Exposure to endocrine disruptors in utero appears to alter epigenetics in the male germ-line and subsequently promote adult-onset disease in subsequent generations. Fetal exposure to bisphenol A (BPA), a highly prevalent endocrine disruptor in environment, has been shown to alter epigenetic modification and result in glucose intolerance in adulthood. However, whether fetal exposure to BPA can induce epigenetic modification and phenotypic changes in their subsequent offspring are still unclear. The present study was designed to investigate whether exposure to BPA in early life induced glucose i

The potential for health effects on humans with exposure to bisphenol A (BPA) has raised concerns, and the adverse effects of low-dose exposure to BPA on reproduction have been controversial. The purpose of the present study was to investigate the effects of low-dose exposure to BPA on reproductive development in F(1) rat offspring. Pregnant female Sprague-Dawley rats (F(0)) were fed a diet containing low doses of BPA (0, 0.33, 3.3, or 33 ppm) from gestational day (GD) 6 through postnatal day (PND) 21. The weanlings (F(1)) from all dose groups were fed a normal diet ad libitum after weaning an

The estrogen receptor ERalpha is emerging as a key molecule involved in glucose and lipid metabolism. The main functions of pancreatic beta-cells are the biosynthesis and release of insulin, the only hormone that can directly decrease blood glucose levels. Estrogen receptors ERalpha and ERbeta exist in beta-cells. The role of ERbeta is still unknown, yet ERalpha plays an important role in the regulation of insulin biosynthesis, insulin secretion and beta-cell survival. Activation of ERalpha by 17beta-estradiol (E2) and the environmental estrogen bisphenol-A (BPA) promotes an increase of insuli

To investigate human exposure to bisphenol A (BPA), a widely used endocrine disruptor, we measured serum BPA concentrations and analyzed the interrelation of BPA with sex-related hormones. BPA was detected in all human sera by a novel enzyme-linked immunosorbent assay. Serum BPA concentrations were significantly higher in normal men (1.49 +/- 0.11 ng/ml; P < 0.01) and in women with polycystic ovary syndrome (1.04 +/- 0.10 ng/ml; P < 0.05) compared with normal women (0.64 +/- 0.10 ng/ml). There were significant positive correlations between serum BPA and total testosterone (r = 0.595, P < 0.001

BACKGROUND: Male infertility (MI) impacts about one in seven couples globally, involving complex gene-environment interactions. Environmental pollutants may disrupt spermatogenesis via mitochondrial dysfunction, but key targets and mechanisms are unclear. Integrating multi-omics with computational toxicology offers a novel strategy to decipher these interactions.
METHODS: We integrated blood transcriptomics from MI patients, GWAS summary statistics, and QTL data (methylation, expression, protein). MI-related mitochondrial genes were identified through differential expression analysis, followe

There is concern about cumulative exposures to compounds that disrupt male sexual differentiation in foetal life, leading to irreversible effects in adulthood, including declines in semen quality, testes non-descent, malformations of the penis and testis cancer. Traditional chemical-by-chemical risk assessment approaches cannot capture the likely cumulative health risks. Past efforts of focusing on combinations of phthalates, a subgroup of chemicals suspected of contributing to these risks, do not go far enough, as they ignore the contribution of other types of chemicals. With the aim of provi

Repeat breeding syndrome (RBS), characterized by repeated artificial insemination failure, is associated with ovarian dysfunction in ruminants and causes economic losses. Environmental endocrine-disrupting chemicals (EDCs), such as bisphenol F (BPF), may contribute to RBS, but the relationship between BPF exposure and RBS in cows remains unclear. In this study, we conducted an epidemiological investigation and clinical examinations in 111 adult Simmental cows, including 7 cows diagnosed with RBS, from two local farms, and measured the BPF levels in the drinking water, feed, and the serum sampl

Endocrine-disrupting chemicals particularly plasticisers such as phthalates, bisphenols and their emerging substitutes, are increasingly recognised as the main environmental toxicants with significant implications for thyroid and cardiovascular health. Due to their widespread use in consumer products, food packaging, medical devices, cosmetics and industrial materials, chronic human exposure occurs via ingestion, inhalation, dermal absorption and parenteral administration. Increasing evidence indicates that these compounds disrupt the hypothalamic-pituitary-thyroid axis by interfering with thy

