Bidirectional neutrophil-macrophage interactions as therapeutic leverage points in sepsis.

Sepsis is a life-threatening condition driven by a dysregulated host response to infection, in which the innate immune axis-particularly the crosstalk between neutrophils and macrophages-plays a central role in dictating disease trajectory. This review delineates the functional, metabolic, and spatial characteristics of these two phagocytic cell types and highlights their bidirectional interactions across distinct immunological phases of sepsis. Multiple intercellular conduits, including extracellular vesicles, cytokine-chemokine axes, extracellular traps, and efferocytosis, mediate context-sp…

Mapping the interaction site for a β-scorpion toxin in the pore module of domain III of voltage-gated Na(+) channels.

Activation of voltage-gated sodium (Na(v)) channels initiates and propagates action potentials in electrically excitable cells. β-Scorpion toxins, including toxin IV from Centruroides suffusus suffusus (CssIV), enhance activation of Na(V) channels. CssIV stabilizes the voltage sensor in domain II in its activated state via a voltage-sensor trapping mechanism. Amino acid residues required for the action of CssIV have been identified in the S1-S2 and S3-S4 extracellular loops of domain II. The extracellular loops of domain III are also involved in toxin action, but individual amino acid residues

Calcium-containing crystals enhance receptor activator of nuclear factor κB ligand/macrophage colony-stimulating factor-mediated osteoclastogenesis via extracellular-signal-regulated kinase and p38 pathways.

OBJECTIVE: Diseases associated with calcium-containing crystal deposition can lead to local bone erosion. We aimed to determine whether calcium-containing crystal-hydroxyapatite, β-tricalcium phosphate and CPPD enhanced osteoclastogenesis and to define underlying mechanisms of action. METHODS: Osteoclastogenesis was studied by culturing murine RAW 264.7 osteoclast precursor cells with RANK ligand (RANKL)/ M-CSF and/or calcium-containing crystals, and observing the tartrate-resistant acid phosphatase (TRAP)-positive multinucleated cells and TRAP activity. Resorption pit formation was used to e

METTL7A-Dependent m6A Methylation Orchestrates CXCR4hi Neutrophil Pathogenicity and NETosis in Skin Inflammation.

Neutrophil extracellular traps (NETs) released by C-X-C motif chemokine receptor 4-high (CXCR4hi) neutrophils are major contributors to skin inflammation in psoriasis, yet the upstream epitranscriptomic mechanisms that regulate NETosis remain poorly defined. Here, we investigated the role of methyltransferase-like protein 7A (METTL7A) in N 6-methyladenosine (m6A)-mediated neutrophil activation and psoriasis pathogenesis. Using transcriptomic profiling of CXCR4hi neutrophils from psoriasis patients, m6A RNA immunoprecipitation, in vitro functional assays, and in vivo murine models of skin infla…

Neutrophils display distinct post-translational modifications in response to varied pathological stimuli.

Neutrophils play a vital role in the innate immunity by perform effector functions through phagocytosis, degranulation, and forming extracellular traps. However, over-functioning of neutrophils has been associated with sterile inflammation such as Type 2 Diabetes, atherosclerosis, cancer and autoimmune disorders. Neutrophils exhibiting phenotypical and functional heterogeneity in both homeostatic and pathological conditions suggests distinct signaling pathways are activated in disease-specific stimuli and alter neutrophil functions. Hence, we examined mass spectrometry based post-translational…

Spin-trapping of free radicals during phthalocyanine photosensitization of lymphoma cells in vitro.

Spin-trapping and electron spin resonance (ESR) spectroscopy were used to detect free radicals generated during light exposure of lymphoma cells sensitized in vitro by metallotetrasulfophthalocyanines (Al-PcS4 and Zn-PcS4). 5,5-dimethyl-1-pyrroline-1-oxide (DMPO) and alpha-phenyl-beta-tert-butylnitrone (PBN) were used as spin-trapping agents. Hydroxyl radical spin-adducts were detected under conditions of both extracellular and intracellular photosensitization. In addition, organic radicals of different origin and/or variable yields were trapped, depending on the photosensitization conditions

Neutrophils in pediatric autoimmune disease.

PURPOSE OF REVIEW: As first immune responders, neutrophils are essential mediators of host defense, and also contribute to chronic pathologic inflammation at the crossroads of innate and adaptive immunity. In this review, we will highlight the current understanding of the role of neutrophils in pediatric rheumatology, with a focus on juvenile idiopathic arthritis (JIA) and lupus. RECENT FINDINGS: In inflamed tissues, neutrophils extrude neutrophil extracellular traps containing autoantigen that potentially drives lupus and rheumatoid factor-positive JIA. However, the contribution of neutrophi…

Voltage sensor-trapping: enhanced activation of sodium channels by beta-scorpion toxin bound to the S3-S4 loop in domain II.

