Schwannomas of the sinonasal tract and nasopharynx.

Approximately 45% of benign peripheral nerve sheath tumors occur in the head and neck region. Of these, schwannomas (neurilemomas) arising from the nasal cavity and paranasal sinuses account for less than 4%. Pathologic features of this subset are not well documented. We report a series of five cases of sinonasal schwannoma and one in the nasopharynx. The male-to-female ratio was equal, and the age at presentation ranged from 38 to 65 years of age (median, 52 yr). Four of the lesions were located within the nasal cavity, one arose from the maxillary sinus, and one originated in the nasopharynx

Co-delivery of glial cell-derived neurotrophic factor (GDNF) and tauroursodeoxycholic acid (TUDCA) from PLGA microspheres: potential combination therapy for retinal diseases.

Retinitis pigmentosa (RP) is a group of genetically diverse inherited disorders characterised by the progressive photoreceptors and pigment epithelial cell dysfunction leading to central vision impairment. Although important advances in the understanding of the pathophysiologic pathways involved in RP have been made, drug delivery for the treatment of ocular disorders affecting the posterior segment of the eye is still an unmet clinical need. In the present study, we describe the development of multi-loaded PLGA-microspheres (MSs) incorporating two neuroprotectants agents (glial cell-line-deri

Controlled release of odontogenic exosomes from a biodegradable vehicle mediates dentinogenesis as a novel biomimetic pulp capping therapy.

Mineralized enamel and dentin provide protection to the dental pulp, which is vital tissue rich with cells, vasculature, and nerves in the inner tooth. Dental caries left untreated threaten exposure of the dental pulp, providing facile access for bacteria to cause severe infection both in the pulp and systemically. Dental materials which stimulate the formation of a protective dentin bridge after insult are necessary to seal the pulp chamber in an effort to maintain natural dentition and prevent pulpal infection. Dental materials to date including calcium hydroxide paste, mineral trioxide aggr

Scalable Production of Multi-Source Hybrid Biomimetic Exosomes Co-Loaded with Therapeutic Plasmids for Treatment of High-Altitude Pulmonary Edema.

Exosomes show therapeutic promise but face limitations in simultaneous targeting and potency. While hybrid exosome strategies combining multiple cell components can address this, current methods rely on native exosome extraction and drug loading, suffering from low yield and encapsulation efficiency. We developed a biological origin-based hybrid biomimetic exosome (BOB-HBE) platform that synthetically assembles exosomes using parent cell-derived components through a water/oil/water emulsion method, overcoming production bottlenecks. For high-altitude pulmonary edema (HAPE) treatment, we engine

Probiotic-derived glucose oxidase exerts therapeutic efficacy against pediatric B-cell acute lymphoblastic leukemia.

Recent advances in therapy have significantly improved long-term survival in pediatric B-cell acute lymphoblastic leukemia (B-ALL). However, relapse still occurs in 15-20 % of cases, and post-relapse survival rates remain suboptimal at 30-60 %. Therefore, novel therapeutic strategies effective against both primary and relapsed B-ALL are needed. In this study, we treated B-ALL cells with the cell-free culture supernatants (CFCS) of various probiotic strains and found that Aspergillus niger exerted the most potent inhibitory effect on B-ALL cell viability. Liquid chromatography-mass spectrometry

CD56-targeted in vivo genetic engineering of natural killer cells mediates immunotherapy for acute myeloid leukemia.

Acute myeloid leukemia (AML) is a heterogeneous hematological malignancy that starts from bone marrow and spreads to other organs. At the time of diagnosis, both innate and defective natural killer (NK) cells are present in AML patients. The dysfunction of the NK cells is due to the absence of NK cell receptors such as NKG2D on tumor cells that help with tumor immune escape, and also the polycomb protein, EzH2, which plays an important role in the commitment and differentiation of NK cells. The inhibition of EzH2 activates NK cells towards enhanced lytic activity. However, the adoptive transfe

Droplet Microfluidics-Based Fabrication of Monodisperse Poly(ethylene glycol)-Fibrinogen Breast Cancer Microspheres for Automated Drug Screening Applications.

