Chronic cocaine but not chronic amphetamine use is associated with perseverative responding in humans.

RATIONALE: Chronic drug use has been associated with increased impulsivity and maladaptive behaviour, but the underlying mechanisms of this impairment remain unclear. We investigated the ability to adapt behaviour according to changes in reward contingencies, using a probabilistic reversal-learning task, in chronic drug users and controls. MATERIALS AND METHODS: Five groups were compared: chronic amphetamine users (n = 30); chronic cocaine users (n = 27); chronic opiate users (n = 42); former drug users of psychostimulants and opiates (n = 26); and healthy non-drug-taking control volunteers (

Identifying and understanding mechanisms in an inhibitory control training in patients with binge-eating-disorder - an exploratory analysis.

Cognitive processes play a central role in the development, maintenance, and remission of mental disorders, like Binge Eating Disorder (BED). Cognitive trainings aim to alter those cognitive processes to achieve amelioration of psychopathology. Regardless, little is known about the underlying mechanisms that can explain effective treatment in BED. Therefore, we analyzed findings from a clinical randomized-controlled trial (RCT) (Giel, Schag et al., 2023) which used an individualized computer-based inhibitory control training programme enhanced by transcranial direct current stimulation (tDCS).

Percutaneous CT-Guided Cryovagotomy.

There are a number of pathologic conditions in the human body that may be modified by the interruption of neural signaling, both related to pain and otherwise. Many of these treatments currently involve implantable neuromodulation or frank surgical neural ligation, representing opportunities for the implementation of percutaneous device-mediated cryoneurolysis in interventional radiology. Computed tomography-guided cryovagotomy for the management of mild to moderate obesity represents one such opportunity currently under investigation. This procedure is designed to attenuate hunger signals by

The Association of Inflammation with Food Intake in Older Hospitalized Patients.

OBJECTIVES: An acute inflammatory process may play a role in inhibiting appetite and food intake particularly in acutely ill older individuals. However, there is a lack of knowledge on the effect of inflammation on food intake in humans. In this study, we sought to investigate the association of C-reactive protein (CRP), as an inflammatory marker, with food intake in acutely ill older hospitalized patients. DESIGN AND SETTING: This cross-sectional study investigated older participants who were consecutively admitted to a geriatric acute care ward. Food intake during previous week was measured

Endocannabinoids, feeding and suckling--from our perspective.

In this overview we have summarized some aspects of our published work related to the effects of the endocannabinoid system on appetite and suckling. As noted also by several other groups we have found that anandamide, a major endocannabinoid, enhances appetite in mice. On partial or full food deprivation over 24 h the levels of 2-arachidonoyl glycerol (2-AG), a second major cannabinoid, are initially elevated in mouse brain; however, partial food deprivation over a longer period causes reduction of 2-AG levels. Blocking the endocannabinoid system with a CB1 antagonist on the 1st day after bir

A placebo controlled randomized trial of the effects of phenylpropanolamine and nicotine gum on cessation rates and postcessation weight gain in women.

With smoking prevalence rates beginning to decline, studies designed to promote cessation in more challenging populations, like weight-concerned smokers, warrant attention. This study assessed the efficacy of two forms of pharmacotherapy [nicotine and phenylpropanolamine (PPA) gums] in addition to a 13-week cognitive behavioral smoking cessation program targeted for women. Participants were 439 females who met rigorous screening criteria and were randomized to one of the three treatment intervention groups (PPA gum, nicotine gum, or placebo gum). All participants attended a 13-week cognitive b

Infusion of the amylin antagonist AC 187 into the area postrema increases food intake in rats.

According to previous studies, the area postrema (AP) of the hindbrain may play an important role in mediating the anorectic effect of the pancreatic hormone amylin. Peripheral amylin has been suggested to directly act on AP neurons to bring about its anorectic effect. Cyclic GMP may act as second messenger in this regard. In the present study, we wanted to further delineate the role of the AP in amylin's effect and to find out whether endogenous amylin might reduce feeding via the AP. Rats with chronic cannulas aiming at the AP were infused with various doses of amylin, its agonist salmon cal

Acute 3rd-ventricular amylin infusion potently reduces food intake but does not produce aversive consequences.

In this study, a conditioned taste aversion (CTA) paradigm was used to assess the possibility that 3rd-ventricular (i3vt) administration of the pancreatic hormone amylin produces aversive consequences that secondarily reduce food intake independently of the normal regulation of energy balance. After 1-h daily access to water for 7 days, rats were given 1-h access to a 0.15% saccharin solution, followed immediately by i3vt amylin (100 pmol) in one group (n=7) and i3vt CSF vehicle in another (n=7). As positive control for the formation of a CTA, a third group of seven rats received intraperitoni

Elemental and configural visual discrimination learning following lesions to perirhinal cortex in the rat.

Rats were tested in a series of two-choice visual discrimination tasks in a computer-controlled testing apparatus. The discriminations used a range of discriminanda, which varied in complexity. The discriminations included relatively simple form discriminations, more complex form discriminations and discriminations between compound stimuli that shared many features. It was found that rats with perirhinal cortex lesions were unimpaired in all discriminations except those that involved the compound stimuli with overlapping features. Using these stimuli, rats with perirhinal cortex lesions were u

Psychological stress impairs spatial working memory: relevance to electrophysiological studies of hippocampal function.