Polypeptide neurotoxins alter ion channel gating by binding to extracellular receptor sites, even though the voltage sensors are in their S4 transmembrane segments. By analysis of sodium channel chimeras, a beta-scorpion toxin is shown here to negatively shift voltage dependence of activation and enhance closed state inactivation by binding to a receptor site that requires glycine 845 (Gly-845) in the S3-S4 loop at the extracellular end of the S4 segment in domain II of the alpha subunit. Toxin action requires prior depolarization to drive the S4 voltage sensors outward, but these effects are

Cellular stress, cell death, and extracellular vesicles: redefining the therapeutic landscape of rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disease driven by dysregulated immune responses, persistent synovial inflammation, and progressive joint destruction. Neutrophil extracellular trap-mediated cell death (NETosis) and autophagy are interrelated processes that amplify inflammation and perpetuate autoimmunity in a feed-forward manner, accelerating synovial damage. Mesenchymal stem cells (MSCs) have attracted interest due to their strong immunomodulatory and regenerative potential; however, the safety concerns, engraftment, and long-term persistence led to focusing on mesenchymal st

Pro-inflammatory mediators and neutrophils are increased in synovial fluid from heifers with acute ruminal acidosis.

BACKGROUND: Acute ruminal acidosis (ARA) is a metabolic disease of cattle characterized by an aseptic synovitis. ARA is the result of an increased intake of highly fermentable carbohydrates that frequently occurs in dairy cattle subjected to high production requirements. In human joint diseases such as rheumatoid arthritis and gout, several pro-inflammatory molecules are increased in the synovial fluid, including cytokines, prostaglandin E2 (PGE2), metalloproteinases, and neutrophil extracellular traps (NETs). The aim of this study was to identify the presence of proinflammatory mediators and …

Mechanisms of Complement Activation and NETosis Interplay in Early Pregnancy Loss: From Autoimmunity to Thrombogenesis.

Early Pregnancy Loss (EPL) represents a formidable challenge in reproductive medicine. Its complex pathophysiology is primarily characterized by dysregulation of both the immune and coagulation systems. In recent years, abnormal activation of the complement system and neutrophil extracellular trap formation (NETosis) have received increasing scientific attention as key molecular events underlying EPL. This interest is fueled significantly by compelling evidence from non-pregnant models, such as antiphospholipid syndrome (APS) and severe infections (e.g., COVID-19), which demonstrate a pathogen

Identification and validation of NETs-associated biomarkers in osteoporosis with diabetes.

Osteoporosis and diabetes represent major global public health challenges. Neutrophil extracellular traps (NETs) serve as key components of the innate immune system by capturing and eliminating pathogens. This exploratory study aimed to preliminarily identify biomarkers associated with NETs in osteoporosis with diabetes and to provide initial insights into underlying molecular mechanisms. A transcriptomic sequencing dataset was integrated to analyze the molecular profiles of comorbid osteoporosis and diabetes (OP-DM). The NETs-related genes (NETs-RGs) were curated from previous literature. As …

Neutrophils as critical orchestrators of chronic inflammation.

Neutrophils are the first key effector innate immune cells recruited toward inflammatory sites. Through the release of neutrophilic extracellular traps (NETs), the production of reactive oxygen species (ROS), degranulation and phagocytosis, neutrophils play a central role in the rapid elimination of invading pathogens. Recently, increasing attention has been given to the role of neutrophils in chronic inflammation, challenging the dichotomy between innate and adaptive immune responses. In chronic inflammatory conditions, neutrophils generally display a hyperinflammatory phenotype via dysregula…

Graduate Student Literature Review: Standardized approaches for neutrophil functional assessment in dairy cattle-A practical guide.

Polymorphonuclear leukocytes (neutrophils) serve as critical indicators of innate immune function in dairy cattle. Accurate assessment of their functional capacities is essential for understanding the status of the innate immune system in health and disease or following immunomodulatory treatment. However, the lack of standardized protocols for evaluating PMN functional capacity in dairy cattle has led to inconsistencies in research methodologies and hindered cross-laboratory comparisons. To address this issue, we propose systematic, evidence-based methodologies for assessing PMN function in d

Neutrophil Fate and Function in Gout: From Sterile Inflammation to Resolution.

Gout is a prototypical sterile inflammatory disease caused by the deposition of monosodium urate (MSU) crystals in joints and periarticular tissues. Although acute gout flares are characterized by rapid and intense neutrophil-dominated inflammation, they often resolve spontaneously, suggesting that neutrophils may contribute not only to inflammatory amplification but also to endogenous resolution. In this review, we summarize current evidence on the multifaceted roles of neutrophils in gout across different stages of disease. We discuss how MSU crystals activate innate immune signaling and pro
