Photodynamic therapy - hypericin tetraether liposome conjugates and their antitumor and antiangiogenic activity.

Photodynamic therapy (PDT) is an established noninvasive tumor treatment. The hydrophobic natural occurring pigment hypericin shows a lot of attractive properties for the application in PDT. Hence, the administration to biological systems or patients requires the formulation in drug carriers enabling sufficient bioavailability. Therefore, free hypericin was encapsulated by the thin film hydration method or a hypericin-hydroxypropyl-β-cyclodextrin inclusion complex (Hyp-HPβCD) was incorporated by dehydration-rehydration vesicle method in either conventional or ultra-stable tetraether lipid (TEL

Resolving liquid-to-glass transitions of water under soft nanoconfinement.

Water has challenged scientists with numerous anomalies and confounding problems since the very early times. The kinetic glass transition of bulk water is a pristine example, as its location remains mostly inaccessible to experimentalists in an area below about -41 °C, where bulk water spontaneously crystallises. Confining water within nanometre-sized domains is an intriguing approach inspired by the cellular environment in nature to completely prevent crystallisation. Here we probe the existence of liquid-to-glass transitions of water nanoconfined between non-freezing lipidic bilayers and fin

Therapeutic Applications and Advancements of Exosome-Based Treatments - A Comprehensive Review.

As tiny extracellular vesicles ranging 30-150 nm in size, exosomes are crucial for cell-to-cell communication and show great promise in therapeutic applications. All cell types release lipid bilayer-enclosed vesicles that carry bioactive molecules such as proteins, lipids, and nucleic acids, which reveal the health conditions of their originating cells. Exosome-based therapeutics have developed rapidly over the past decade due to their natural biocompatibility and minimal immune response, with the added benefit of traversing biological barriers such as the blood-brain barrier. This review pres

Extracellular vesicles for next-gen therapeutics and drug delivery.

Extracellular vesicles (EVs) are membrane-bound, nano-sized particles released by diverse cell types. They serve as key mediators of intercellular communication by transporting a broad repertoire of proteins, lipids, nucleic acids, and metabolites to recipient cells within a protective lipid bilayer. Owing to their natural origin, intrinsic stability, low immunogenicity, and ability to cross biological barriers, EVs are highly attractive candidates for next-generation therapeutics and drug delivery platforms. In this review, we summarize the biology of EVs and highlight the features critical f

Small extracellular vesicles as biomarkers and therapeutic targets in breast Cancer.

Small extracellular vesicles (sEVs) represent a novel frontier in oncology, functioning as critical mediators of intercellular communication that profoundly influence the biology of breast cancer. These nano-sized particles, typically ranging from 30 to 200 nm in diameter, are released by virtually all cell types into the extracellular space and circulate in biofluids like blood, urine, and saliva. Encased within a lipid bilayer, sEVs carry a complex molecular payload including proteins, lipids, various nucleic acids like microRNAs (miRNAs) and long non-coding RNAs (lncRNAs), and metabolites t

Biological properties and potential pathogenic mechanisms of oral bacterial-derived extracellular vesicles in oral and systemic diseases.

This review summarizes the biological characteristics of oral bacterial extracellular vesicles (O-BEVs) and their proposed roles in disease pathogenesis. As nanosized lipid bilayer structures secreted by oral bacteria, O-BEVs encapsulate virulence factors, nucleic acids, and metabolites, enabling them to traverse biological barriers and modulate host cellular functions. Studies suggest that O-BEVs are involved in the progression of oral diseases, including periodontitis, dental caries, and oral cancer through mechanisms such as immune activation, tissue destruction, and cellular behavior modul

An update on schistosomicide and tumoricide in the future: arachidonic acid.

Blood-dwelling schistosomes and cancer severely impact human health, welfare, and survival, essentially because of their ability to evade host immune cells and humoral effectors. The host immune defenses interact with the surface membrane of the worm and tumor cell. Schistosomes use their sphingomyelin (SM)-rich outer lipid bilayer to form with the surrounding water molecules a tight hydrogen bond barrier in which they open, at will, pores that allow entry of nutrients but not lethal complement components and antibodies. In parasite-free humans, impaired expression of one or more cellular gene

Recent Progress of Exosomes in Hematological Malignancies: Pathogenesis, Diagnosis, and Therapeutic Strategies.

Hematological malignancies originate from the hematopoietic system, including lymphoma, multiple myeloma, leukaemia, etc. They are highly malignant with a high incidence, a poor prognosis and a high mortality. Although the novel therapeutic strategies have partly improved the clinical efficacy of hematological malignancies, patients still face up with drug resistance, refractory disease and disease relapse. Many studies have shown that exosomes play an important role in hematological malignancies. Exosomes are nanoscale vesicles secreted by cells with a size ranging from 40 to 160 nm. They con

The role of exosome in autoimmune connective tissue disease.

Exosomes have generated significant interest in the last few decades owing to their important roles in a diverse range of biological pathways. They are nano-sized lipid bilayer membrane vesicles of endosomal origin, and are produced by a vast number of cell types. They are released into the extracellular environment and are found in most biological fluids. Exosomes can contain proteins, lipids and nucleic acids. The cargo of exosomes allows them to play roles in cell communication, antigen presentation, as biomarkers and in immune regulation. Substantial efforts have been made to understand th

Caffeine sensitivity of native RyR channels from normal and malignant hyperthermic pigs: effects of a DHPR II-III loop peptide.

Enhanced sensitivity to caffeine is part of the standard tests for susceptibility to malignant hyperthermia (MH) in humans and pigs. The caffeine sensitivity of skeletal muscle contraction and Ca(2+) release from the sarcoplasmic reticulum is enhanced, but surprisingly, the caffeine sensitivity of purified porcine ryanodine receptor Ca(2+)-release channels (RyRs) is not affected by the MH mutation (Arg(615)Cys). In contrast, we show here that native malignant hyperthermic pig RyRs (incorporated into lipid bilayers with RyR-associated lipids and proteins) were activated by caffeine at 100- to 1…

Light-Triggered Mechanotherapy of Membrane-Mimicking Conjugated Electrolytes Potentiates Antibacterial Efficacy.

The rise of multidrug-resistant (MDR) pathogens has created an urgent need for antibacterial strategies that operate beyond conventional biochemical targets. Here, a light-triggered molecular mechanotherapeutic antibacterial platform based on membrane-mimicking conjugated electrolytes (MMCEs) is reported. Owing to their membrane-compatible architecture, MMCEs can spontaneously intercalate into lipid bilayers while remaining relatively non-disruptive in the dark. To enhance photoinduced mechanical activity, ZBT was developed from the classical BT scaffold by replacing the stilbene-like wing wit…

Blocking glycosphingolipid production alters autophagy in osteoclasts and improves myeloma bone disease.

Glycosphingolipids (GSLs) are key constituents of membrane bilayers playing a role in structural integrity, cell signalling in microdomains, endosomes and lysosomes, and cell death pathways. Conversion of ceramide into GSLs is controlled by GCS (glucosylceramide synthase) and inhibitors of this enzyme for the treatment of lipid storage disorders and specific cancers. With a diverse range of functions attributed to GSLs, the ability of the GSC inhibitor, eliglustat, to reduce myeloma bone disease was investigated. In pre-clinical models of multiple myeloma, osteoclast-driven bone loss was reduc
