Radioimmunoscintigraphy of intracranial glioma xenograft with a technetium-99m-labeled mouse monoclonal antibody specifically recognizing type III mutant epidermal growth factor receptor.

The type III mutant epidermal growth factor receptor (EGFR) is expressed on the cell surface of a subset of glioma, but not of normal tissues. In this study, we investigated the in vivo kinetics of 3C10 mouse monoclonal antibody (mAb), specifically recognizing the type III mutant EGFR (EGFRvIII), using athymic nude mice bearing the intracranial glioma xenograft overexpressing the EGFRvIII. Human glioma cell line, U87MG, expressing the wild type EGFR and the transfectant, named U87MG x deltaEGFR, expressing the EGFRvIII, were transplanted subcutaneously or intracranially to nude mice. 3C10 mAb

High-dose myeloablative radioimmunotherapy of mantle cell non-Hodgkin lymphoma with the iodine-131-labeled chimeric anti-CD20 antibody C2B8 and autologous stem cell support. Results of a pilot study.

Источник пометил публикацию как отозванную

BACKGROUND: CD20 has been used successfully as a target molecule for conventional low-dose, as well as high-dose, myeloablative radioimmunotherapy (RIT) of B-cell non-Hodgkin lymphoma (NHL). Mantle cell lymphoma (MCL) is an especially aggressive, prognostically unfavorable subtype of B-cell NHL, associated with an overall 5-year survival rate of less than 20%. Recent evidence has failed to show convincing therapeutic efficacy of conventional, nonmyeloablative RIT in patients with MCL. The aim of this pilot study was to investigate whether high-dose, myeloablative RIT with the iodine-131 ((131)

Trifunctional conjugation reagents. Reagents that contain a biotin and a radiometal chelation moiety for application to extracorporeal affinity adsorption of radiolabeled antibodies.

A method of removing radiolabeled monoclonal antibodies (mAbs) from blood using a device external to the body, termed extracorporeal affinity-adsorption (EAA), is being evaluated as a means of decreasing irradiation of noncancerous tissues in therapy protocols. The EAA device uses an avidin column to capture biotinylated-radiolabeled mAbs from circulated blood. In this investigation, three trifunctional reagents have been developed to minimize the potential deleterious effect on antigen binding brought about by the combination of radiolabeling and biotinylation of mAbs required in the EAA appr

Pertuzumab Enhances the Antitumor Activity of T-DXd in HER2-Positive Gastric Cancer Cells.

For HER2-positive advanced gastric cancer, a recent major therapeutic advancement is the development of trastuzumab deruxtecan (T-DXd), a HER2-directed antibody-drug conjugate. In this disease, simultaneously targeting HER2 and HER3 pathways has the potential to be a promising therapeutic strategy. However, the therapeutic approach of combining T-DXd with pertuzumab, which disrupts HER2-HER3 heterodimerization, has not yet been explored in gastric cancer, making this study a pioneering effort. In vitro, T-DXd efficacy correlated with high levels of membrane HER2 expression. Among the 12 cell l

Expression of the antibody-drug conjugate targets TROP2, CLDN6, B7-H3, and DLL3 in mesonephric-like adenocarcinoma of the female genital tract.

OBJECTIVE: Mesonephric-like adenocarcinoma (MLA) of the endometrium and ovary is a rare and aggressive malignancy with limited treatment options. Antibody-drug conjugates (ADCs) targeting cell-surface proteins such as TROP2, CLDN6, B7-H3, and DLL3 are emerging therapies in gynecologic cancers; however, their expression has never been systematically investigated in MLA. This study aimed to characterize the expression of these potential therapeutic targets in MLA. METHODS: Immunohistochemical staining for TROP2, CLDN6, B7-H3, and DLL3 was performed in a cohort of 22 MLAs (11 endometrial and 11

Mosunetuzumab plus polatuzumab vedotin for relapsed/refractory MCL after BTK inhibitor therapy: a phase 2 study.

Patients with relapsed/refractory (R/R) mantle cell lymphoma (MCL), especially those progressing after Bruton tyrosine kinase (BTK) inhibitor and/or chimeric antigen receptor (CAR) T-cell therapy and those with high-risk features, have poor outcomes. The bispecific antibody, mosunetuzumab, combined with the antibody-drug conjugate (ADC), polatuzumab vedotin (Mosun-Pola), targets CD20 and CD79b via independent cell-killing mechanisms. In this multicenter phase 2 study, patients with MCL who had received ≥2 previous lines of therapy, including a BTK inhibitor, were enrolled. Patients received ou

Legumain-Cleavable Top1i Antibody-Drug Conjugates without Self-Immolative Spacers Demonstrate Potent Antitumor Activity.

