Rotenone induced olfactory deficit in Parkinson's disease rat model: The protective role of adenosine A2A receptors antagonist.

BACKGROUND: Parkinson's disease is both a motor and non-motor disorder. Despite the non-motor being an intrinsic feature of PD, it has been poorly researched and understood in clinical practices; olfactory deficit is one of the first established non motor symptom and nearly all ∼90 % of sporadic PD cases are associated with olfactory dysfunction and there is inconsistency in various pharmacological approaches. Hence this study aimed to evaluate the impact of caffeine at the A2A receptors of the olfactory bulb of a rotenone rat model of Parkinson's disease. MATERIALS AND METHODS: About 50 male…

The proboscis in human cyclopia: an anatomical study in two dimensions.

A 2-dimensional anatomical study has been undertaken of the proboscis and its contribution to the roof of the median orbit in human cyclopia … e of the ethmoidal notch forming a collection of tissue similar to the inferior layer of the normal olfactory bulb. The data indicate that the proboscis represents the anterosuperior part of the normal nasal cavity developed in the absence of median components. It is suggested that the cyclops face constitutes a model for the study of the development of the norm …

Behavioural disturbances and sensory pathology following allylnitrile exposure in rats.

Animals exposed to allylnitrile develop permanent abnormalities in motor behaviour, similar to those caused by 3,3'-iminodipropionitrile (ID … ir cell loss by scanning electron microscopy. Corneal transparency and concentrations in retina and olfactory bulbs of glial fibrillary acidic protein (GFAP), a marker for reactive gliosis, were also determined, as they are known targets of IDPN toxicity. In a dose-dependent manner, allylnitrile caused corneal opacity and gliosis in the retina and olfactory bulb …

Immunohistochemical and Ultrastructural characterization of the terminal nerve ganglion cells of the ayu, Plecoglossus altivelis (Salmoniformes, Teleostei).

BACKGROUND: Little is known on the cytological properties of the terminal nerve ganglion (TNG) cells in teleosts (Demski, 1993. Acta Anat., 148:81-95). MATERIALS AND METHODS: To characterize the TNG cells of a salmonoid fish, Plecoglossus altivelis, we adopted immunohistochemistry and transmission electron microscopy (TEM). RESULTS: The majority of the TNG cells formed a compact mass halfway between the olfactory sac and the olfactory bulb, whereas a few cells were scattered in the ventromedial region of the olfactory bulb. The cell had a voluminous perikaryon that was positive to antisera a…

Ultramicronized Palmitoylethanolamide and Luteolin Supplement Combined with Olfactory Training to Treat Post-COVID-19 Olfactory Impairment: A Multi-Center Double-Blinded Randomized Placebo- Controlled Clinical Trial.

BACKGROUND: Olfactory training is the only evidence-based treatment for post-viral olfactory dysfunction. Smell disorders after SARS-CoV-2 infection have been attributed to neuroinflammatory events within the olfactory bulb and the central nervous system. Therefore, targeting neuroinflammation is one potential strategy for promoting recovery from post-COVID-19 chronic olfactory dysfunction. Palmitoylethanolamide and luteolin (PEA-LUT) are candidate antiinflammatory/ neuroprotective agents. OBJECTIVE: To investigate recovery of olfactory function in patients treated with PEA-LUT oral supplemen…

Tumor necrosis factor alpha inhibits olfactory regeneration in a transgenic model of chronic rhinosinusitis-associated olfactory loss.

BACKGROUND: Olfactory loss is a debilitating symptom of chronic rhinosinusitis (CRS). Although olfactory sensory neurons (OSNs) are normally … , the role of the CRS-associated cytokine TNF-alpha was investigated using surgical ablation of the olfactory bulb to stimulate synchronous OSN turnover. We find that TNF-alpha expression prevents normal OE recovery, supporting the role of suppressed olfactory regeneration in the pathophysiology of CRS-associated olfactory loss.

Topographic coding of odorant quality is maintained at different concentrations in the salamander olfactory epithelium.

In a recent study in the tiger salamander, Ambystoma tigrinum, were demonstrated topographic patterns of responsivity across the olfactory epithelium which were characteristic for each odorant. The present study was initiated to investigate whether these patterns remain constant when odorant concentration is varied. Odorant-induced electro- olfactograms were recorded from at least 12 sites on each epithelium. The odorants used were pinene, amyl acetate and propanol. Each epithelium was tested with one odorant, delivered at 3 concentrations. For comparison between animals, the epithelia were di

Illuminating the terminal nerve: Uncovering the link between GnRH-1 neuron and olfactory development.

