A Comprehensive Assessment of the Shared Genetic Architecture between Myopia and Open-Angle Glaucoma.

OBJECTIVE: Individuals with high myopia have an increased prevalence of open-angle glaucoma (OAG). We aim to clarify the possibly shared genetic architecture of myopia and OAG, in particular in high myopes with myopic macular degeneration (MMD), where OAG screening is highly challenging. DESIGN: Individual participant data meta-analysis of one-sample Mendelian randomization analyses and pleiotropic analysis under a composite null hypothesis. PARTICIPANTS: A total of 34 825 participants from 6 population-based cohort studies and 1 high myopia case-control study, including 708 OAG and 1953 hig…

Exploring the shared genetic architecture and causal relationship between childhood maltreatment and psychiatric disorders.

Childhood maltreatment (CM) is a major risk factor for psychiatric disorders, yet the shared genetic etiology, potential causal effects, and mediating pathways underlying these associations remain incompletely understood. We investigated the shared genetic architecture, potential causal relationships, and candidate mediating pathways linking genetically proxied reported CM with major psychiatric disorders. Summary statistics were obtained from large-scale genome-wide association studies of CM and 10 psychiatric disorders. Genetic correlations were estimated using linkage disequilibrium score r…

Jaw-specific stem cell dynamics are associated with distinct enamel architecture in mouse incisors.

BACKGROUND: The continuously growing mouse incisor serves as a model for stem cell-driven tooth renewal. Although mice possess maxillary and mandibular incisors, most studies have focused on the mandible. Potential differences in structure and renewal between these serially homologous organs remain insufficiently characterized. RESULTS: Mandibular incisors are longer and contain greater enamel volume, mineral density, and mineral content than maxillary incisors, whereas maxillary incisors are more curved and display a larger cross-sectional area. Mineral density increases along the apical-to-

Integrative genomic analysis of 21 orofacial diseases identifies shared genetic architecture with systemic diseases.

Orofacial diseases are closely linked to systemic health, yet their genetic architecture and causal relationships remain incompletely understood. Here we show that integrative genomic analyses of 21 orofacial diseases in the UK Biobank, combining genome-wide, transcriptome-wide, rare variant association studies and Mendelian randomization, identify 48 novel loci and prioritize 160 putative causal genes. Among these, a synonymous variant in ALDH1A2 associated with periodontitis, encoding a key enzyme in retinoic acid synthesis, reduces translational efficiency, as shown by CRISPR-Cas9 editing. …

Macrophage Polarization and Spatial Architecture Characterize the Tumor Immune Microenvironment and Prognosis in Hepatoblastoma.

Hepatoblastoma, the most common primary pediatric liver malignancy, exhibits resistance to contemporary treatments in a subset of patients. Understanding the tumor immune microenvironment is essential for the development of novel immuno-oncological therapies. Herein, the microenvironment of hepatoblastoma was characterized by mapping the spatial distribution of immune cells, identifying prognostic immune features, assessing chemotherapy-induced alterations, and exploring potential immunotherapeutic targets. Whole-slide multiplex immunofluorescence staining (n = 35) was performed on formalin-fi

Adenoviral-Mediated SIRT1 Expression Restores Lung Architecture and Reduces Fibrotic Progression in Experimental Models.

Background: Pulmonary fibrosis (PF) involves remodeling of lung architecture and limited therapeutic options are available. Sirtuin 1 (SIRT1), an epigenetic regulator, has emerged as a promising antifibrotic target due to its role in apoptosis, inflammation, and chromatin remodeling. Methods: We evaluated the effects of a recombinant adenovirus expressing human SIRT1 (Ad-SIRT1) both in vitro and in vivo. Human alveolar epithelial cells (A549) were transduced with Ad-SIRT1 or AdGFP and subjected to bleomycin-induced injury. Transduction efficiency, cell viability (MTT assay), SIRT1 localization…

TyG-Centered Endocrine-Metabolic Architecture in Patients with Thyroid Dysfunction: A Systems Phenotyping Study.

BACKGROUND: Metabolic dysfunction is a complex process arising from coordinated interactions among insulin resistance, dyslipidemia, hepatic dysfunction, obesity, low-grade inflammation, and endocrine alterations. The triglyceride-glucose (TyG) index is a simple, reproducible surrogate of insulin resistance, but most studies have evaluated it through isolated association-based analyses. We characterized the endocrine-metabolic architecture associated with TyG in patients with thyroid dysfunction using a systems-phenotyping approach. METHODS: In this retrospective, single-center, cross-section…

A Multimodal TinyML-Based Predictive Maintenance Architecture for Industrial IoT in the 6G Era.

