Engineering MRI-Based Programmable Genetic Sensors Using the MAPPER Platform.

Genetically encodable reporters that produce signals detectable in deep tissues offer a powerful tool for noninvasive monitoring of molecular events in vivo. Although magnetic resonance imaging (MRI) is a standard technique for noninvasive clinical imaging, its wider application in detecting molecular activities has been constrained by the lack of programmable sensors. This limitation is in stark contrast to the widespread use of fluorescent reporter-derived sensors in cultured cells and in transparent specimens. To overcome this limitation, we recently developed the modular aquaporin-based pr

Non-insulin dependent diabetes--the past, present and future.

Diabetes, known since antiquity, has been defined by glycosuria. In 1886, when Minkowski demonstrated that pancreatectomized dogs developed diabetes, the islets of Langerhans became a focus of the search for an active principle culminating in the discovery and the isolation of insulin in 1921 by Banting, Best and Collip. In 1959, the radioimmunoassay of Yalow and Berson solidified the concept of insulin resistance in non-insulin dependent diabetes (NIDDM). In 1971, the insulin receptor was defined as a cell surface protein that initiated the insulin signal transduction cascade. Today, we know

Messing with Signal 1: How Perturbed MHC Class I Antigen Presentation Contributes to Cancer.

The antigen presentation machinery processes proteins for presentation to T cells, thereby controlling activation of the adaptive cellular immune response. Perturbation of this machinery has been linked to the development of various diseases. This review describes the function of the Major Histocompatibility Complex class I antigen presentation machinery and highlights how its perturbation can lead to compromised immune function and disease progression in the context of cancer. We categorize these perturbations into four distinct mechanistic levels: peptide generation, peptide loading, MHC cla

Human β-defensin-3 as a transcriptional convergence point linking innate immunity, endocrine signals, and tissue repair.

Human β-defensin 3 (hBD-3) is a multifunctional host defense peptide with exceptional antimicrobial, immunomodulatory, and tissue-repair properties. Beyond its potent activity against gram-positive and gram-negative bacteria, fungi, and selected viruses, hBD-3 plays a central role in epithelial barrier integrity, chemotactic signaling, and wound healing. Its expression is tightly regulated by an intricate network of pattern-recognition receptors, inflammatory cytokines, growth factors, vitamins, and endocrine mediators. Emerging evidence indicates that hBD-3 functions as a transcriptional conv

AKAP6β-a conductor in the orchestra of pathological cardiomyocyte gene transcription.

Heart failure is a major cause of morbidity and mortality worldwide, representing the end stage of cardiovascular diseases that induce pathological cardiac remodeling. At the cellular level, pathological remodeling includes myocyte hypertrophy and decreased survival, changes in myocyte metabolism and contractility, and interstitial myocardial fibrosis, all of which are controlled by a complex network of intracellular signaling pathways. At the outer nuclear envelope of the cardiomyocyte, A-kinase anchoring protein (AKAP) 6β (also known as mAKAPβ) organizes multimolecular signaling complexes ca

Functional diversity of natural human IFNB1 variants in innate immunity.

Human interferon-β (IFN-β), a type I IFN, is critical for effective innate immunity and is also an approved disease-modifying therapeutic. While genetic variations in several components of the type I IFN system are associated with infectious and immune diseases, consequences of IFNB1 variation remain unexplored. Here, we functionally evaluated 70 naturally occurring rare protein-altering IFNB1 variants detected in humans globally. Twenty-five percent of all protein variants were found to be substantially less active than the common reference IFN-β, a phenotype that for many correlated with ret

Beyond Single Variants: A Pathway-Based Approach to Explore the Genetic Basis of Memory.

Memory plays a crucial role in human cognitive processes and daily functioning. While memory has a genetic basis, identifying the specific genetic factors influencing memory performance has proven challenging. This challenge arises because memory is a complex trait, whose genetic architecture likely comprises an accumulation of many low-effect size common variants. Thus, study sample sizes are easily insufficient, leading to underpowered genetic association analyses. This limitation is especially pronounced in studies that prioritize deep phenotyping over broader recruitment, resulting in smal

From Tumor Biology to Clinical Perspectives: Novel Biomarkers and Therapeutic Insights in Gastric Cancer.

Gastric cancer (GC) is a leading cause of cancer-related mortality worldwide. Accurate early detection, timely diagnostic stratification, and robust prognostic risk assessment are essential for optimizing clinical outcomes and improving survival duration. However, conventional serological biomarkers demonstrate limited diagnostic performance owing to suboptimal sensitivity and specificity, while standard chemotherapy and targeted therapies provide only modest survival benefits in GC. Marked inter- and intratumoral heterogeneity further characterizes GC as a biologically complex and treatment-r

How epigenetics and miRNA affect gene expression in dental pulp inflammation: A narrative review.

