BACKGROUND AND OBJECTIVE: Small lymphocytic lymphoma (SLL) is morphologically and immunologically similar to chronic lymphocytic leukemia (CLL), and the new REAL classification refers to them as a single disease termed SLL/CLL. Recently, frequent losses in 6q, 11q and/or 13q were observed in CLL using comparative genomic hybridization (CGH). We performed CGH analyses in order to find out whether these two entities contain the same DNA copy number changes.
DESIGN AND METHODS: Seventeen patients with stage IV disease and one with stage III disease were studied by CGH. CGH is based on quantitati

The micronucleus assay is a convenient, in situ method for observing cell damage resulting from exposure to clastogenic agents and has been widely used as a dosimeter of human exposure to radiation or chemicals. It also is a complement to the classic clonogenic cell survival assay in that it can be used to examine radiation damage vs dose as a function of cell type or radiation quality. Digitized imaging densitometry was conducted on CHO cells that have undergone one division and in which further cytokinesis was blocked to collect data on the distributions of percentage total cellular DNA per

It is of critical importance to our understanding of Alzheimer's disease (AD) pathology to determine how key pathological factors are interconnected and implicated in nerve cell death, clinical symptoms, and disease progression. The formation of extracellular beta-amyloid (Aβ) plaques is the major pathological hallmark of AD and Aβ has been suggested to be a critical inducer of AD, driving disease pathogenesis. Exactly how Aβ plaque formation begins and how ongoing plaque deposition proceeds and initiates subsequent neurotoxic mechanisms is not well understood. The primary aim of our research

The clinical assessment, treatment, and prognostic evaluation of breast cancer liver metastasis (BCLM) remain clinically challenging. Hepatic involvement is often identified only after substantial intrahepatic tumor burden, radiologically evident parenchymal involvement, abnormal liver function, or symptoms attributable to hepatic impairment have developed. Standard evaluation relies on molecular subtype, tumor burden, liver function, imaging, metastatic-site biopsy, receptor reassessment, and selected genomic testing; however, these variables do not fully explain why only a subset of dissemin

Atypical spindle cell/pleomorphic lipomatous tumor (ASPLT) is a recently defined adipocytic neoplasm with broad morphological variability. While generally benign, a small subset of ASPLT can demonstrate locally aggressive behavior with recurrences after incomplete resection. There is only limited description of the radiologic features of ASPLT in the literature. This case report presents a unique longitudinal case of a 75-year-old woman with an ASPLT in the subcutaneous tissue of the left lower leg. Following two incomplete resections, the tumor recurred twice over 38 months, each time with in

Breast cancer remains one of the leading causes of cancer-related mortality among women worldwide. Early and accurate detection is critical for improving survival rates and enabling effective treatment. This research proposes a fine-tuned Multi-Deep Neural Network model with Multiple Datasets (FT-MDNNMDs). The proposed system employs a hierarchical classification strategy, first distinguishing cancer from normal cases, and subsequently classifying cancer-positive cases into Stage II and Stage III categories. The model integrates data from The Cancer Genome Atlas (TCGA) and a private clinical i

Hepatocellular carcinoma (HCC) exhibits substantial interpatient heterogeneity, leading to markedly variable outcomes and survival even among patients with similar stages and imaging phenotypes. Mainstream staging systems remain suboptimal, whereas pathology-dependent factors and high-cost genomic assays are neither scalable nor timely for clinical decision-making. Existing algorithms provide coarse risk stratification or static binary predictions, failing to capture the time-varying risk of death. We developed and externally validated TEMPO-HCC, a multimodal deep survival model with hierarchi

Auricular anomalies, including microtia and preauricular tags (PATs), are frequently overlooked findings on prenatal ultrasound, yet they may represent the earliest and sometimes the only clue to a wide spectrum of craniofacial and multisystem disorders. Distinguishing isolated anomalies from syndromic conditions remains a major diagnostic challenge in fetal medicine. In this illustrated, state of the art case-based review, we propose a practical, imaging-driven diagnostic framework for the prenatal evaluation of auricular anomalies. Drawing on a decade of clinical experience and a curated ser

AIMS: Accurately and efficiently identifying mitotic figures (MFs) is crucial for diagnosing and grading various cancers, including glioblastoma (GBM), a highly aggressive brain tumour requiring precise and timely intervention. Traditional manual counting of MFs in whole slide images (WSIs) is labour-intensive and prone to interobserver variability. Our study introduces a deep active learning framework that addresses these challenges with minimal human intervention.
METHODS AND RESULTS: We utilized a dataset of GBM WSIs from The Cancer Genome Atlas (TCGA). Our framework integrates convolution