Predictive maintenance (PdM) is central to Industry 5.0 strategies for reducing unplanned downtime in rotating machinery. This work proposes and evaluates, as a proof of concept on a controlled single-machine testbed, a multimodal TinyML edge architecture for PdM designed to remain compatible across the application plane's evolution toward sixth-generation (6G) networks. Three complementary modalities run local inference on commercial off-the-shelf smart sensor nodes-vibration, acoustic, and thermography-with an embedded gateway bridging per-modality decisions to a serverless cloud back-end. Transcriptomic Architecture of Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Risk in Mexican Americans.

Hispanics of Mexican American descent in South Texas show a very high prevalence of MASLD, with some studies reporting rates as high as 50% in adults. However, assessment of genetic risk factors underlying this prevalence is complicated by a high co-occurrence of other metabolic disorders and variable endogenous and exogenous environmental risk factors. To map the transcriptomic architecture of MASLD hepatic steatosis risk, we conducted an epidemiological-scale investigation using human induced pluripotent stem cell (iPSC)-derived hepatocyte cultures from 193 participants in our longitudinal S…

Islet delta-cell architecture is remodelled in the human pancreas during type 1 diabetes.

Delta cells participate in regulating hormone secretion in adjacent alpha- and beta cells and a general assumption is that cells with a shorter distance to the secreting cell receive a higher concentration of the secretory compounds. Isolated islets obtained from donors with type 1 diabetes have a reduced glucagon secretion during low glucose levels, but adding a somatostatin receptor inhibitor increases the glucagon secretion. Despite this, information regarding the delta-cell architecture during diabetes is sparse. The aim of the current study was to determine intra-islet and extra-islet del…

Dissecting the shared genetic architecture between nonalcoholic fatty liver disease and type 2 diabetes.

Observational studies have reported a bidirectional correlation between nonalcoholic fatty liver disease (NAFLD) and type 2 diabetes (T2D), but the shared genetic basis between the two conditions remains unclear. Using genome-wide association study (GWAS) summary data from European-ancestry populations, we examined the cross-trait genetic correlation and identified genomic overlaps and shared risk loci. We employed a latent causal variable model and Mendelian randomization (MR) analysis to infer causal relationships. Colocalization analysis and conditional/conjunctional false discovery rate (c

Patient-specific functional brain architecture explains cortical patterns of tau PET in Alzheimer's disease.

The spatial distribution of tau pathology, the core driver of neurodegeneration in Alzheimer's disease (AD), varies markedly across individu … prognostic value. Together, these findings provide strong evidence that individual functional brain architecture shapes tau propagation in humans, supporting the network spread hypothesis by showing that variability in connectivity translates into heterogeneity in tau distribution. This work advances biological understanding of tau propagation in AD, highlight …

Dynamic architecture of mammalian paternal chromatin: histone-to-protamine exchange and post-fertilization reprogramming.

Chromatin remodeling in male germ cells and after fertilization plays a pivotal role in genetic transmission and early embryonic development. During spermatogenesis, histone-based chromatin undergoes progressive reorganization: canonical histones are gradually replaced by testis-specific variants, then by transition proteins, and ultimately by highly basic protamines (PRM1 and PRM2). This hierarchical replacement, modulated by histone post-translational modifications-including hyperacetylation, ubiquitination, and dynamic methylation-and supported by molecular chaperones and chromatin remodele

S100A proteins show a spatial distribution of inflammation associated with the glioblastoma microenvironment architecture.

Background: Glioblastoma IDH wild type (GBM IDH wt) has a poor prognosis and a strongly associated with inflammatory processes. Inflammatory … r findings define different functions for S100A9, A11 and A13 proteins that are associated with the architecture of the glioblastoma microenvironment and define its progression. Moreover, these alterations can be reversed by the RAGE inhibitor, Azeliragon which is in a phase I/II clinical trial NCT05635734.

Biomechanical and inflammatory pathways underlying the genetic architecture of keratoconus: A genomic SEM study.

BACKGROUND: The genetic architecture of keratoconus (KC) is complex, and the interplay between its biomechanical and inflammatory etiological components remains poorly defined. We aimed to quantitatively resolve these distinct genetic pathways to provide a mechanistic framework for disease pathogenesis. METHODS: We applied genomic structural equation modeling (genomic SEM) to large-scale GWAS summary statistics to model two latent genetic factors: a Corneal Biomechanical Stability Factor (CBSF) and an Atopic-Inflammatory Liability (AIL) factor. Pathway-specific multivariate GWAS were subseque…