Virtually all antibody-drug conjugates employ a cleavable linker that is attached to a self-immolative spacer element. The linker and self-immolative spacer are widely known to have a dramatic influence on ADC stability, pharmacokinetics, and therapeutic efficacy. In 2021, our group described a highly polar legumain-cleavable linker that could be used to generate highly potent MMAE-based ADCs. Herein, we build on this finding by describing the design of legumain-cleavable ADCs that release a potent topoisomerase-I inhibitor (TOP1i) without the need for a self-immolative spacer. These ADCs empl

Folate receptor alpha as a successful biomarker in the treatment of low-grade serous ovarian cancer patients using preclinical and clinical models.

OBJECTIVE: Low-grade serous ovarian cancer is a rare epithelial ovarian cancer subtype characterized by high resistance to chemotherapy. Development of novel, effective, targeted treatments for recurrent low-grade serous ovarian cancer remains an unmet medical need. We evaluated FOLR1 expression in a cohort of low-grade serous ovarian cancer patients and the preclinical and clinical activity of mirvetuximab soravtansine, an antibody-drug conjugate targeting FOLR1, in vivo in a patient-derived xenograft model and in a heavily pretreated low-grade serous ovarian cancer patient progressing after

Expression and therapeutic potential of TROP2 in cisplatin-resistant germ cell tumors.

BACKGROUND AND OBJECTIVES: Cisplatin-based chemotherapy remains the standard of care for metastatic germ cell tumors (GCTs), but patients with cisplatin-refractory disease have limited effective options and poor outcomes. There is a clear unmet need for new therapies in this setting. Antibody–drug conjugates (ADCs) have emerged as a promising class of targeted agents for chemoresistant solid tumors. This study aimed to evaluate the expression of TROP2 in cisplatin-resistant GCT metastases [MET(-R)] and to assess the cytotoxic efficacy of the anti-TROP2 ADC sacituzumab govitecan (SG) in GCT cel

KK2845, a PBD dimer-containing antibody-drug conjugate targeting TIM-3-expressing AML.

Acute myeloid leukemia (AML) is a common hematopoietic malignancy with high recurrence rates, and there is an urgent need for new therapeutic agents. T-cell immunoglobulin mucin-3 (TIM-3) is expressed on the surface of both LSCs and blasts in most AML patients, but not in normal hematopoietic stem cells (HSCs). We have developed KK2845, an antibody drug conjugate (ADC) that consists of an anti-TIM-3 fully human IgG1 antibody, a valine-alanine linker and a highly potent DNA cross-linking pyrrolobenzodiazepine (PBD) dimer SG3199. KK2845 exhibited potent cytotoxicity against AML cells both in vit

An engineered antibody-lectin conjugate targeting the HIV glycan shield protects humanized mice against HIV challenge.

Enveloped viruses responsible for global health pandemics often display a glycan shield on their surface envelope glycoproteins. In HIV, the glycan shield is formed by clusters of high-mannose glycans and plays essential roles in viral fitness and immune evasion. A few mannose-binding lectins potently inactivate HIV but have not been fully exploited due to poor pharmacokinetics and short serum half-lives. To address this, we engineered an antibody-lectin conjugate comprising the anti-HIV lectin griffithsin (GRFT) to the Fc region of human IgG1, with the aim of extending its serum half-life and

Adverse events of antibody-drug conjugates: comparative analysis of agents with a common payload using the adverse event spontaneous reporting database.

BACKGROUND: Antibody-drug conjugates (ADCs) have been implicated in myelosuppression and peripheral neuropathy (PN), possibly owing to their payload. Thus, their systemic toxicity should be considered. However, reports documenting adverse event (AE) occurrences in clinical settings are limited. This study analyzed AEs associated with polatuzumab vedotin, brentuximab vedotin, and enfortumab vedotin-which share a common payload-based on patient background factors using Japanese and United States AE spontaneous reporting databases. Polatuzumab vedotin in CNS lymphoma: proof-of-concept study for blood-brain barrier penetration.

Polatuzumab vedotin (pola) is a CD79B-targeting antibody-drug conjugate with significant clinical activity in systemic diffuse large B-cell lymphoma. However, the ability of pola to penetrate the blood-brain barrier (BBB) and induce responses in central nervous system (CNS) lymphoma (CNSL) is unknown. Since other antibody-based therapies can partially penetrate the BBB to induce clinical responses in CNSL and other CNS malignancies, we hypothesized that pola would also partially penetrate the BBB, which could support its evaluation in future CNSL clinical trials. To test this hypothesis, we fi

Heterogeneity and Scoring Reproducibility of Folate Receptor 1 Immunohistochemistry in High-grade Serous Carcinoma.

Mirvetuximab soravtansine (MIRV) is an antibody-drug conjugate approved for the treatment of adult patients with folate receptor 1 (FRα; FOLR1) positive, platinum-resistant epithelial ovarian, fallopian tube, or primary peritoneal cancer, who have received one to three prior systemic treatment regimens. Per the FDA approval, FOLR1 positivity is defined as ≥75% of viable tumor cells showing moderate (2+) or strong (3+) membranous immunostaining ("PS2+"). Given this disease's high recurrence rate and relatively limited therapeutic options, there is utility in exploring consistency in FOLR1 repor