Influenza A virus (IAV) is a major threat to global human health and is a topic of intense research. With the continuous problem of seasonal influenza and the threat of potential pandemics due to frequent emergence of new viral strains, development of new, broad-spectrum antivirals is an urgent priority. In antiviral development against influenza, the process of host cell entry of IAV is of particular interest as inhibiting the virus at the entry step should stop infection early on, blocking the downstream infection processes including viral replication and transcription. Therefore, a detailed

The use of machine learning (ML) in biomarker analysis for predicting Down syndrome exemplifies an innovative strategy that enhances diagnostic accuracy and enables early detection. Recent studies demonstrate the effectiveness of ML algorithms in identifying genetic variations and expression patterns associated with Down syndrome by comparing genomic data from affected individuals and their typically developing peers. This review examines how ML and biomarker analysis improve prenatal screening for Down syndrome. Advancements show that integrating maternal serum markers, nuchal translucency me

Due to the availability of various mouse strains with well-defined genomes, mice have become pivotal models for ocular developmental research. Accurate and highly repeatable biometric measurements are crucial for investigating myopia in experimental animals. This study aimed to develop a precise methodology for comprehensive assessment of ocular biometrics in mice. We developed the custom-built spectral domain optical coherence tomography (SD-OCT) system with an axial resolution of 4.3 μm, and an imaging depth of 4.1 mm in tissue. Three-dimensional biometric parameters exhibited superior preci

Formaldehyde (FA) is both a one-carbon (1C) metabolite and a potent genotoxin in living cells. FA plays beneficial roles in endogenous catabolic processes and cellular signaling, but its potent electrophilicity necessitates strict regulation. This dichotomy is especially important in the nucleus, where endogenously produced FA has been shown to promote toxicity and disease by generating deleterious DNA adducts. More broadly, the sources and scavenging mechanisms of FA differ across subcellular compartments, underscoring the need for imaging sensors with subcellular spatial resolution to accura

The use of functional quantitative biomarkers extracted from routine PET-CT scans to characterize clinical responses in patients with lymphoma is gaining increased attention, and these biomarkers can outperform established clinical risk factors. Total metabolic tumour volume enables individualized estimation of survival outcomes in patients with lymphoma and has shown the potential to predict response to therapy suitable for risk-adapted treatment approaches in clinical trials. The deployment of machine learning tools in molecular imaging research can assist in recognizing complex patterns an

Skull-stripping, an important pre-processing step in neuroimage computing, involves the automated removal of non-brain anatomy (such as the skull, eyes, and ears) from brain images to facilitate brain segmentation and analysis. Manual segmentation is still practiced, but it is time-consuming and highly dependent on the expertise of clinicians or image analysts. Prior studies have developed various skull-stripping algorithms that perform well on brains with mild or no structural abnormalities. Nonetheless, they were not able to address the issue for brains with significant morphological changes

Although there are pronounced sex differences for psychiatric disorders, relatively little has been published on the heterogeneity of sex-specific genetic effects for these traits until very recently for adults. Much less is known about children because most psychiatric disorders will not manifest until later in life and existing studies for children on psychiatric traits such as cognitive functions are underpowered. We used results from publicly available genome-wide association studies for six psychiatric disorders and individual-level data from the Adolescent Brain Cognitive Development (AB

OBJECTIVE: Although immunoglobulin A (IgA) is abundantly expressed in the gut and known to be an important component of mucosal barriers against luminal pathogens, its precise function remains unclear. Therefore, we tried to elucidate the effect of IgA on gut homeostasis maintenance and its mechanism.
DESIGN: We generated various IgA mutant mouse lines using the CRISPR/Cas9 genome editing system. Then, we evaluated the effect on the small intestinal homeostasis, pathology, intestinal microbiota, cytokine production, and immune cell activation using intravital imaging.
RESULTS: We obtained tw

Alzheimer disease (AD) was originally conceived as a rare disease that caused presenile dementia but has come to be understood as the most prevalent cause of dementia at any age worldwide. It has an extended preclinical phase characterized by sequential changes in imaging and cerebrospinal fluid biomarkers with subtle memory decline beginning more than a decade before the emergence of symptomatic memory loss heralding the beginning of the mild cognitive impairment stage. The apolipoprotein E ε4 allele is a prevalent and potent risk factor for AD that has facilitated research into its preclinic

Written and verbal languages are neurobehavioral traits vital to the development of communication skills. Unfortunately, disorders involving these traits-specifically reading disability (RD) and language impairment (LI)-are common and prevent affected individuals from developing adequate communication skills, leaving them at risk for adverse academic, socioeconomic and psychiatric outcomes. Both RD and LI are complex traits that frequently co-occur, leading us to hypothesize that these disorders share genetic etiologies. To test this, we performed a genome-wide association study on individuals
