Epidemiology, morphology, and molecular characterization of Stephanurus dentatus (Nematoda: Syngamidae) in wild boars from southwestern South Korea.

BACKGROUND: Stephanurus dentatus Diesing, 1839 (Nematoda: Syngamidae), commonly known as the swine kidney worm, parasitizes the renal pelvis, ureters, and perirenal fat of pigs and wild boars, occasionally causing severe pathological effects. Despite reports of low prevalence in domestic pigs in South Korea, no epidemiological data exist for wild boars. This study aimed to investigate the prevalence, morphology, and molecular characteristics of S. dentatus infection in wild boars from the southwestern regions of South Korea. METHODS: A total of 167 wild boars were examined between 2009 and 20

Phyllodistomum kupermani n. sp. from the European perch, Perca fluviatilis L. (Perciformes: Percidae), and redescription of Phyllodistomum macrocotyle (Lühe, 1909) with notes on the species diversity and host specificity in the European Phyllodistomum spp. (Trematoda: Gorgoderidae).

BACKGROUND: European species of the large genus Phyllodistomum Braun, 1899 had historically been erected based solely on morphological characters. Unfortunately, many of them are still poorly known and inadequately described. Molecular approaches are critical to delineate species which were impossible to differentiate based on morphology alone. METHODS: New samples of adult Phyllodistomum spp. were collected from the urinary bladder and/or ureters of European freshwater fishes and fixed to conduct a light and scanning electron microscopy study, and to obtain sequences of nuclear (ITS2 spacer

Genotyping and phylogenetic analysis of unusually located hydatid cysts isolated from humans in north-east Iran.

Despite several studies conducted to determine the genotypes of cystic echinococcosis (CE) agents in humans and other intermediate hosts, the relationship between parasite genotype and clinical presentation of hydatidosis is yet to be well defined. The aim of this study was to compare the genotypes/haplotypes of CE agents of human extra-hepatopulmonary hydatid cysts and common hydatid cysts of the liver. A comparative analysis was carried out between partial cox1 sequences of ten extra-hepatopulmonary hydatid cysts, two liver cysts and reliable sequences from the GenBank database. All the stud

Phylogenetic Position of Pseudosellacotyla lutzi (Freitas, 1941) (Digenea: Cryptogonimidae), A Parasite of Hoplias malabaricus (Bloch) in South America, through 28S rDNA Sequences, and New Observations of the Ultrastructure of Their Tegument.

The freshwater fish digenean Pseudosellacotyla lutzi ( Freitas, 1941 ) Yamaguti, 1954 has had an unsettled taxonomic history, and has at various times been classified as a member of Nanophyetidae, Heterophyidae, Microphallidae, Faustulidae, and Cryptogonimidae. Nine individual specimens of the trahira, Hoplias malabaricus (Bloch, 1794), were sampled in the Paraná River basin, Paraná State, Brazil; 22 specimens of P. lutzi were collected. One specimen of P. lutzi was used to obtain a sequence of the domains D1-D3 of the 28S rRNA gene, and to perform a phylogenetic analysis to assess their posit

Genotype and Phenotype of Echinococcus granulosus Derived from Wild Sheep (Ovis orientalis) in Iran.

The aim of the present study is to determine the characteristics of genotype and phenotype of Echinococcus granulosus derived from wild sheep and to compare them with the strains of E. granulosus sensu stricto (sheep-dog) and E. granulosus camel strain (camel-dog) in Iran. In Khojir National Park, near Tehran, Iran, a fertile hydatid cyst was recently found in the liver of a dead wild sheep (Ovis orientalis). The number of protoscolices (n=6,000) proved enough for an experimental infection in a dog. The characteristics of large and small hooks of metacestode were statistically determined as th

Molecular and phylogenetic status of Fasciola sp., of cattle in Qena, Upper Egypt.

The species of liver fluke of the genus Fasciola (phylum platyhelminthes, order Digenea, Family Fasciolidae) are obligatory parasites that inhabit the large biliary ducts of herbivore animals as well as man. Reports on the species of Fasciola present in the Nile Delta, Egypt, appear controversial. In the current study a precise identification of Fasciola isolates from cattle in Qena province, Upper Egypt was done based on examination of the second Internal Transcribed Spacer (ITS2) and the mitochondrial cytochrome oxidase subunit I (COI). Amplification, sequencing and phylogenetic examination

Is Dirofilaria repens endemic in the Havelland district in Brandenburg, Germany?

In Brandenburg, northeastern Germany, an outbreak of zoonotic Dirofilaria repens in a sledge dog kennel was detected in January, 2012. Using the modified Knott test, 8 out of 28 adult dogs were microfilaria positive. A D. repens-specific PCR based on the internal transcribed spacer 2 region detected filarial DNA in 11 of these dogs. Northeastern Germany is currently considered to be free of Dirofilaria. The 39% prevalence within the kennel and the fact that the dogs transiently stayed in endemic areas only during the winter suggest that autochthonous infections occurred. Analysis of weather da

PCR assays for detection of Baylisascaris procyonis eggs and larvae.

