) catalysis in an integrated system.

The sustainable synthesis of bio-based monomers from renewable biomass intermediates is a central goal in green chemistry and biorefinery innovation. This study introduces a synergistic catalytic-enzymatic strategy for the efficient and eco-friendly oxidation of 5-hydroxymethylfurfural (5-HMF) into 2,5-furandicarboxylic acid (FDCA), a key monomer for next-generation biodegradable plastics. The catalytic phase employed non-noble metal catalysts, MnO2 and Co-Mn supported on activated carbon (Co-Mn/AC), under mild batch reaction conditions at 90 °C. Through systematic optimization, a metal-modifi

Wave height fluctuations in the waveforms of an endoscopic pressure study integrated system have the potential to predict acid reflux in gastroesophageal reflux disease (with video).

OBJECTIVES: The endoscopic pressure study integrated system (EPSIS) is a useful diagnostic tool for gastroesophageal reflux disease (GERD). Although wave height fluctuations have been observed in EPSIS waveforms, their clinical significance remains unclear. We hypothesized that the magnitude of these fluctuations may reflect lower esophageal sphincter functionality. This study aimed to evaluate the association between wave height fluctuations and objective acid reflux parameters. METHODS: A retrospective analysis was conducted on patients with GERD symptoms who underwent both EPSIS and 24-h m

Versatile Biosensing Tool: CRISPR-Cas12a System-Integrated Electrochemical Biosensor for Severe Fever with Thrombocytopenia Syndrome Virus Detection in Clinical and Environmental Conditions.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is a highly lethal zoonotic virus with a maximum mortality rate of 30%. Despite its risks and potential for human-to-human transmission, the standard diagnostic method has been absent for SFTSV detection. Therefore, this study introduces a versatile SFTSV biosensing technology using the electrochemical-clustered regularly interspaced short palindromic repeat (EC-CRISPR) system. The sensing membrane is functionalized with the 2WJ DNA@Au-MoS2 signal probe as a strategy to amplify the EC response resulting from target detection events of t

Locally-tailored vs. centrally-administered strategies for implementation of primary human papillomavirus (HPV) screening in an integrated healthcare system: a qualitative research study.

INTRODUCTION: Primary human papillomavirus (HPV) testing is recommended for cervical cancer screening for women aged 30-65 years without a history of abnormal results. However, there is little clear guidance regarding effective strategies for implementing primary HPV screening. As part of an ongoing randomized trial comparing implementation strategies for primary HPV testing (a centrally administered + usual care strategy vs. centrally administered + locally tailored strategy), we evaluated clinician experiences and perceptions of large-scale implementation of primary HPV screening in an integ

Enhanced Integrative Diagnosis and Therapeutic Strategy for Hepatic Ischemia-Reperfusion Injury through HOCl-Sensitive Cysteine Delivery System for Targeted Antioxidant Modulation.

Hepatic ischemia-reperfusion injury (HIRI) is a major cause of postoperative liver insufficiency and liver failure, and its pathological process is closely related to the synergistic dysregulation of oxidative stress. Currently, the diagnosis of HIRI is separated from its treatment, making it difficult to achieve effective intervention for HIRI progression. Considering that HOCl is an important upregulated marker at HIRI sites, this study developed an integrated diagnostic and therapeutic strategy based on HOCl-activated cysteine release. The designed smart molecular system, which specifically

Integrated Physiologically-Based Pharmacokinetic Model with a Quantitative Systems Pharmacology and Toxicology Model for Statins in Disease Population. Part 1: Model Development and Validation.

It is important to understand the key factors affecting the pharmacokinetics (PK), pharmacological response and toxicity of a drug to ensure clinical therapeutic efficacy and safety across disease populations. Traditionally, label dose-adjustment recommendations for patient populations are based on drug plasma concentrations. However, plasma PK may not be an appropriate surrogate for response and/or toxicity for drugs like statins with intracellular targets and tissue distribution influenced by membrane transporters. This study presents the integration of a physiologically-based pharmacokineti
