Application of representational difference analysis to identify sequence tags expressed by Metarhizium anisopliae during the infection process of the tick Boophilus microplus cuticle.

Metarhizium anisopliae is a well-characterized biocontrol agent of a wide range of plagues, including insects and acari. To identify genes involved in the infection process, representational difference analysis was performed using cDNA generated from germinated conidia of M. anisopliae in the tick Boophilus microplus cuticle, and cDNA generated during fungal growth in glucose-rich medium. Sequence determination of approximately 135 clones and comparison analysis using public databases led to the identification of 34 sequences and 14 expressed sequence tags with known orthologs. As expected, al

Thermal constant estimation in tropical horse tick, Anocentor nitens (Acari:Ixodidae).

Experience has demonstrated that the exclusive use of acaricides as a control method against cattle ticks causes several problems, and is not totally effective. Other methods need to be added in order to achieve better and more profitable control of these parasites. Groups of 12 to 15 engorged ticks, bred on bovines, were individually incubated at six temperatures (T = 24, 26, 28, 30, 32, and 34 degrees C) and four relatives humidities (RH = 100, 80, 75.5, and 70%), creating 24 variants. The onset of oviposition and eclosion (emergence of larvae from eggs) was observed in all ticks, and preecl

Communal farmers and tick control--a field study in Zimbabwe.

Intensive dipping of cattle to control ticks and tick-borne diseases precludes development of endemic stability to the latter. Alternative control strategies have been recommended which reduce the frequency of dipping and permit the expression of tick resistance and endemic stability. However, such strategies imply acceptance by cattle owners of higher tick burdens. In this study the views of a group of communal farmers to tick control are described and the impact of these views upon adoption of other strategies is discussed.

Progressive sensitization of circulating basophils against Ixodes ricinus L. antigens during repeated infestations of rabbits.

The sensitivity of rabbit basophils to antigens from Ixodes ricinus females has been studied by a degranulation test. Observations of basophil numbers and degranulation were made on the 6th day of each of four sequential infestations. Maximal degranulation of cells was observed after challenge of cells with antigen at a concentration of 10(6) and 10(7) pg/ml. At these concentrations, during a 1st infestation, 21.8 and 23.6% of cells degranulated. During a 2nd infestation, these percentages increased (34.8 and 33.8%) and reached 59.8 and 63.8% by the 4th infestation. A plasma factor which parti

[Characteristics of the "tache noire" of boutonneuse fever].

Humans show little sensitivity to the bites of healthy Rhipicephalus sanguineus and the skin lesion is extremely slight. The "tache noire" - innoculation eschar - corresponding to the infecting bite of the tick with R. conori is discussed and histopathological findings reported. The patient is frequently unaware of the eschar on his body and elements relating to the duration of the infecting bite are deduced. Apart from its typical features, the tache noire may be very small, with surrounding hyper-ergic reaction, or it may even not be present, depending on different conditions of tissue react

Effect of agro-ecological zone and grazing system on incidence of East Coast Fever in calves in Mbale and Sironko Districts of Eastern Uganda.

Between May 2002 and February 2003 a longitudinal survey was carried out in Mbale and Sironko Districts of Eastern Uganda to determine the influence of agro-ecological zones (AEZ) and grazing systems on tick infestation patterns and incidence of East Coast Fever (ECF) in bovine calves. The study area was stratified into AEZ (lowland, midland and upland) and grazing systems {zero grazing (ZG), restricted-outdoor grazing (ROG) and communal grazing (CG)}, whose strata had previously been shown to influence the prevalence of ECF, babesiosis and anaplasmosis. One hundred and eighty-five smallholder

Prevalence and diversity of spotted fever group Rickettsia species in ixodid ticks from domestic dogs in Chad, Africa.

Tick-borne pathogens in the genus Rickettsia are the causative agents of severe and potentially fatal spotted fever group (SFG) and typhus group diseases in dogs and humans. Climate, habitat, and land-use changes are impacting vector ranges, with expansions potentially resulting in novel pathogens being introduced into naïve locations. Despite the public health importance of SFG Rickettsia, there are relatively few data on the prevalence and diversity of rickettsial pathogens in sub-Saharan Africa. The aim of this study was to characterize the SFG Rickettsia prevalence and diversity in ixodid

Photosensitizers in the fight against ticks: safranin as a novel photodynamic fluorescent acaricide to control the camel tick Hyalomma dromedarii (Ixodidae).

