Nerve resuturation decreases autotomy in the rat.

Effects of different kinds of nerve lesions on denervation-induced self-mutilation (autotomy) was studied in the rat. Four different types of lesion were used: nerve transection, nerve transection combined with ligation and extirpation of the distal part of the transected nerve, nerve resuturation with a microsurgical technique, and lidocaine treatment after transection but before and during nerve resuturation. To determine the degree of nerve regeneration, the extensor muscles of the ankle joint were prepared and weighed after the follow-up period. If the transected nerves were resutured the

Distinct substance P and calcitonin gene-related peptide immunoreactive nerves in the guinea pig eye.

Using a double labeling indirect immunofluorescent technique, we studied the guinea pig trigeminal ganglion and eye for co-localization of substance P and calcitonin gene-related peptide. In the trigeminal ganglion, the number of neurons immunoreactive for calcitonin gene-related peptide significantly outnumber those immunoreactive for substance P, but virtually all substance P positive neurons are immunoreactive for calcitonin gene-related peptide. In the eye, a complex pattern of co-localization is present; both peptides co-localize in most immunoreactive nerve fibers. Nerve fibers immunorea

Myospasm and Fasciculation After Botulinum Toxin Injection.

Botulinum toxin type A (BTA) is commonly used to treat dystonia; however, reports of dystonia caused by BTA are rare. In this case study, the authors aimed to illustrate the instances of dystonia after cosmetic injections of BTA. Three cases were retrospectively analyzed, with follow-up evaluations conducted to assess the development and resolution of symptoms. Injection techniques, including microdroplet and intradermal methods, were examined, and potential hypotheses regarding the etiology of dystonia were explored. Recommendations for mitigating adverse effects were formulated based on the

Lymphatic dysfunction correlates with inflammation in a mouse model of amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a rapidly progressive, ultimately fatal neurodegenerative disease, without effective modifying treatments. It affects both lower and upper motor neurons, causing skeletal muscle denervation and paralysis. Regardless of the mechanisms that initiate and drive ALS, chronic neuroinflammation and systemic immune system activation play key roles in disease progression. The lymphatic system is a network of vessels and organs essential for immune surveillance, tissue fluid balance and lipid absorption, critical for the resolution and progression of inflammation i…

Tetramethylpyrazine nitrone improves motor dysfunction and pathological manifestations by activating the PGC-1α/Nrf2/HO-1 pathway in ALS mice.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive loss of upper and lower motor neurons that results in skeletal muscle atrophy, weakness and paralysis. Oxidative stress plays a key role in the pathogenesis of ALS, including familial forms of the disease arising from mutation of the gene coding for superoxide dismutase (SOD1). We have used the SOD1G93A ALS mouse model to investigate the efficacy of 2-[[(1,1-dimethylethyl)oxidoimino]-methyl]-3,5,6-trimethylpyrazine (TBN), a novel tetramethylpyrazine derivative armed with a powerful free-radica

Inhibition of Calpain Protects Against Tauopathy in Transgenic P301S Tau Mice.

Alzheimer's disease (AD) and other tauopathies are characterized by intracellular accumulation of microtubule-associated tau protein leading to neurodegeneration. Calpastatin is the endogenous inhibitor of calpain, a calcium-dependent cysteine protease that has been increasingly implicated in tauopathies. In this study, we generated a neuron specific calpastatin overexpressing knock-in transgenic mouse model and crossed it with the PS19 tauopathy mouse model expressing human P301S mutant tau protein. The forced expression of calpastatin in neurons significantly alleviated tau hyperphosphorylat

Changes in a rat facial muscle after facial nerve injury and repair.

This study describes changes in a rat facial muscle innervated by the mandibular and buccal facial nerve branches 4 months after nerve injury and repair. The following groups were studied: (A) normal controls; (B) spontaneous reinnervation by collateral or terminal sprouting; (C) reinnervation after surgical repair of the mandibular branch; and (D) chronic denervation. The normal muscle contained 1200 exclusively fast fibers, mainly myosin heavy chain (MyHC) IIB fibers. In group B, fiber number and fiber type proportions were normal. In group C, fiber number was subnormal. Diameters and propor

[Evaluation of autonomic nervous function by pupil dynamics recording].

Pupil is richly innervated by autonomic nerves. Therefore, its size or movement well reflects the autonomic nervous function. By analyzing dynamics of pupil movement for light stimulus followed by infrared video-pupillography, the abnormality of autonomic nervous system can be expressed in a quantitative manner. Clinical applications of pupillography and pharmacological examination to Horner syndrome, Adie syndrome, diabetes mellitus, pupillary involvement of Behçet disease, pupil of brain death and patients with chemical sensitivity have been described. Segmental spasm of iris in Horner and A

On the appearance of acetylcholine receptors in denervated rat diaphragm, and its dependence on nerve stump length.