Bisphenol A (BPA) is a synthetic organic compound commonly utilized in the manufacturing of polycarbonate plastics and epoxy resins. Its widespread presence in everyday items, including water bottles, food containers, and thermal paper, has raised considerable health concerns due to its potential as an endocrine disruptor. BPA has the ability to mimic estrogen and bind to estrogen receptors, which can lead to disruptions in hormonal signaling pathways. This interference poses risks to reproductive health, especially among younger individuals, as it may result in irregular menstrual cycles, ovu

Persistent organic pollutants (POPs) pose severe environmental risks because of their chemical stability, bioaccumulation, and toxicity. The Bio-Fenton process, coupling microbial metabolism with Fenton chemistry to generate •OH in situ, offers a green and sustainable remediation strategy. However, its application is hindered by inefficient Fe(III) reduction and insufficient endogenous H2O2 production. In this study, we developed a self-sustained microbial Fenton system driven by Shewanella oneidensis MR-1 under oxygen-limited conditions. Fine-tuning the dissolved oxygen level enabled simultan

Diapers are a staple care product for infants, yet concerns persist regarding the potential risks posed by dermal exposure to chemicals through their usage. This review provides a comprehensive summary of reported chemicals, highlighting the frequent detection of polychlorodibenzo-p-dioxins (PCDDs), phthalates (PAEs), volatile organic compounds (VOCs), polycyclic aromatic hydrocarbons (PAHs), bisphenols (BPs), organotins, and heavy metals. Disposable diapers commonly exhibit higher concentrations of VOCs, PAEs, BPs, and heavy metals than other chemicals. Our estimation reveals formaldehyde as

One of the aims of the European Human Biomonitoring Initiative, HBM4EU, was to provide examples of and good practices for the effective use of human biomonitoring (HBM) data in human health risk assessment (RA). The need for such information is pressing, as previous research has indicated that regulatory risk assessors generally lack knowledge and experience of the use of HBM data in RA. By recognising this gap in expertise, as well as the added value of incorporating HBM data into RA, this paper aims to support the integration of HBM into regulatory RA. Based on the work of the HBM4EU, we pro

Background Bisphenol A (BPA) is an estrogenic, endocrine-disrupting compound widely used in the industry. It is also a ubiquitous environmental pollutant. Its presence was confirmed in human fetuses, which results from maternal exposure during pregnancy. The mechanisms behind maternal-fetal transfer, and relationships between pregnant women and fetal exposures remain unclear. The aim of this study was to assess the impact of maternal exposure to BPA on the exposure of the fetus. Methods Maternal plasma and amniotic fluid samples were collected from 52 pregnant women undergoing amniocentesis fo

Maternal thyroid hormones (THs) play an essential role in the embryonic and larval development of fish. Previous studies in fish have reported that parental exposure to thyroid disrupting chemicals (TDCs) changed maternal TH levels in the offspring; however, whether this transgenerational thyroid endocrine disruption can further disturb the early development of the offspring still remains largely unknown. Bisphenol S (BPS), a substitute of bisphenol A, has been reported to be a potential TDC. In this study, zebrafish (F0) were exposed to environmentally relevant concentrations (1, 10, and 100

Endocrine-disrupting chemicals (EDCs) comprise structurally diverse contaminants that are frequently detected in human biological matrices and may interfere with endocrine homeostasis even at low exposure levels. Developing practical multi-residue methods for biomonitoring remains challenging because EDC classes differ markedly in polarity, ionization behavior, and urinary matrix susceptibility. In this study, we developed and validated an ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method for the simultaneous determination of 32 target EDCs in urine fro

Formaldehyde is a volatile organic compound that may remain in, or be released from, epoxy adhesive formulations. Prolonged or high-level exposure can cause chronic respiratory injury and elevate cancer risk. Accordingly, developing epoxy curing agents from renewable bioresources without introducing formaldehyde is of significant importance for safeguarding human health and reducing environmental impacts. In this study, a biobased phenalkamine curing agent was synthesized from cardanol via a Mannich reaction incorporating 1,4-butanediamine and citronellal, and subsequently combined with bisphe

In excess of 13,000 chemicals are added to plastics ('additives') to improve performance, durability, and production of plastic products. They are categorized into numerous chemical classes including flame retardants, light stabilizers, antioxidants, and plasticizers. While research on plastic additives in the marine environment has increased over the past decade, there is a lack of methodological standardization. To direct future measurement of plastic additives, we compiled a first-of-its-kind dataset of literature assessing plastic additives in marine environments, delineated by sample type
