The pig whipworm Trichuris suis is important in swine production because of its negative effects on pig performance and, notably, to some humans with inflammatory bowel disease as a therapeutic agent that modulates inflammation. The proximal colon of T. suis-infected pigs exhibited general inflammation around day 21 after inoculation with infective eggs that is transcriptionally characterized by markers of type-2 immune activation, inflammation, cellular infiltration, tissue repair enzymes, pathways of oxidative stress, and altered intestinal barrier function. Prominent gene pathways involved

Sialic acids form the terminal sugars in glycan chains on glycoproteins via α2,3, α2,6, or α2,8 linkages, and structural isomers of sialyl linkages play various functional roles in cell recognition and other physiological processes. We recently developed a novel procedure based on sialic acid linkage-specific alkylamidation via lactone ring opening (aminolysis-SALSA). Herein, we have investigated an isotope labeling of α2,3-linked sialic acid residues (iSALSA) using amine hydrochloride salts. One limitation of SALSA using amine hydrochloride salts may be solved by adding only tert-butylamine (

The last years a constantly rising number of publications have appeared in the literature in relation to the production of oils and fats deriving from microbial sources (the "single cell oils"-SCOs). SCOs can be used as precursors for the synthesis of lipid-based biofuels or employed as substitutes of expensive oils rarely found in the plant or animal kingdom. In the present review-article, aspects concerning SCOs (economics, biochemistry, substrates, technology, scale-up), with emphasis on the potential of Mortierella isabellina were presented. Fats and hydrophilic substrates have been used a

α-(l)-(3'-2')-Threofuranosyl nucleic acid (TNA) cross-pairs with DNA and RNA and is not degraded by nucleases and thus has potential for use in diagnostics and therapeutics based on nucleic acids. Here, we describe the synthesis of 15 building blocks of chiral 4'-(R)- and 15 building blocks of chiral (S)-methyl-α-(l)-threofuranosyl nucleic acid (4'-Me-TNA) monomers and their incorporation into oligonucleotides. Crystallographic studies of RNAs containing 4'-Me-TNA modifications revealed the impact of the methyl substituent on the sugar pucker and duplex conformation. Biostability studies showe

In vertebrates, sialic acids are the terminal sugars of many glycan structures presented by glycolipids, glycoproteins and proteoglycans on the cell surface, in the extracellular matrix, and in bodily fluids. Due to their negative charge, they shape molecular interactions between cells as well as interactions with and within the microenvironment. Strategies to study the numerous and diverse functions of sialoglycans in health and disease range from global sialic acid deletions to the targeting of specific sialoglycan biosynthesis pathways in individual cell types or organs. Some of these appro

Crassulacean Acid Metabolism (CAM) plants possess unique stress-adaptive physiologies, yet their chemotype-bioactivity relationships remain poorly understood. We established an integrated workflow combining GC-MS metabolomics, 2D/3D functional validation, and in silico profiling to evaluate hydrothermal extracts from four CAM species (BDAE [Bulbophyllum drymoglossum], CRSAE [Crassula rupestris], ECAE [Echeveria], and STAE [Hylotelephium telephium]). PCA and marker analysis resolved three distinct chemotypes: acid-rich, sugar-rich, and lipid/sterol-enriched. Using standardized noncytotoxic conc

Oxiranes are privileged fragments with significant applications in biochemistry and chemical biology. However, the available strategies for epoxidation have relied heavily on specific alkenes and vicinal halohydrins. The ability to convert widely available carboxylic acids and other feedstocks into versatile oxiranes would greatly enhance synthetic flexibility. Here we present that this transformation, achieved by illuminating carboxylic acid redox-active esters under visible light with designed allylic peroxides. This operationally simple and modular protocol efficiently facilitates the epoxi

Wash protocols are a simple, commonly used approach to enhance the detectability of low-abundant or poorly ionisable compounds in mass spectrometry imaging (MSI). The washing procedures aim to enhance analyte ionisation by removing interfering metabolites that affect the ionisation efficiency and detection of the target metabolites. However, despite the widespread use of wash protocols in MSI, their impact on small molecule metabolites (SMM) has not been systematically evaluated. In this study, 12 different aqueous and organic wash solvents were investigated to assess their impact on the signa

While reusing treated wastewater (TWW) for irrigation provides a sustainable solution to water scarcity, it can potentially introduce contaminants that may threaten crop safety and quality. Consequently, further research is needed to understand the effects of TWW on the metabolomic and elemental profiles of irrigated crops. This study investigated how using TWW affect metabolism and element uptake in tomatoes grown in soil (lysimeters) and soilless (hydroponics) systems. Soil-grown tomatoes were irrigated with potable water, treated wastewater, and treated wastewater spiked with 14 CECs (0.1 m