Ticks transmit more pathogen species than any other group of blood-feeding arthropods worldwide, affecting humans, livestock, and companion animals. Hyalomma dromedarii is the predominant tick species infesting camels, and its effective control is of pivotal importance. In this research, we compared the phytoefficacy of safranin (SF), a fluorescent dye applied as an acaricide for the first time, to that of tetramethrin (TM) against engorged females of H. dromedarii through in vitro immersion bioassays. Furthermore, the effect of SF exposure was evaluated on the reproductive potential of surviv

Detection and characterization of amitraz resistance in the southern cattle tick, Boophilus microplus (Acari: Ixodidae).

Amitraz, a formamidine acaricide, plays an important role in the control of the southern cattle tick, Boophilus microplus (Canestrini), and other tick species that infest cattle, dogs, and wild animals. Although resistance to amitraz in B. microplus was previously reported in several countries, the actual measurement of the level of amitraz resistance in ticks has been difficult to determine due to the lack of a proper bioassay technique. We conducted a survey, by using a newly reported technique that was a modification of the standard Food and Agriculture Organization larval packet test, to m

The dog mummy, the ticks and the louse fly: Archaeological report of severe ectoparasitosis in Ancient Egypt.

The authors report the first archaeological case of dog ectoparasitosis in Ancient Egypt. The study of a mummified young dog dated to the Roman period suggests a significant infestation by two blood sucking ectoparasites, respectively the brown dog tick Rhipicephalus sanguineus Latreille, 1806 (Acari: Ixodidae), the louse fly Hippobosca longipennis Fabricius, 1805 (Diptera: Hippoboscidae) as well as puparia of sarcosaprophagous flies (Diptera: Sarcophagidae and Calliphoridae), a potential source of myiasis. All of these species are known to be potential vectors of different pathogens which may

Life cycle of Amblyomma integrum (Acari: Ixodidae) under laboratory conditions.

Amblyomma integrum is a hard tick infesting mainly buffalo and cattle and has been identified as an agent of human otoacariasis in Sri Lanka. Data on the life cycle pattern of A. integrum were collected by experimental infestation on New Zealand white rabbits under laboratory conditions. Wild-caught females laid 55-7389 eggs for 2-35 days after spending a latent period of 10-25 days. Egg incubation period was 31-105 days and the newly emerged larvae started feeding after 4-11 days. Larvae dropped off after feeding and they moulted into nymphs after 10-16 days. Nymphs actively fed on rabbits fo

Natural and experimental infection of white-tailed deer (Odocoileus virginianus) from the United States with an Ehrlichia sp. closely related to Ehrlichia ruminantium.

An Ehrlichia sp. (Panola Mountain [PM] Ehrlichia sp.) closely related to Ehrlichia ruminantium was recently detected in a domestic goat experimentally infested with lone star ticks (LSTs, Amblyomma americanum) collected from Georgia, USA. The infected goat exhibited pyrexia and mild clinical pathologic abnormalities consistent with ehrlichiosis. At least two other Ehrlichia species (Ehrlichia chaffeensis and Ehrlichia ewingii) are maintained in nature by a cycle involving LSTs as the primary vector and white-tailed deer (Odocoileus virginanus) as a known or suspected reservoir. Molecular screening and genetic diversity of tick-borne pathogens associated with dogs and livestock ticks in Egypt.

BACKGROUND: The Middle East and North Africa (MENA) offer optimal climatic conditions for tick reproduction and dispersal. Research on tick-borne pathogens in this region is scarce. Despite recent advances in the characterization and taxonomic explanation of various tick-borne illnesses affecting animals in Egypt, no comprehensive examination of TBP (tick-borne pathogen) statuses has been performed. Therefore, the present study aims to detect the prevalence of pathogens harbored by ticks in Egypt. METHODOLOGY/PRINCIPAL FINDINGS: A four-year PCR-based study was conducted to detect a wide range

Molecular detection of parapoxvirus in Ixodidae ticks collected from cattle in Corsica, France.

BACKGROUND: Several viruses belonging to the family Poxviridae can cause infections in humans and animals. In Corsica, livestock farming (sheep, goats, pigs, and cattle) is mainly mixed, leading to important interactions between livestock, wildlife, and human populations. This could facilitate the circulation of zoonotic diseases, and makes Corsica a good example for studies of tick-borne diseases. OBJECTIVES: To gain understanding on the circulation of poxviruses in Corsica, we investigated their presence in tick species collected from cattle, sheep, horses, and wild boar, and characterized
