A Photoresponsive Azo-Functionalized 2,4-Dinitrophenylhydrazone Polymer for Selective Fluoride Sensing in Environmental and Biological Media.

Fluoride (F-) plays an essential role in dental health and osteoporosis treatment; however, excessive intake can lead to severe disorders, including dental fluorosis and urolithiasis, necessitating sensitive monitoring in environmental and biological systems. Herein, we report a photoresponsive azo-2,4-dinitrophenylhydrazone (AZO-DNP) platform comprising monomeric and polymeric architectures for selective fluoride detection. Both systems enable rapid naked-eye recognition through an intense yellow-to-purple color transition, accompanied by a pronounced bathochromic shift and ratiometric UV-vis

Assessment of hydrochemistry, fluoride distribution, and non-carcinogenic health risks in groundwater of the Manyara region, Tanzania.

Fluoride (F-) contamination in groundwater is a major global public health concern. Prolonged intake of F⁻ above 1.5 mg/L and 10 mg/L may lead to skeletal fluorosis and crippling fluorosis, respectively. The Manyara region, located within Tanzania's fluoride belt in the Eastern African Rift Valley, is one of the areas most affected by elevated F- levels in groundwater. The extent of F⁻ pollution and associated health risks in this region remains poorly documented. This study assessed the hydrochemistry, spatial distribution of F-, non-carcinogenic health risks, and the suitability of groundwat

Effect of a sodium fluoride rinse on the composition of early oral biofilms formed in situ on enamel and dentine.

OBJECTIVES: Dental caries results from dysbiotic shifts in the microbial community, so modulating biofilm composition represents a valid preventive strategy. The aim of this double-blind randomized crossover in situ study was to investigate how a fluoride rinse can modify the composition of the early oral biofilm formed on enamel and dentine. MATERIALS AND METHODS: Twelve volunteers wore mandibular appliances containing 3 enamel and 3 dentine specimens for 1 min, to allow basal pellicle formation, then rinsed (1 min) with a NaF solution (500 ppm F⁻) or deionized water (DW) and kept the applia

Fluoride induced chondrocyte extracellular matrix degradation by inhibition of glycolysis by the PHD2/HIF1α signaling pathway.

Studies have shown that fluoride induces cartilage damage, however, the specific mechanism is unknown. In this study, we investigated the effect of fluoride on extracellular matrix metabolism and its molecular regulatory mechanism, using well-established experimental models SW1353 cell line and neonatal rat tibia organ culture. We found that 5 × 10-4 M fluoride reduced proteoglycan synthesis by 20%, significantly downregulated the expression levels of Aggrecan, Collagen Type II and X Alpha 1 Chain, and upregulated various matrix metalloproteinases. Based on RNA sequencing results, we found tha

Silver diamine fluoride, a caries restorative agent, induces cytotoxicity in primary human gingival fibroblasts.

Silver diamine fluoride (SDF) is commonly used in clinical settings to manage dental caries in young children. A major drawback of SDF is its tendency to darken carious lesions, turning them brown or black, which has drawn criticism. This study investigates the cytotoxic effects of SDF on human gingival fibroblasts (hGF).hGF cells were exposed to varying concentrations of SDF (0.0001%, 0.001%, and 0.005%) for a 24-hour period. We analysed cytotoxicity, the potential for reactive oxygen species (ROS) production, mitochondrial function, morphological indicators of apoptosis, and key molecular ma

The impact of light curing on the efficacy of silver diamine fluoride in arresting dentinal caries: an in vitro study.

OBJECTIVE: Silver diamine fluoride (SDF) is widely used for caries arrest due to its ease of application, minimal caries removal, low patient discomfort, and cost-effectiveness. However, the recommended 60-second (sec) application time may be impractical in patients with limited compliance. This study evaluated the effects of 10-sec versus 60-sec SDF application, with and without light curing, on arresting artificial dentinal caries by analyzing three parameters-percentage change in mean lesion depth (%∆LD), percentage change in mean mineral density (%∆MD), and penetration depth (PD)- assessin

Silver Diamine Fluoride in Arresting Dental Caries Among Young Children: a Randomized Clinical Trial.

BACKGROUND: Dental caries is a biofilm-mediated, sugar-driven disease in which acids from bacterial fermentation of dietary carbohydrates demineralize tooth structure. While early enamel lesions are reversible, progression into dentin allows rapid bacterial advancement toward the pulp due to the tissue's tubular structure. OBJECTIVE: The primary objective of this study is to evaluate the effectiveness of SDF as a minimally invasive approach for arresting dental caries in young children. The secondary objective was to determine whether the alternating SDF+ FV regimen achieves higher caries arr

Aluminum-[18F]fluoride radiolabeling of triarylphosphines for cell labeling via the perfluoroaryl azide Staudinger ligation.

BACKGROUND: The development of safer and more effective cell-based therapies requires robust methods for tracking cells in vivo. Positron emission tomography (PET) is a highly sensitive nuclear imaging technique capable of quantitatively tracking the in vivo fate of cells after administration. Here, we investigated a cell-labeling strategy based on metabolic glycoengineering (MGE) to introduce azides on the cell surface, followed by radiolabeling via the bioorthogonal perfluoroaryl azide (PFAA)-Staudinger reaction. We studied the metabolic incorporation of a tetraacetylated PFAA-derivatized ma

High-valent sulfur fluorides as reactivity switches for PFAS-free benzene-azepine skeletal editing.

The development of non-PFAS fluorinated heterocycles is a critical challenge in sustainable molecular design. Here we report a reactivity-switching skeletal editing strategy that transforms benzenes into azepines through visible-light activation mediated by high-valent sulfur fluorides. The pentafluorosulfanyl (SF5) and tetrafluorosulfanyl (SF4) groups act as powerful electronic activators that stabilize singlet nitrenes, lowering the barrier for 6π-electrocyclization and enabling efficient benzene-to-azepine interconversion in high yields. Mechanistic and DFT studies reveal that the strong el