Acetylcholine (ACh) sensitivity and extrajunctional receptor distribution of the rat diaphragm were closely monitored during the early period following denervation. Both contracture in response to 10 microgram/ml of ACh and extrajunctional binding of [125I]alpha-bungarotoxin ([125I]alpha-BTX)) were first detectable 30 h after cutting the phrenic nerve in the thorax. If the nerve were cut more proximally, leaving a 3.5 cm distal nerve stump, the same level of ACh contracture and [125I]alpha-BTX binding did not appear until 40 h after operation. This 10-h delay was far longer than the 3-h delay

EMG-based control for a C5/C6 spinal cord injury upper extremity neuroprosthesis.

The goal of this project is to enhance the benefits of functional electrical stimulation (FES) for individuals with cervical mid-level spinal cord injury (C5-C6 SCI) by providing upper arm function that complements the hand function provided by current FES systems. As a result of stimulation to selected shoulder and elbow muscles, individuals are able to increase their range of motion, their reaching ability, and improve their overall shoulder stability. An approach that provides a natural way of controlling arm stimulation is proposed. The controller extracts information from recorded EMG act

The fourth dorsal interosseus pedis muscle: a useful muscle in routine electromyography.

The aim of the study was to assess the utility of needle examination of the fourth dorsal interosseus pedis (FDIP) muscle in routine EMG. FDIP is an easily accessible muscle and may be less prone to blunt trauma as compared with the other foot muscles but despite its potential utility in electrophysiology, little if any data are available on the FDIP muscle. Detailed neurologic examination, nerve conduction, and needle EMG studies were performed in 118 individuals referred to the EMG laboratory of a tertiary referral center. The FDIP was studied by inserting a needle obliquely at an angle of a

miR-23a suppression accelerates functional decline in the rNLS8 mouse model of TDP-43 proteinopathy.

Skeletal muscle dysfunction may contribute to the progression and severity of amyotrophic lateral sclerosis (ALS). In the present study, we characterized the skeletal muscle pathophysiology in an inducible transgenic mouse model (rNLS8) that develops a TAR-DNA binding protein (TDP-43) proteinopathy and ALS-like neuropathology and disease progression; representative of >90% of all familial and sporadic ALS cases. As we previously observed elevated levels of miR-23a in skeletal muscle of patients with familial and sporadic ALS, we also investigated the effect of miR-23a suppression on skeletal m

Evaluation of the lower motor neuron integrity of upper extremity muscles in high level spinal cord injury.

PURPOSE: To evaluate the lower motor neuron (LMN) integrity of upper extremity muscles of persons with high tetraplegia (C1-C4) in order to determine muscles available for stimulation. METHODS: Fourteen subjects (23 arms) were evaluated for LMN integrity. Muscles that elicited a functional response (grade 3 or better) to surface electrical stimulation were considered to have intact LMN and good candidates for FES. Strength-duration (S-D) curves were generated on muscles that showed weak (less than grade 3) or no response to surface stimulation. Muscles were considered denervated if S-D curves

Acute cervical motor radiculopathy induced by neck and limb immobilization in a patient with Parkinson disease.

A 68-year-old woman with Parkinson disease (PD) presented with acute monoplegia of her left upper extremity after the neck and limb immobilization for several hours. Her sensory function was normal, and the chest X-ray showed left phrenic nerve palsy. Electrophysiological studies showed multi-segment muscle involvement (C3 to T1) including denervation potentials and reduced interference of motor units in needle electromyography. M wave amplitude in peripheral nerve stimulation was preserved except for the ulnar nerve, suggesting both axonal injury and conduction block at the anterior spinal ro

[Exercise-induced partial damage of the n. peroneus profundus].

Over a period of two years a 26-year-old man active in sports developed progressively more marked pain in the area of the left lateral tibial edge and signs of foot flexor paresis after physical exertion. These symptoms lasted for several hours. Finally there also developed sensory dysfunction in the area of the deep peroneal nerve. Electrophysiological tests revealed normal peroneal nerve conduction velocity but denervation signs in the anterior tibial muscle. In view of the diagnosis of an exercise-induced incomplete chronic anterior tibial syndrome a fasciotomy was performed revealing conne

[Comparative activity of ethaverine and papaverine on chronotropic and dromotropic cardiac functions in the anesthetized dog].

Ethaverine inhibits both atrioventricular conduction and heart rate, starting at a dosage of 1 mg/kg, injected i.v. into the anaesthetized dog. In the experimental conditions used, ethaverine is clearly different from papaverine, which stimulates both at a dosage between 1 and 10 mg/kg, and which only produces an inhibitory effect starting at a dosage of 20 mg/kg, with the appearance of cardiotoxic phenomena. After total cardiac denervation the inhibitory action of ethaverine persists partially, especially at the atrioventricular level, which is probably due to a direct effect on the heart mus

Morphological variability is greater at developing than mature mouse neuromuscular junctions.

The neuromuscular junction (NMJ) is the highly specialised peripheral synapse formed between lower motor neuron terminals and muscle fibres. Post-synaptic acetylcholine receptors (AChRs), which are found in high density in the muscle membrane, bind to acetylcholine released into the synaptic cleft of the NMJ, thereby enabling the conversion of motor action potentials to muscle contractions. NMJs have been studied for many years as a general model for synapse formation, development and function, and are known to be early sites of pathological changes in many neuromuscular diseases. However, inf
