Genetics and development of neural tube defects.

Congenital defects of neural tube closure (neural tube defects; NTDs) are among the commonest and most severe disorders of the fetus and newborn. Disturbance of any of the sequential events of embryonic neurulation produce NTDs, with the phenotype (eg anencephaly, spina bifida) varying depending on the region of neural tube that remains open. While mutation of > 200 genes is known to cause NTDs in mice, the pattern of occurrence in humans suggests a multifactorial polygenic or oligogenic aetiology. This emphasizes the importance of gene-gene and gene-environment interactions in the origins of

Enhancing neural communication via activating calcium-dependent signals toward spinal cord injury repair.

Spinal cord injury (SCI) is a severe neurological disease resulting in the formation of a harsh microenvironment that hinders neural regeneration. By preserving the components and structure of natural tissues, the decellularized extracellular matrix (dECM) mimics the neural regeneration microenvironment and provides the biochemical signals and structural support for neural regeneration. However, the regenerated neurons lack the ability to autonomously restore neural communication. In this study, we assemble the conductive polymer poly(3,4-ethylenedioxythiophene) (PEDOT) within spinal cord-deri

Single-cell transcriptomics identifies neural fate disruption and glial reprogramming caused by RARS2 deficiency.

BACKGROUND: Pontocerebellar hypoplasia type 6 (PCH6) is caused by biallelic pathogenic variants in RARS2, encoding mitochondrial arginyl-tRNA synthetase. Although mitochondrial dysfunction is a recognised feature, how RARS2 deficiency disrupts neural lineage development remains unclear. METHODS: We generated rars2-deficient zebrafish using the clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9 (CRISPR/Cas9) system and performed single-cell RNA sequencing (scRNA-seq) at 48 hours postfertilisation, complemented by immunofluorescence, in situ hybridisation, beh

The Notch pathway regulates autophagy after hypoxic-ischemic injury and affects synaptic plasticity.

Following neonatal hypoxic-ischemia (HI) injury, it is crucial factor to reconstruct neural circuit and maintain neural network homeostasis for neurological recovery. A dynamic balance between the synthesis and degradation of synaptic protein is required for maintaining synaptic plasticity. Protein degradation is facilitated by autophagy. This study aimed to investigate the regulation of synaptic structural plasticity by the Notch pathway, by assessing changes in Notch pathway activation and their effects on synaptic proteins and autophagy after HI injury. The study involved 48 male newborn Yo

Responses of neurons in rostral ventromedial medulla to nociceptive stimulation of craniofacial region and tail in rats.

The rostral ventromedial medulla (RVM) plays a key role in the endogenous modulation of nociceptive transmission in the central nervous system (CNS). The primary aim of this study was to examine whether the activities of RVM neurons were related to craniofacial nociceptive behaviour (jaw-motor response, JMR) as well as the tail-flick response (TF). The activities of RVM neurons and TF and JMR evoked by noxious heating of the tail or perioral skin were recorded simultaneously in lightly anaesthetized rats. Tail or perioral heating evoked the TF and JMR, and the latency of the JMR was significan

[Vertical vergence pathways].

Two pathways for vertical vergence probably exist: the cortical one carrying a psycho-optical reflex, the other sub-cortical connected to the vestibular apparatus and more probably to the otolith system. The sub-cortical pathway is essentially a vestibulo-cerebellar oculomotor pathway; but in addition there certainly exist direct vestibulo-oculomotor connections and long tracts passing through the interstitial nucleus of Cajal and the reticular formation.

Bypass neural interfaces for paralysis: clinical translation, challenges and future directions.

Advances in neural recording, real-time decoding and bioelectronic stimulation have enabled a new class of systems that re-establish functional communication across disrupted neural pathways. In the context of paralysis, these neural bypass interfaces link upstream neural intent to effector activation downstream of the lesion, effectively circumventing sites of injury within the nervous system to restore volitional movement. In this review, we define neural bypass interfaces as an emerging category of bioelectronic medicine, distinct from conventional brain-computer interfaces and neuromodulat…

Definition and connections of the entopallium in the zebra finch (Taeniopygia guttata).

In birds the entopallium (formerly known as the core region of ectostriatum) is the major thalamorecipient zone, within the telencephalon, of the tectofugal visual system. Here we sought to redefine the entopallium in the zebra finch, particularly with respect to a laterally adjacent zone, known as the perientopallium (formerly known as the periectostriatal belt), and to determine its projections. We show that the entopallium can be defined by the almost complete overlap of dense terminations of thalamic rotundal afferents and intense cytochrome oxidase activity and parvalbumin immunoreactivit

Magnetic graphene oxide functionalized composite nanofibrous stem cell-based neural scaffolds.

Conductive nerve scaffolds have emerged as a promising alternative to autologous grafts for promoting nerve regeneration. However, optimizing the electroactive properties of scaffold materials and elucidating their regulatory mechanisms on neural stem cell (NSC) differentiation remain critical challenges. Magnetic graphene oxide (MGO), an innovative nanomaterial integrating magnetic responsiveness with two-dimensional carbon-based conductivity, exhibits potential for modulating neural regeneration. Nevertheless, its application in guiding NSC fate within nanofibrous scaffolds is still limited.

Competing mechanisms of plasticity impair compensatory responses to repetitive apnoea.