Spheroidal cancer microtissues are highly advantageous for a wide range of biomedical applications, including high-throughput drug screening, multiplexed target validation, mechanistic investigation of tumor-extracellular matrix (ECM) interactions, among others. Current techniques for spheroidal tissue formation rely heavily on self-aggregation of single cancer cells and have substantial limitations in terms of cell-type-specific heterogeneities, uniformity, ease of production and handling, and most importantly, mimicking the complex native tumor microenvironmental conditions in simplistic mod

Paederia scandens-derived exosome-like nanoparticles as a delivery system for andrographolide to treat ulcerative colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by relapsing colon inflammation. Side effects and drug resistance limit current therapies. Andrographolide (AG), an NF-κB pathway inhibitor, shows promise in UC treatment but suffers from poor oral bioavailability. In this study, Paederia scandens-derived exosome-like nanoparticles (P-ELNs) were used as a delivery system to enhance the therapeutic efficacy of AG in UC. P-ELNs were extracted from Paederia scandens leaves and characterized for size, zeta potential, and morphology using transmission electron microscopy

Effects of hydrophobic chain lengths on the structure and properties of polymeric micelles: experimental and simulation studies.

Amphiphilic block copolymers with polyester-based hydrophobic segments, particularly PEG-PLA, exhibit considerable potential as carrier materials for delivering water-insoluble small-molecule drugs via self-assembled micelles. A comprehensive understanding of relationship between carrier material structures and micelle properties is critical for optimizing the drug-loading capacities, drug release behaviors and tumor cell inhibitory properties of micelle formulations. This study aims to elucidate the influences of different hydrophobic chain lengths of mPEG-PLLA on self-assembling to micelles

Co-Encapsulation of Curcumin and α-Tocopherol in Bicosome Systems: Physicochemical Properties and Biological Activity.

A novel co-encapsulation system called bicosomes (bicelles within liposomes) has been developed to overcome the limitations associated with the topical application of curcumin (cur) and α-tocopherol (α-toc). The physicochemical properties and biological activity in vitro of bicosome systems were evaluated. Bicelles were prepared with DPPC, DHPC, cur, and α-toc (cur/α-toc-bicelles). Liposomal vesicles loading cur/α-toc-bicelles were prepared with Lipoid P-100 and cholesterol-forming cur/α-toc-bicosomes. Three cur/α-toc-bicosomes were evaluated using different total lipid percentages (12, 16, an

Vaccination of carp against SVCV with an oral DNA vaccine or an insect cells-based subunit vaccine.

We recently reported on a successful vaccine for carp against SVCV based on the intramuscular injection of a DNA plasmid encoding the SVCV glycoprotein (SVCV-G). This shows that the intramuscular (i.m.) route of vaccination is suitable to trigger protective responses against SVCV, and that the SVCV G-protein is a suitable vaccine antigen. Yet, despite the general success of DNA vaccines, especially against fish rhabdoviruses, their practical implementation still faces legislative as well as consumer's acceptance concerns. Furthermore, the i.m. route of plasmid administration is not easily comb

Preparation, characterization and properties of sterically stabilized paclitaxel-containing liposomes.

Paclitaxel (Taxol) is a diterpenoid isolated from Taxus brevifolia, approved by the FDA for the treatment of ovarian and breast cancers. Due to its low solubility in water, it is clinically administered dissolved in Cremophor EL, (polyethoxylated castor oil) and ethanol, which cause serious side effects. Inclusion of paclitaxel in liposomal formulations has proved to be a good approach to eliminating this vehicle and improving the drug's antitumor efficacy. We prepared different conventional and PEGylated liposomes containing paclitaxel and determined encapsulation efficiency, physical stabili

Bioengineered curcumin-SCFA co-loaded PLGA nanoparticles for modulating inflammation and epithelial barrier integrity in radiation-induced enteritis.

Radiation enteritis (RE) is a common side effect of abdominal radiotherapy, which causes intestinal inflammation and impairment of epithelial barrier. In this study, we developed a bioengineered dual-delivery nanosystem by encapsulating curcumin and microbiota-derived short-chain fatty acids (SCFAs: acetate, propionate, and butyrate) into PLGA nanoparticles (Cur-PLGA NPs@SCFAs). The formulation encapsulated 66% of curcumin and released it slowly over 48 h, with about 72.6% of it being released at pH 7.4. In vitro studies using Caco-2 cells, the Cur-PLGA NPs@SCFAs maintained good cytocompatibil

Chk1-Targeted Hesperidin Liposomes for Breast Cancer Therapy: An Integrated In Silico, In Vitro, and In Vivo Evaluation.

Checkpoint 1 kinase (Chk1) regulates the cell cycle and facilitates DNA repair processes, which are essential for maintaining genomic integrity. In breast cancer, Chk1 is frequently overexpressed, enabling tumor cells to endure DNA damage. By blocking Chk1, a crucial regulator of the development and spread of malignancies, the current study investigates the potential of natural products to identify different treatment approaches for breast cancer. Based on the molecular docking, absorption, distribution, metabolism, excretion, and toxicity profiling and molecular dynamics simulations, hesperid
