Size and spatial orientation of uterine tissue transplants on the peritoneum crucially determine the growth and cyst formation of endometriosis-like lesions in mice.

BACKGROUND: In many studies in rodents, intraperitoneal endometriosis-like lesions are surgically induced by syngeneic or autologous transplantation of uterine tissue samples, which are sutured to the abdominal wall. However, until now the surgical techniques have not been standardized, and we address this issue here. METHODS: Uterine tissue samples were transplanted to the peritoneum of C57BL/6 mice (four study groups, n = 7 each). Using non-invasive high-resolution ultrasound imaging over a period of 4 weeks, we analyzed growth characteristics and cyst formation of the endometriosis-like le

The outcomes of living donor renal transplants with multiple renal arteries: a large cohort study with a mean follow-up period of 10 years.

BACKGROUND: Living donor kidney transplants with multiple arteries are presumed to be associated with an increased risk of complications. OBJECTIVES: The aim of the study was to compare the outcomes in living donor transplantation with the specific intention of comparing long-term outcomes in which the donor kidney had 1 or more renal arteries. The study was undertaken in 2 large transplant centers. METHODS: A retrospective analysis of 201 living donor kidney transplants with multiple arteries that were performed between January 1985 and December 2004 was undertaken. We recorded patient and …

Transplants of adrenal medullary chromaffin cells reduce forelimb and hindlimb allodynia in a rodent model of chronic central pain after spinal cord hemisection injury.

In the majority of patients, spinal cord injury (SCI) results in abnormal pain syndromes in which non-noxious stimuli become noxious (allody … Nonimmunosuppressed injured animals received either control-striated muscle (n = 7) or AM (n = 10) transplants. Nociceptive behavior was tested for 4 weeks posttransplant as measured by paw withdrawals to von Frey filaments, radiant heat, and pin prick stimuli. Hemisected animals receiving AM demonstrated statistically significant reductions in both fore- an …

Control of bone resorption by hematopoietic tissue. The induction and reversal of congenital osteopetrosis in mice through use of bone marrow and splenic transplants.

The reciprocal transplantation of hematopoietic tissues was carried out on young, lethally irradiated mice of inbred, microphthalmic stock. The cell infusions prepared from the bone marrow or spleen of a normal littermate fully restored capacity to resorb bone and cartilage in the osteopetrotic recipients. Conversely, cell infusions prepared from the spleen of microphthalmic mice induced osteopetrosis in their irradiated, normal littermates. It is concluded that resorption of skeletal matrix is controlled by migratory cells, possibly osteoclastic progenitors, derived from the myelogenous tissu

Pre-transplant hepatocellular carcinoma is associated with increased risk of post-transplant non-HCC cancer in liver transplant recipients.

BACKGROUND: Post-transplant malignancy (PTM) contributes substantially to late morbidity and mortality after liver transplantation (LT), yet contemporary data on cancer spectra across cirrhosis etiologies remain limited. We assessed PTM prevalence, determinants, and cancer-type distribution by pre-LT etiology in a Canadian cohort to inform risk-adapted surveillance in routine practice. METHODS: We conducted a retrospective cohort study of adults undergoing LT at a tertiary Ontario center (2007-2018) with 60-month follow-up. De novo and recurrent malignancies were ascertained through June 2023

Simultaneous islet-kidney transplantation as an alternative for patients ineligible for pancreas transplant: Initial experience at a North American transplant center.

Simultaneous pancreas-kidney (SPK) transplantation is the gold standard for patients with diabetes mellitus and end-stage renal disease, but many are ineligible due to surgical risk. Simultaneous islet-kidney (SIK) transplantation offers a less invasive alternative by combining kidney transplantation with islet infusion. We report our initial North American experience with SIK. Between July 2022 and April 2025, 13 patients with type 1 diabetes mellitus and end-stage renal disease underwent SIK after being declined for SPK, primarily due to peripheral vascular disease. Follow-up ranged from 4 t

Pre-transplant morphometry by computed tomography scan and post-transplant dialysis risk in overweight or obese kidney transplant recipients.

PURPOSE: Adipose distribution and sarcopenia could better assess kidney transplantation outcomes than body mass index (BMI) and have been poorly evaluated among obese and overweight recipients. We aimed to evaluate morphometric radiologic markers to predict post-operative dialysis within this population. METHODS: We conducted a retrospective study including patients with a BMI > 25 kg/m2 undergoing kidney transplantation during 5 years. Subcutaneous adipose tissue surface (SAT), visceral adipose tissue surface (VAT), and psoas surface were measured on CT scans sections. A model predictive of

Pre-transplant IFN-gamma ELISPOTs are associated with post-transplant renal function in African American renal transplant recipients.

Final crossmatch testing is routinely used to assess the risk of antibody-mediated graft injury/rejection post-transplant. Analogously, we postulated that quantitative measurements of anti-donor effector/memory T cells pre-transplant would independently assess post-transplant risk. To address this hypothesis, we determined the frequencies of pre-transplant, donor-specific interferon-gamma (IFN-gamma) enzyme-linked immunosorbent spots (ELISPOTs) and correlated the results with post-transplant outcomes in 37 African American recipients of deceased donor kidney transplants treated with tacrolimus…

Cost-utility analysis of living-donor kidney transplantation followed by pancreas transplantation versus simultaneous pancreas-kidney transplantation.

