[The differentiation of viruses of the tick-borne encephalitis complex by means of RNA-DNA hybridization].

Nucleic acid spot hybridization with cloned cDNA of tick-borne encephalitis (TBE) virus, strain Sofjin, was used to differentiate strains of TBE and other flaviviruses. The cDNA probe reacted with strains of TBE and flaviviruses of TBE subgroup with the exception of Powassan virus. The probe did not react with viruses of Japanese encephalitis and Gendue subgroups. The viruses of TBE subgroup and some strains of TBE virus were differentiated from TBE strain Sofjin by thermal stability of RNA-DNA hybrids. Negishi and Louping ill viruses were found to be most closely related to TBE strain Sofjin

Identification of rabbit immunoglobulin latent Ckappa1 allotype genes alters the concept of allelic inheritance.

In the rabbit, expression of immunoglobulin Ckappa1 light chain genes is believed to be under allelic control. Conventionally, four nominal allotypic variants, b4, b5, b6 and b9 have been shown to be co-dominantly expressed at the Ckappa1 gene locus. Analogously, the heavy chain allotypes, VHa1, VHa2 and VHa3, found in the V region, are also believed to be inherited co-dominantly. However, after our earlier discovery of non-allelic or latent allotypes in the serum and on cell surfaces. we subsequently reported that cDNA sequences for latent b5 and b6 were identical to nominal b5 and b6, respec

Role of ROS modified human DNA in the pathogenesis and etiology of cancer.

The effect of hydroxyl radical, generated by ultraviolet (UV) irradiation of hydrogen peroxide, on human placental DNA was monitored by UV spectroscopy, melting temperature studies, S1 nuclease digestibility and hydroxyapatite column chromatography. Immunological data indicated that reactive oxygen species (ROS) modified human DNA induced high titer antibodies. In ELISA, serum antibodies from various cancer patients showed a higher recognition of ROS-human DNA as compared to native DNA. Retarded mobility of the immune complex formed between IgG, isolated from cancer sera, and ROS-human DNA pro

Perinatal findings and molecular cytogenetic analysis of trisomy 16q and 22q13.3 deletion.

OBJECTIVES: To present the perinatal findings and molecular cytogenetic analysis of a case with concomitant trisomy 16q and 22q13.3 deletion … 1 (present) and D22S1168 (absent). CONCLUSION: The use of LSI DiGeorge/VCFS region/ARSA dual color DNA probes to examine distal 22q would miss some subtle terminal deletions of 22q13. However, the use of 22q telomeric probes would detect these minute deletions. Fetuses having trisomy 16q and 22q13.3 deletion may prenatally manifest decreased fetal movement, …

Prenatal molecular cytogenetic diagnosis of partial tetrasomy 10p due to neocentromere formation in an inversion duplication analphoid marker chromosome.

Neocentromeres are fully functional centromeres found on rearranged or marker chromosomes that have separated from endogenous centromeres. N … a chromosome 10 origin of the marker, a complete panel of chromosome-specific centromeric satellite DNA probes failed to hybridize to any portion of the marker. The presence of a neocentromere on the marker chromosome was confirmed by the absence of hybridization of an all-human-centromere alpha-satellite DNA probe, which hybridizes to all normal centromeres …

Modified primers for rapid and direct electrochemical analysis of coeliac disease associated HLA alleles.

Direct detection of PCR product via hybridisation assay, would facilitate the development of rapid tools for genetic analysis. Here, a PCR primer designed to generate a PCR amplicon tagged with single stranded DNA tails at each end of the duplex, which can be used for direct hybridisation with a surface immobilised probe and an enzyme labelled reporter probe is presented. Four modified sequence specific primers (SSP) pairs were designed for the selective amplification of coeliac disease associated alleles (DQA1*05, DQB1*02, DQB1*03:02 alleles), and human growth hormone (positive control). Fluorescence lifetime imaging of G-quadruplex RNA dynamics in Alzheimer's disease using a novel nucleic acid-sensitive probe.