BACKGROUND: Pulpitis, an inflammatory condition of the dental pulp, typically arises due to caries. It can remain asymptomatic for extended periods, complicating its diagnosis. The inflammatory response induced by bacterial invasion encompasses both cell-mediated and humoural immunity, accompanied by neural and vascular changes. The primary aim of inflammation is to eradicate invading pathogens from the pulp. However, failure to eliminate pathogens may result in necrosis of the pulp. Before direct bacterial contact with cells occurs, the pulp initiates protective responses like the formation o

Shared and Context-Specific Mechanisms of EMT and Cellular Plasticity in Cancer and Fibrotic Diseases.

Cellular plasticity enables cells to dynamically adapt their phenotype in response to environmental cues, a process central to development, tissue repair, and disease. Among the most studied plasticity programs is epithelial-mesenchymal transition (EMT), a transcriptionally controlled process by which epithelial cells acquire mesenchymal traits. Originally described in embryogenesis, EMT is now recognized as a key driver in both tumor progression and fibrotic remodeling. In cancer, EMT and hybrid epithelial/mesenchymal (E/M) states promote invasion, metastasis, stemness, therapy resistance, an

Hedgehog signaling pathway: A research review on a new therapeutic target for rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by progressive joint destruction, with existing therapies limited by adverse effects and incomplete efficacy. The Hedgehog signaling pathway, abnormally activated in RA, plays a pivotal pathogenic role by promoting synovial fibroblast proliferation/invasion, amplifying inflammatory responses, inducing chondrocyte matrix degradation, and enhancing angiogenesis. This review summarizes therapeutic strategies targeting this pathway, including small-molecule inhibitors (Smo/Gli antagonists), gene therapy (CRISPR-Cas, SMO-siRNA)

Phosphoinositide signaling in the nucleus: Impacts on chromatin and transcription regulation.

Phosphoinositides also called Polyphosphoinositides (PPIns) are small lipid messengers with established key roles in organelle trafficking and cell signaling in response to physiological and environmental inputs. Besides their well-described functions in the cytoplasm, accumulating evidences pointed to PPIns involvement in transcription and chromatin regulation. Through the description of previous and recent advances of PPIns implication in transcription, this review highlights key discoveries on how PPIns modulate nuclear factors activity and might impact chromatin to modify gene expression.

miRNAs dysregulation in ankylosing spondylitis: A review of implications for disease mechanisms, and diagnostic markers.

Epigenetic processes, including non-coding RNA, histone modifications, and DNA methylation, play a vital role in connecting the environment to the development of a disorder, especially when there is a favorable genetic background. Ankylosing Spondylitis (AS) is a chronic type of spinal arthritis that highlights the significance of epigenetics in diseases related to autoimmunity and inflammation. MicroRNAs (miRNAs) are small non-coding RNAs that are involved in both normal and aberrant pathological and physiological gene expression. This study focuses on the pathophysiological pathways to clari

Engineered AAV13 variants with enhanced transduction and confined spread.

Precise targeting of specific regions within the central nervous system (CNS) is crucial for both scientific research and gene therapy in the context of brain diseases. Adeno-associated virus 13 (AAV13) is known for its restricted diffusion range within the CNS, making it an ideal choice for precise labeling and administration within small brain regions. However, AAV13 mediates relatively low expression of target genes. Here, we introduced specifically engineered modifications to the AAV13 capsid protein to enhance its transduction efficiency. We first constructed AAV13-YF by mutating tyrosine

Genome-wide association study across five cohorts identifies five novel loci associated with idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a chronic lung condition with poor survival times. We previously published a genome-wide meta-analysis of IPF risk across three studies with independent replication of associated variants in two additional studies. To maximise power and to generate more accurate effect size estimates, we performed a genome-wide meta-analysis across all five studies included in the previous IPF risk genome-wide association studies. We used the distribution of effect sizes across the five studies to assess the replicability of the results and identified five robust novel ge

Impact of Endocannabinoid System Manipulation on Neurodevelopmental Processes Relevant to Schizophrenia.

The neurodevelopmental hypothesis of schizophrenia has received much support from epidemiological and neuropathological studies and provides a framework to explain how early developmental abnormalities might manifest as psychosis in early adulthood. According to this theory, the onset of schizophrenia is likely the result of a complex interplay between a genetic predisposition and environmental factors whose respective influence might contribute to the etiology and progression of the disorder. The two most sensitive windows for neurodevelopment are the prenatal/perinatal and the adolescent win

FNDC5: A novel player in metabolism and metabolic syndrome.

Half a decade ago, transmembrane protein fibronectin type III domain-containing protein 5 (FNDC5) was found to be cleaved as a novel myokine irisin, which burst into prominence for browning of white adipose tissue during exercise. However, FNDC5, the precursor of irisin, has been paid relatively little attention compared with irisin despite evidence that FNDC5 is associated with the metabolic syndrome, which accounts for one-fourth of the world's adult population and contributes to diabetes, cardiovascular disease and all-cause mortality. Besides N-terminal and C-terminal sequences, the FNDC5