For a type I diabetic with end-stage renal disease, the choice between a kidney-alone transplant from a living-donor (KA-LD) and a simultaneous pancreas kidney (SPK) transplant remains a difficult one. The prevailing practice seems to favor KA-LD over SPK, presumably due to the superior long-term renal graft survival in KA-LD and the elimination of the lengthy waiting time on the cadaver transplant list. In this study, two treatment options, KA-LD followed by pancreas-after-kidney (PAK) and SPK transplant, are compared using a cost-utility decision analysis model. The decision tree consisted o

Reduced size liver transplantation, split liver transplantation, and living related liver transplantation in relation to the donor organ shortage.

Because of the shortage of cadaveric donors, three techniques of partial liver grafting have been developed. These techniques are placed in perspective in relation to the organ shortage. Reduced size liver transplantation (RSLTx) is widely used and has results comparable to those from whole liver grafting. However, this technique, while benefitting pediatric patients, reduces the adult donor liver pool. It also makes inefficient use of an available adult donor liver. In split liver transplantation (SPLTx), the whole liver is used after bipartition for two recipients. The results are comparable

Impacts of Pre-transplant Panel-Reactive Antibody on Post-transplantation Outcomes: A Study of Nationwide Heart Transplant Registry Data.

BACKGROUND AND OBJECTIVES: The number of sensitized heart failure patients on waiting lists for heart transplantation (HTx) is increasing. Using the Korean Organ Transplantation Registry (KOTRY), a nationwide multicenter database, we investigated the prevalence and clinical impact of calculated panel-reactive antibody (cPRA) in patients undergoing HTx. METHODS: We retrospectively reviewed 813 patients who underwent HTx between 2014 and 2021. Patients were grouped according to peak PRA level as group A: patients with cPRA ≤10% (n= 492); group B: patients with cPRA >10%, <50% (n=160); group C p

Meeting report of the STAR-Sensitization in Transplantation Assessment of Risk: Naïve Abdominal Transplant Organ subgroup focus on kidney transplantation.

The development of de novo donor-specific HLA antibody (dnDSA) is a critical feature contributing to late allograft failure. The complexity of the issue is further complicated by organ-specific differences, detection techniques, reliance of tissue histopathology and changing diagnostic criteria, ineffective therapies, and lack of consensus. To tackle these issues, the Sensitization in Transplantation Assessment of Risk (STAR) 2017 was initiated as a collaboration of the American Society of Transplantation and American Society of Histocompatibility and Immunogenetics consisting of 8 working gro

[Treatment strategy for rejection-free corneal transplantation--transition from full-thickness corneal transplantation to corneal endothelium transplantation].

The avoidance of allograft rejection is the most critical factor for favorable surgical outcome after corneal transplantation. We report experimental data including distribution of white blood cells in human corneas for rejection-free corneal transplantation. We focused on leukocyte trafficking based on the immunological mechanism leading to allograft rejection in a mouse full-thickness corneal transplantation model. We identified two chemokine-receptors, CCR1 and CCR7 which are functionally relevant to the occurrence of allograft rejection. These chemokine receptors can be new targets for the

Pre-transplant evaluation of immunological risk in paediatric kidney transplant recipients.

Kidney transplantation is the preferred treatment for children with kidney failure. Clinicians focus on improving graft outcomes by ensuring an optimal immunological environment for recipients. The objective of this review is to provide an overview of the immunological evaluation of a potential paediatric kidney transplant recipient. This review focuses on the role of Human Leucocyte Antigen (HLA) laboratory testing and its clinical applications in the pre-transplant immunological risk assessment for a transplant physician. It provides a basic insight on the current technologies, available tes

Residential and Transplant Center Neighborhood Segregation and Live Donor Liver Transplant.

IMPORTANCE: Neighborhood segregation, a mechanism of structural racism, is associated with racial and ethnic disparities in health care access and outcomes. Live donor liver transplant (LDLT) is the ideal treatment for cirrhosis, improving survival and quality of life. Understanding the role of segregation in LDLT access is important to address disparities. OBJECTIVE: To assess the associations between residential and transplant center neighborhood segregation and LDLT access. DESIGN, SETTING, AND PARTICIPANTS: This cohort study used data from a US national transplant registry on adult candi

Pre-transplant JAK-inhibition improves post-transplant outcome in myelofibrosis.

Allogeneic hematopoietic stem cell transplantation (alloHSCT) remains the only curative approach in myelofibrosis (MF). Janus-Associated Kinase inhibitors (JAKi) have changed the treatment paradigm of MF patients, becoming a therapeutic mainstay in those with splenomegaly and/or constitutional symptoms. The impact of JAKi prior to allogeneic transplant is still under investigation. The aim of the current study was to analyze the outcome and prognostic factors, especially JAKi impact on transplantation outcome in MF patients. The study comprised 96 patients (64 - MF, 21 - post-polycythemia vera