Real-time monitoring of G-quadruplex (G4) RNA dynamics is vital for understanding their biological roles in the progression and treatment of Alzheimer's disease. However, this remains challenging due to two key difficulties. First, current fluorescent probes lack sufficient selectivity for G4 RNA over G4 DNA and other non-G4 secondary structures in live-cell competitive environments. Second, fluorescence intensity-based imaging cannot detect subtle changes in G4 RNA because of variations in fluorophore uptake and photobleaching. Herein, we report a novel thiazole orange derivative (TOGR) for f

Linkage exclusion between the autosomal dominant polycystic kidney disease locus and chromosome 16 markers in a new family.

A family segregating for autosomal dominant polycystic kidney disease (ADPKD) is reported. The clinical picture was typical for ADPKD in some family members, although others showed mild involvement. DNA from family members was probed with seven chromosome 16 single-copy DNA sequences that mapped to the telomere of the short arm of the chromosome. The most likely order of six of the probes from the telomere is palpha3'HVR.64 at the designated locus D16S85, CRI-0327 at D16S63, CRI-090 at D16S45, CRI-0129 at D16S56, CRI-0133 at D16S58, and CRI-0136 at D16S60, with the PKD1 locus for ADPKD between

Label- and wash-free electrochemical nucleic acid detection via a LAMP platform coupled with silver ion aptamer probes.

We present a novel electrochemical nucleic acid detection strategy based on loop-mediated isothermal amplification (LAMP) coupled with silver ion (Ag+) aptamer probes. The strategy employs specially designed hairpin-shaped probes comprising an Ag+-binding aptamer sequence in the stem region and a target-complementary sequence in the loop region. In the presence of the target nucleic acid, the LAMP reaction is initiated, and the resulting amplicons hybridize with the loop region of the probe. This interaction disrupts the hairpin structure, leading to the release of Ag+ from the stem region. Th

Reversible RNA ADP-ribosylation on uracil bases.

ADP-ribosylation is a conserved modification that uses NAD+ as a co-substrate to regulate essential cellular processes, such as genome stability and transcription, with Poly(ADP-ribose) Polymerases (PARPs) serving as the major catalyzing enzymes in humans. Historically defined as a protein post-translational modification, ADP-ribosylation on nucleic acids has been increasingly recognized in recent years, particularly in bacterial systems, but remains poorly understood in higher organisms. Here, we identify human PARP10 as a candidate enzyme that ADP-ribosylates nucleic acid bases, showing appa

Comprehensive analysis of pore properties of ultra-wide pore size exclusion chromatography packing materials.

Accurate characterization of size exclusion chromatography (SEC) packing materials is critical for understanding separation behavior, particularly for the emerging class of large biotherapeutic and nanoparticle analytes. In this study, we present a comprehensive and multi-technique assessment of the pore structure of modern wide-pore and ultra-wide-pore SEC columns. Mercury intrusion porosimetry (MIP) and advanced scanning electron microscopy (SEM) image analysis with broad ion beam milling were employed alongside SEC calibration using rigid gold nanoparticles (AuNPs) and double-stranded DNA (

Detection of NSCLC biomarker miRNAs via localized catalytic hairpin self-assembly and laser-induced fluorescence on a microfluidic platform.

This study introduces an innovative microfluidic platform integrating localized catalytic hairpin assembly (LCHA) amplification with laser-induced fluorescence (LIF), for sensitive detection of non-small cell lung cancer (NSCLC) biomarkers miRNA-20a and miRNA-223. The enzyme-free LCHA strategy leverages two hairpin probes (Hp1/Hp2) anchored on a DNA tetrahedron framework, where spatial confinement enhances reaction kinetics by elevating local probe concentrations. Hp2 is dual-labeled with 6-FAM and BHQ-1 for fluorescence quenching control. Target miRNA initiates a cascade reaction through magn

Bivalent Display of Dicysteine on Peptide Nucleic Acids for Homogenous DNA/RNA Detection through in Situ Fluorescence Labelling.

Fluorogenic probes that signal the presence of specific DNA or RNA sequences are key enabling tools for molecular disease diagnosis and imaging studies. Usually, at least one fluorophore is attached through covalent bonding to an oligonucleotide probe. However, the additional conjugation step increases costs. Here we introduce a method that avoids the requirement for the preparation of fluorescence-labelled oligonucleotides and provides the opportunity to alter the fluorogenic reporter dye without resynthesis. The method is based on adjacent hybridization of two dicysteine-containing peptide n