The objective of this study was to develop polymerase chain reaction (PCR) assays for detection of Baylisascaris procyonis eggs and larvae in fecal, environmental, and tissue samples. We have optimized conventional and real-time PCR assays for B. procyonis using the mitochondrial cytochrome oxidase 2 gene as the target for amplification. The lower limit of detection of the parasite genomic DNA was 10 pg in the conventional PCR and 100 fg in the real-time PCR. In both PCR assays, specific amplification of a 146 bp product was achieved with DNA extracted from a single in vitro hatched B. procyon

The FAR proteins of filarial nematodes: secretion, glycosylation and lipid binding characteristics.

The FAR proteins of nematodes are small ( approximately 20 kDa), helix-rich, fatty acid and retinol-binding (FAR) proteins that appear to be confined to nematodes. We have carried out a comparative sequence and biochemical analysis of selected FAR proteins often species of filarial parasites (from the genera Onchocerca, Brugia, Wuchereria, Loa, Acanthocheilonema and Litomosoides). The sequences fall into two main groups corresponding broadly to the onchocercal and lymphatic filariasis parasites, and only those with unsheathed microfilariae were found to produce glycosylated FAR proteins. The p

sem-4 promotes vulval cell-fate determination in Caenorhabditis elegans through regulation of lin-39 Hox.

Vulval cell-fate determination in Caenorhabditis elegans requires the action of numerous gene products, including components of the Ras/Raf/MAPK signaling cascade and the hox gene lin-39. sem-4 encodes a zinc finger protein with previously characterized roles in fate specification of sex myoblasts, coelomocytes, and multiple neuronal lineages in C. elegans (M. Basson and R. Horvitz, 1996, Genes Dev. 10, 1953-1965). By characterizing three new alleles of sem-4 that we identified in a screen for vulval-defective mutants, we determined that loss of sem-4 activity results in abnormal specification

Ascaris infections in humans from North America: molecular evidence for cross-infection.

Cases of human Ascaris infection occur sporadically in areas such as N. America and Western Europe, where this parasite is thought to be non-endemic. Clinical case histories suggest that many of these cases may be cross-infections from pigs. I describe patterns of variation in the ribosomal DNA of Ascaris from 9 such cases. For comparative purposes, I also describe patterns of variation in parasites obtained from pigs and humans from worldwide locations. A Hae III restriction site distinguishes two classes of rDNA repeats; repeats bearing this restriction site were found in > 96% of parasites

DNA probe-based classification of Simulium damnosum s. l.-borne and human-derived filarial parasites in the onchocerciasis control program area.

The development of polymerase chain reaction-based methods using strain- and species-specific DNA probes for Onchocerca volvulus has permitted classification of individual parasites from every stage of the parasite's life cycle. This technology has been applied on a large scale basis by Onchocerciasis Control Program (OCP) in West Africa. The primary objective of the OCP in using the DNA probes was to obtain accurate estimates of the annual transmission potential of the blinding strain of O. volvulus. The DNA probe classification of larvae collected throughout the OCP area demonstrated that la

Molecular characterization of muscle-parasitizing didymozoids in marine fishes.

Classification and identification of muscle-parasitizing didymozoids found in marine fish is difficult because of their novel parasitism and morphology. Recent sequence analysis has helped, but only seven sequences are available. Therefore, the usefulness of molecular methods for differentiation of muscle-parasitizing didymozoids, as well as genetic differences between the muscle and the other site-parasitizing didymozoids are quite unclear. In the present study, six unidentified didymozoid isolates from the trunk muscles of four marine fish species (Diagramma pictum, Plectorhinchus cinctus, P

Optimization and validation of a TaqMan real-time PCR for the detection of Heterobilharzia americana in dog feces.

Heterobilharzia americana is a trematode parasite that causes canine schistosomiasis, a disease capable of causing significant morbidity and mortality in dogs. Reliable diagnosis is essential for limiting disease progression. This study aimed to optimize and validate a previously developed TaqMan qPCR assay for detection of H. americana DNA in feces. The assay targets a highly repetitive non-coding DNA sequence located across multiple loci within the H. americana genome. PCR optimization involved gradient thermocycling and serial dilutions to refine annealing temperature and efficiency. PCR pr

Entomological survey of Dirofilaria spp. in Sardinia (Italy): molecular detection and mosquito species distribution.

BACKGROUND: Dirofilariosis, caused by Dirofilaria immitis and Dirofilaria repens, is expanding across Europe because of climate change, raising veterinary and zoonotic concerns. Among over 70 mosquito species considered to be implicated in their transmission, Culex pipiens and Aedes albopictus are recognized as major vectors. In Sardinia, where canine dirofilariosis is endemic, entomological information on Dirofilaria circulation was previously lacking. This study investigated mosquito species composition and assessed the presence of D. immitis and D. repens across five sites on the island. M