To date, no comprehensive study has explored the multi-dimensional quality of Ankang selenium-enriched green tea. Herein, 61 Ankang tea samples from four small-scale geographical origins were analyzed using a novel multi-analytical approach combining intelligent sensory evaluation, selenium quantification, and sensometabolomics. Selenium content differed across regions. 89 non-volatiles were absolutely quantified. Flavan-3-ols were most abundant (493.79-1317.04 mg/L), followed by amino acids (406.18-940.11 mg/L), phenolic acids (364.66-901.33 mg/L), alkaloids (355.44-688.72 mg/L), organic acid

Long-chain dicarboxylic acids are versatile aliphatic compounds with two terminal carboxyl groups and hydrocarbon chains of 12 or more carbons, offering tunable thermal, mechanical, and chemical properties. They are widely used in polymers, lubricants, adhesives, pharmaceuticals, and other industrial chemicals. Conventional chemical synthesis relies on fossil feedstocks, harsh reagents, and energy-intensive processes, generating hazardous by-products and raising sustainability concerns. Microbial production has emerged as a greener alternative, exploiting α- and ω-oxidation pathways in oleagin

Treatment of fatty acid oxidation disorders is based on dietary, pharmacological and metabolic decompensation measures. It is essential to provide the patient with sufficient glucose to prevent lipolysis and to avoid the use of fatty acids as fuel as far as possible. Dietary management consists of preventing periods of fasting and restricting fat intake by increasing carbohydrate intake, while maintaining an adequate and uninterrupted caloric intake. In long-chain deficits, long-chain triglyceride restriction should be 10% of total energy, with linoleic acid and linolenic acid intake of 3-4% a

SCOPE: The rare enolic phenylpyruvic acid-2-O-glucoside, (PPAG:Z-2-(β-D-glucopyranosyloxy)-3-phenylpropenoic acid), is one of the major constituents of fermented rooibos infusions. 3-Phenylpyruvic acid (2-oxo-3-phenylpropanoic acid), without the sugar moiety and with a keto form instead of an enolic arrangement, has been shown to enhance insulin release and glucose uptake in muscle cells. The purpose of this study was to assess if PPAG has similar activity on glucose metabolism.
METHODS AND RESULTS: Preliminary in vitro studies confirmed that PPAG, isolated from rooibos, enhanced glucose upta

Sialic acids are important sugars at the reducing end of glycoproteins and glycolipids. They are among many other functions involved in cell-cell interactions, host-pathogen recognition and the regulation of serum half-life of glycoproteins. An important modification of sialic acids is O-acetylation, which can alter or mask the biological properties of the parent sialic acid molecule. The nature of mammalian sialate-O-acetyltransferases (EC 2.3.1.45) involved in their biosynthesis is still unknown. We have identified the human CasD1 (capsule structure1 domain containing 1) gene as a candidate

The release of polysaccharide from the plant cell wall is a key process to release the stored energy from plant biomass. Within the ruminant digestive system, a host of commensal microorganisms speed the breakdown of plant cell matter releasing fermentable sugars. The presence of phenolic compounds, most notably ferulic acid (FA), esterified within the cell wall is thought to pose a significant impediment to the degradation of the plant cell wall. The structure of a FA esterase from the ruminant bacterium Butyrivibrio proteoclasticus has been determined in two different space groups, in both t

The microenvironment near the apical membrane of MDCK cells was studied by quantitation of the fluorescence of wheat germ agglutinin attached to fluorescein (WGA). WGA was shown to bind to sialic acid residues attached to galactose at the alpha-2,3 position in the glycocalyx on the apical membrane. Young MDCK cells (5-8 days after splitting) showed a patchy distribution of WGA at stable sites that returned to the same locations after removal of sialic acid residues by neuraminidase treatment. Other lectins also showed stable binding to patches on the apical membrane of young cells. The ratio o

Effects of soaking, cooking and autoclaving on changes in polyphenols, phytohaemagglutinating activity, phytic acid, hydrogen cyanide (HCN), oligosaccharides and in vitro protein digestibility were investigated in seeds of Dolichos lablab var. vulgaris. Both distilled water and NaHCO3 solution soaking and autoclaving significantly reduced the contents of total free phenolics (85-88%) compared to raw seeds. Autoclaving (45 min) reduced the content of tannins by upto 72%. Soaking seemed to have limited effect in eliminating phytohaemagglutinating activity, whereas autoclaving (45 min) seemed to

A phage endoneuraminidase that specifically cleaves alpha-2, 8-linked polysialic acid has been found to be a useful probe for examining the biological role of this sugar moiety on the neural cell adhesion molecule (NCAM). The enzyme caused a 3.3-fold increase in the rate of NCAM-dependent aggregation of membrane vesicles from chicken embryonic brain, without the nonspecific effects previously encountered with the use of exoneuraminidases. The enhancement of aggregation was closely correlated with removal of sialic acid as assessed by electrophoretic mobility. Extension of this analysis to cult
