The pattern of thalamocortical and brain stem projections to the vibrissae-related sensory and motor cortices in de-whiskered congenital hypothyroid rats.

The present study is designed to investigate the plastic organization of the thalamo-cortical (TC) and brain stem afferents of whisker primary sensory (wS1) and motor (wM1) cortical areas in congenital hypothyroid (CH) pups following whisker deprivation (WD) from neonatal to adolescence period. Maternal hypothyroidism was induced by adding propylthiouracil (PTU) to the drinking water from early embryonic day 16 to postnatal day (PND) 60. Pregnant rats were divided into intact and CH groups (n = 8). In each group, the total whiskers of pups (4 of 8) were trimmed continuously from PND 1 to PND 6…

Regulation of endogenous neural stem/progenitor cells for neural repair-factors that promote neurogenesis and gliogenesis in the normal and damaged brain.

Neural stem/precursor cells in the adult brain reside in the subventricular zone (SVZ) of the lateral ventricles and the subgranular zone (SGZ) of the dentate gyrus in the hippocampus. These cells primarily generate neuroblasts that normally migrate to the olfactory bulb (OB) and the dentate granule cell layer respectively. Following brain damage, such as traumatic brain injury, ischemic stroke or in degenerative disease models, neural precursor cells from the SVZ in particular, can migrate from their normal route along the rostral migratory stream (RMS) to the site of neural damage. This neur

Cordocytes-stem cells cooperation in the human brain with emphasis on pivotal role of cordocytes in perivascular areas of broken and thrombosed vessels.

This study is based on data analysis by light and transmission electron microscopy of the surgical cases in cerebral tumors, cerebrovascular malformations, thromboses in the carotid system, and other injuries such as perivascular hemorrhage. We examined cortical arteries and veins, perivascular areas with old hematic masses, vasculogenic foci, and broken large vessels. We identified, characterized, and compared both undifferentiated cells and well-differentiated cordocytes within periadventitial areas where these cells cooperate very well with precursor/stem cells to perform vital functions fo

Leukoencephalopathy with brain stem and spinal cord involvement and lactate elevation is associated with cell-type-dependent splicing of mtAspRS mRNA.

LBSL (leukoencephalopathy with brain stem and spinal cord involvement and lactate elevation) is an autosomal recessive white matter disorder with slowly progressive cerebellar ataxia, spasticity and dorsal column dysfunction. Magnetic resonance imaging shows characteristic abnormalities in the cerebral white matter and specific brain stem and spinal cord tracts. LBSL is caused by mutations in the gene DARS2, which encodes mtAspRS (mitochondrial aspartyl-tRNA synthetase). The selective involvement of specific white matter tracts in LBSL is striking since this protein is ubiquitously expressed. …

[A case of chronic methyl bromide intoxication showing symmetrical lesions in the basal ganglia and brain stem on magnetic resonance imaging].

A 30-year-old man developed paresthesia in the feet, unstable gait and blepharoptosis subsequent to pharyngeal pain while working in a warehouse of imported fruit and vegetable fumigated with methyl bromide and cyanate. Neurological examination showed bilateral blepharoptosis, diplopia, ataxic gait and paresthesia below the inguinal region. The serum concentration of thiociane, a metabolite of cyanide, was within normal limits, but that of bromide was elevated (43.7 mg/l, normal: 3.7-8.6). Thus we diagnosed the patient as intoxication of methyl bromide. Magnetic resonance imaging of the brain

The plated bayonet: a new instrument to facilitate surgery for intra-axial neoplasms of the spinal cord and brain stem. Technical note.

A new instrument for use in surgically treating intra-axial neoplasms of the spinal cord and brain stem is described. The plated bayonet allows neoplastic tissue in the spinal cord to be separated from functioning neural elements without perforating the adjacent spinal cord. In addition, the plated bayonet facilitates exposure through the very small incision necessary to remove tumors of the brain stem without damaging cranial nerves or other vital structures.

A combined autoradiographic and immunocytochemical study of 3H-corticosterone target neurons and catecholamine neurons in rat and mouse lower brain stem.

Target neurons for 3H-corticosterone were identified by autoradiography and tyrosine hydroxylase containing neurons were identified by immunocytochemistry in the same sections of lower brain stem of 38-day-old rats, 10-day-old rats and adult mice. In all animals studied, the same topographic distribution of neurons was found which exhibited a nuclear accumulation of 3H-corticosterone. Target neurons for 3H-corticosterone were widely distributed in the brain stem. Motor neurons of the cranial nerves and certain neurons of the reticular formation were among the most heavily labeled cells. Neuron…

A Rare Case of Successful Perioperative Management of a Child With Oropharyngeal Sharp Foreign Body Penetrating the Brain stem: A Case Report.