During embryonic development, the olfactory placode (OP) generates migratory neurons, including olfactory pioneer neurons, cells of the terminal nerve (TN), gonadotropin-releasing hormone-1 (GnRH-1) neurons, and other uncharacterized neurons. Pioneer neurons from the OP induce olfactory bulb (OB) morphogenesis. In mice, GnRH-1 neurons appear in the olfactory system around mid-gestation and migrate via the TN axons to different brain regions. The GnRH-1 neurons are crucial in controlling the hypothalamic-pituitary-gonadal axis. Kallmann syndrome is characterized by impaired olfactory system dev…

Ultrastructural evidence for multiple mucous domains in frog olfactory epithelium.

This study showed that the olfactory mucus is a highly structured extracellular matrix. Several olfactory epithelial glycoconjugates in the frog Rana pipiens were localized ultrastructurally using rapid-freeze, freeze-substitution and post-embedding (Lowicryl K11M) immunocytochemistry. Two of these conjugates were obtained from membrane preparations of olfactory cilia, the glycoproteins gp95 and olfactomedin. The other conjugates have a carbohydrate group which in the olfactory bulb appears to be mostly on neural cell-adhesion molecules (N-CAMs); in the olfactory epithelium this carbohydrate i…

Transplantation of neural stem cells in anosmic mice.

OBJECTIVES: Treating olfactory dysfunction is a challenge for physicians. One of the therapeutic options could be transplantation of stem cells. In this study, neural stem cells were transplanted into anosmic mice. METHODS: Neural stem cells were generated from the olfactory bulb of green fluorescent protein (GFP)-transgenic C57BL6 mice. Anosmia were induced by injection of intraperitoneal 3-methylindole. The neural stem cells were transplanted transnasally on the next day. The olfactory function was evaluated by a food-finding test once a week. The olfactory neuroepithelium was harvested for…

Neuronal nitric oxide synthase in the olfactory system, forebrain, pituitary and retina of the adult teleost Clarias batrachus.

Immunocytochemical application of antibodies against nNOS to the brain sections of Clarias batrachus revealed intense immunoreactivity in several olfactory receptor neurons (ORNs), in their axons over the olfactory nerve, and terminals in the olfactory glomeruli. Several basal cells in the olfactory epithelium showed NOS immunoreactivity. Application of post-embedding immunoelectron microscopy showed nNOS labeled gold particles in apical cilia, dendrites and soma of the ORNs and also in the axon terminals in the glomeruli of the olfactory bulb. nNOS containing fibers were also encountered in t…

The olfactory route for cerebrospinal fluid drainage into the peripheral lymphatic system.

Drainage of the cerebrospinal fluid through the olfactory nerves into the nasal lymphatics has been suggested repeatedly. To investigate precisely the morphology of this pathway, India ink was injected into the subarachnoidal space of the rat brain, and samples including the olfactory bulbs, olfactory tracts and the nasal mucosa were observed by light and electron microscopy. Under the dissecting microscope, ink particles were found within the subarachnoid space and along the olfactory nerves. At the nasal mucosa, a lymphatic network stained in black was identified near the olfactory nerves, w…

Tracing neural pathways in snail olfaction: from the tip of the tentacles to the brain and beyond.

The anatomical organization of the olfactory system of terrestrial snails and slugs is described in this paper, primarily on the basis of ex … s synaptic glomeruli whose ultrastructure is similar to that of the glomeruli seen in the mammalian olfactory bulb and the insect olfactory lobe. All of the olfactory neuropils, but especially the tentacle ganglion, contain large numbers of morphologically symmetrical chemical synapses. The procerebrum is a unique region of the snail brain that possesses further …

Coculture of Axotomized Rat Retinal Ganglion Neurons with Olfactory Ensheathing Glia, as an In Vitro Model of Adult Axonal Regeneration.

Olfactory ensheathing glia (OEG) cells are localized all the way from the olfactory mucosa to and into the olfactory nerve layer (ONL) of the olfactory bulb. Throughout adult life, they are key for axonal growing of newly generated olfactory neurons, from the lamina propria to the ONL. Due to their pro-regenerative properties, these cells have been used to foster axonal regeneration in spinal cord or optic nerve injury models. We present an in vitro model to assay and measure OEG neuroregenerative capacity after neural injury. In this model, reversibly immortalized human OEG (ihOEG) is culture…