KEY POINTS: Intermittent reductions in respiratory neural activity, a characteristic of many ventilatory disorders, leads to inadequate ventilation and arterial hypoxia. Both intermittent reductions in respiratory neural activity and intermittent hypoxia trigger compensatory enhancements in inspiratory output when experienced separately, forms of plasticity called inactivity-induced inspiratory motor facilitation (iMF) and long-term facilitation (LTF), respectively. Reductions in respiratory neural activity that lead to moderate, but not mild, arterial hypoxia occludes plasticity expression, i

Neurotoxic effects of 4-hydroxy-4'-isopropoxydiphenylsulfone exposure on zebrafish embryos.

The central nervous system (CNS) is particularly vulnerable to endocrine-disrupting chemicals, especially bisphenol analogues. Bisphenol A (BPA), a widely studied compound, has been associated with various neurological disorders, leading to restrictions on its use and the subsequent adoption of alternative chemicals such as 4-hydroxy-4'-isopropoxydiphenylsulfone (BPSIP). However, concerns regarding the potential neurotoxicity of BPSIP have emerged. To evaluate the effects of BPSIP on neural development, zebrafish were exposed to a gradient of concentrations (0.05 mg/L, 0.5 mg/L, 5 mg/L) for 96

Advances in research on dental occlusion and brain function: neuroimaging correlates and molecular mechanisms.

The relationship between dental occlusion and brain function is bidirectional, involving neuroanatomical, neurophysiological, and clinical disorders. Occlusal sensory input modulates sensorimotor control, cognitive functions, and emotional states via neural pathways, while aberrant brain function can conversely disrupt occlusion homeostasis. This review synthesizes recent progress in deciphering the bidirectional interactions between dental occlusion and brain function, with a focus on the impacts of occlusion on brain structure and function, the underpinning molecular and physiological mechan…

Anatomical Features of the Ventricular System Relevant to Endoscopic Surgery for Hydrocephalus.

In endoscopic surgery for hydrocephalus and associated intraventricular lesions, a thorough understanding of the required microsurgical anatomy is paramount. Endoscopic procedures in hydrocephalus treatment typically fall into two categories: ventriculocisternostomy and ventriculostomy for obstructive hydrocephalus. In the former, precise knowledge of intraventricular structures, such as the configuration of the ventricles and the path of internal vessels within them, is essential. In the latter, a comprehensive grasp of neural pathways, neural nuclei, and especially venous pathways beneath th…

Asthma, the central nervous system, and neurocognition: Current findings, potential mechanisms, and treatment implications.

Accumulating behavioral evidence suggests that asthma is associated with cognitive deficits. A number of studies have identified potential biological contributions to cognition in asthma; however, mechanistic pathways of central nervous system (CNS) involvement in asthma are yet to be established. We therefore conducted a literature review to identify studies examining potential CNS contributions to cognition in asthma. In this review, we discuss our general understanding of the CNS in asthma in the context of cognitive performance and outline a working model of mechanistic pathways linking th

[Dissemination pathways for poliovirus cells to animals models].

It is considered there are two main pathways for poliovirus dissemination towards the central nervous system in humans. One is the pathway through the blood brain barrier. The orally ingested virus invades into the blood circulation, and then the virus permeates into the central nervous system through the blood brain barrier. The other is the neural pathway. In this pathway, the intramuscularly-inoculated virus is transported through the axons from the synapse to the cell body in the central nervous system. We have developed the oral infection system using the mouse models. Moreover, we propos

Ischemic stroke and neurogenesis in the subventricular zone.

The subventricular zone (SVZ) of the lateral ventricle contains neural stem and progenitor cells that generate neuroblasts, which migrate to the olfactory bulb where they differentiate into interneurons. Ischemic stroke induces neurogenesis in the SVZ and these cells migrate to the boundary of the ischemic lesion. This article reviews current data on cytokinetics, signaling pathways and vascular niche that are involved in processes of proliferation, differentiation, and migration of neural progenitor cells after stroke.

Migratory potential of transplantable neural tumor cell lines.

Experimentally induced primitive neuroectodermal tumor (PNET) cell lines were transplanted into neonatal and adult rat brain and examined neuropathologically for their tumorigenic potential. Both cell lines showed a striking migratory behavior in both neonatal and adult brain. Migration of tumor cells was found in host brain parenchyma, along white matter tracts and associated with CSF pathways. These neural tumor cell lines provide a valuable tool for the development of strategies against strongly migrating neural tumors.

Electroacupuncture alleviates PTSD-like behaviors by modulating hippocampal synaptic plasticity via Wnt/β-catenin signaling pathway.

Abnormalities in hippocampal synaptic plasticity contribute to the pathogenesis of post-traumatic stress disorder (PTSD). The Wnt/β-catenin signaling pathway is critical for the regulation of synaptic plasticity. PTSD symptoms can be alleviated by correcting impaired neural plasticity in the hippocampus (Hipp). Electroacupuncture (EA) has a therapeutic effect by relieving PTSD-like behaviors. However, little is known about whether the Wnt/β-catenin pathway is involved in EA-mediated improvements of PTSD symptoms. In this study, we found that enhanced single prolonged stress (ESPS)-induced PTSD

Neuroimmune crosstalk in pancreatic ductal adenocarcinoma: integrating neuroscience and immuno-oncology.

Pancreatic ductal adenocarcinoma (PDAC) is among the most lethal malignancies and remains largely refractory to immunotherapy because of its profoundly immunosuppressive tumor microenvironment (TME). Emerging advances in cancer neuroscience suggest that immune resistance in PDAC may not be fully explained by immune-centered models alone, and increasingly implicate neural signaling as a potential context-dependent regulator of tumor progression and microenvironmental remodeling. In this review, we synthesize current evidence on the anatomical basis of pancreatic innervation, tumor-associated ne