BACKGROUND: Penetrating foriegn bodies extending into brain stem are rare in children and present unique anesthetic and surgical challenges. Airway management is particularly difficult because of restricted oral access, risk of displacement leading to catastrophic hemorrhage and neurological complications. CASE: We describe the perioperative management of achild who presented in emergency with accidentally impacted foriegn body. The metallic foriegn body occupied the oral cavity significantly limiting the airway access and posing a risk of airway obstruction. A multidisciplinary team involvin…

Update on the interaction of notch and Wnt/β-catenin signaling in neural stem cells and brain homeostasis during ischemic stroke.

Stroke is the second leading cause of death globally and a major contributor to long-term disability. Neural stem cells (NSCs) offer a promising therapeutic strategy for ischemic conditions by modulating neuroinflammation, enhancing neuroplasticity, facilitating tissue remodeling, and promoting neural tissue regeneration. Preclinical evidence strongly indicates that NSCs migrate to damaged brain regions and play a crucial role in repair by differentiating into various functional neural cell types. Recent research has underscored the significance of specific molecular pathways, particularly the

The stems of Syringa pinnatifolia alleviate insomnia based on the brain-gut-microbiota axis mediated by 5-HT in mice and rats.

The peeled stems of Syringa pinnatifolia Hemsl. (SP) have long been used for the treatment of insomnia in Inner Mongolia, China, but their hypnotic constituents and underlying pharmacological basis remain poorly defined. In this study, we investigated the hypnotic activity of SP and identified its active fraction and major constituents using an equal mass to equivalent effect transmission (EMEET)-guided fractionation strategy. SP extracts and fractions were evaluated in pentobarbital-induced sleep tests and a PCPA-induced insomnia model in mice and rats. The phenolic fraction (Ps) significantl

The effect of maternal sleep deprivation on differentiation of mesenchymal stem cells in the presence of neonates brain cerebrospinal fluid of Wistar rats.

OBJECTIVE: Cerebrospinal fluid (CSF) contains proliferation, differentiation and maturation signals that are essential factors for brain development. Due to presence of such factors this fluid has proliferative and differentiation potential. A previous study showed that maternal sleep deprivation (MSD) decrease the number of newborn neurons in development of hippocampus. Also, it impairs hippocampus-dependent spatial learning and memory in the young offspring rat. MSD can change CSF factors. In the present study, the effect of MSD-CSF on differentiation of mesenchymal stem cells to neural cell

Novel Regenerative Therapies Based on Regionally Induced Multipotent Stem Cells in Post-Stroke Brains: Their Origin, Characterization, and Perspective.

Brain injuries such as ischemic stroke cause severe neural loss. Until recently, it was believed that post-ischemic areas mainly contain necrotic tissue and inflammatory cells. However, using a mouse model of cerebral infarction, we demonstrated that stem cells develop within ischemic areas. Ischemia-induced stem cells can function as neural progenitors; thus, we initially named them injury/ischemia-induced neural stem/progenitor cells (iNSPCs). However, because they differentiate into more than neural lineages, we now refer to them as ischemia-induced multipotent stem cells (iSCs). Very recen

Human Cytomegalovirus IE2 Protein Disturbs Brain Development by the Dysregulation of Neural Stem Cell Maintenance and the Polarization of Migrating Neurons.

Despite the high incidence of severe defects in the central nervous system caused by human cytomegalovirus (HCMV) congenital infection, the mechanism of HCMV neuropathogenesis and the roles of individual viral genes have not yet been fully determined. In this study, we show that the immediate-early 2 (IE2) protein may play a key role in HCMV-caused neurodevelopmental disorders. IE2-transduced neural progenitor cells gave rise to neurospheres with a lower frequency and produced smaller neurospheres than control cells in vitro, indicating reduction of self-renewal and expansion of neural progeni

A comparison of auditory brain stem responses elicited by click and chirp stimuli in adults with normal hearing and sensory hearing loss.

OBJECTIVES: The aim of this study was to compare the human auditory brain stem response (ABR) elicited by clicks and chirps with overall behavioral hearing thresholds in participants with normal hearing and with sensory hearing loss. The authors hypothesized that ABRs to chirps would be more robust and that thresholds would be more similar to overall behavioral hearing thresholds compared with ABRs to clicks. DESIGN: Twenty-five adults with normal hearing and 25 adults with sensory hearing loss were recruited. Subjects were without middle ear or neurological pathologies at the time of testing…

Homing of neural stem cells from the venous compartment into a brain infarct does not involve conventional interactions with vascular endothelium.

Human neural stem cells (hNSCs) hold great potential for treatment of a wide variety of neurodegenerative and neurotraumatic conditions. Heretofore, administration has been through intracranial injection or implantation of cells. Because neural stem cells are capable of migrating to the injured brain from the intravascular space, it seemed feasible to administer them intravenously if their ability to circumvent the blood-brain barrier was enhanced. In the present studies, we found that interactions of hNSCs in vitro on the luminal surface of human umbilical vein endothelial cells was enhanced