Stress blocks hippocampal primed-burst potentiation, a low threshold form of long-term potentiation, thereby suggesting that stress should also impair hippocampal-dependent memory. Therefore, the effects of stress on working (hippocampal-dependent) and reference (hippocampal-independent) memory were evaluated. Rats foraged for food in seven arms of a 14-arm radial maze. After they ate the food in four of the seven baited arms, they were placed in an unfamiliar environment (stress) for a 4-hr delay. At the end of the delay they were returned to the maze to locate the food in the 3 remaining bai

Corticosterone facilitation of inhibition of fat intake by enterostatin (Val-Pro-Asp-Pro-Arg).

Enterostatin or Val-Pro-Asp-Pro-Arg (VPDPR) is the amino-terminal pentapeptide of procolipase; VPDPR is generated during tryptic activation of procolipase to lipase. In rodents, exogenous VPDPR has been shown to cause a selective decrement in fat appetite. To understand the mechanism(s) underlying the action of this peptide, we have studied the effects of corticosterone, an adrenal hormone known to modulate caloric intake, on VPDPR-mediated inhibition of appetite. The results of this study show a significant increase in the inhibition of total caloric intake by 250 micrograms/kg VPDPR followin

Agonist-induced homologous and heterologous sensitization to D-1- and D-2-dependent contraversive turning.

A low dose of the D-1 receptor agonist SKF 38393 (2.0 mg/kg s.c.) 14 days post-lesion failed to induce contraversive turning in male Sprague-Dawley rats lesioned unilaterally in the medial forebrain bundle with 6-hydroxydopamine (6OHDA). Priming with a single administration of the specific D-2 agonist LY 171555 (0.2 mg/kg s.c. 3 days before), which itself elicited contraversive turning, made SKF 38393 (2.0 mg/kg s.c.) very active to produce contraversive turning. Priming with LY 171555 (0.2 mg/kg s.c. 3 days before) potentiated the contraversive turning in response to a low dose of LY 171555 (

Feeding suppression induced by a fecal anorexigenic substance (FS-T).

Intraperitoneal (IP) injection of a fecal anorexigenic substance (FS-T) induced significant suppression of feeding and this suppression recovered on the second day. At 2 hr after IP injection, at the time of maximum feeding suppression, plasma glucose, insulin and free fatty acid (FFA) levels did not change but amino acid level decreased. Intra-third cerebral ventricle (ICV) infusion of FS-T induced parallel but more potent feeding suppression. Analysis of meal patterns demonstrated that suppression of feeding after ICV treatment continued into the second day. FS-T was applied electrophoretica

Study of symptoms in middle life with special reference to the menopause.

In an attempt to clarify the nature of the "menopausal syndrome" a survey of symptoms was carried out by means of postal questionnaires in a population sample of 1120 women and 510 men. Special care was taken to avoid letting the participants know that the survey was mainly concerned with the menopause. Response rates were 72% for women and 68% for men. Analysis of patterns of symptoms by age and sex showed that peaks of prevalence of flushing and sweating were closely associated with the mean age of menopause, coinciding with it or occurring a little after it. Less impressive peaks of prevale

Anorexigenic effects of d-amphetamine and l-DOPA in the rat.

The effect of amphetamine and l-dopa was compared in 22-hr food- and water-deprived rats. Amphetamine produced marked anorexia, and l-dopa significantly reduced food intake at 200 mg/kg. Following pretreatment with RO 4-4602, a decarboxylase inhibitor, 100 mg/kg of l-dopa, a dose that did not significantly affect eating, produced marked anorexia. The anorectic effect of both amphetamine and l-dopa was antagonized by propranolol, a beta adrenergic antagonist. Phentolamine, an alpha-adrenergic antagonist, potentiated the anorectic effect of amphetamine and l-dopa. Haloperidol (0.1 mg/kg), a dopa

Dextroamphetamine Treatment in Children With Hypothalamic Obesity.

INTRODUCTION: Hypothalamic obesity (HO) in children has severe health consequences. Lifestyle interventions are mostly insufficient and currently no drug treatment is approved for children with HO. Amphetamines are known for their stimulant side-effect on resting energy expenditure (REE) and suppressing of appetite. Earlier case series have shown positive effects of amphetamines on weight in children with acquired HO. We present our experiences with dextroamphetamine treatment in the, up to now, largest cohort of children with HO. METHODS: A retrospective cohort evaluation was performed of ch

Exercise Timing and its Effects on Eating Behavior, Weight, and Body Composition: A Systematic Scoping Review to Identify Gaps and Inform Future Research.

PURPOSE: This scoping review aims to: (1) describe the existing evidence and, based on that, examine study designs, intervention strategies, participant characteristics (e.g., age, sex), and exercise protocols (e.g., modality, intensity, duration); (2) Identify knowledge gaps related to the effects of exercise timing, and provide recommendations for future research directions. RESULTS: Fragmented evidence of time-of-day effects was identified across acute studies, particularly for subjective appetite ratings, food preference, energy intake, and relative energy intake. In chronic studies, isola

Endometriosis and eating disorders: epidemiology, shared neurobiology, and clinical implications.

Growing evidence suggests that women with endometriosis may be particularly vulnerable to disordered eating behaviors (DEBs) and clinically defined eating disorders (EDs). This narrative review aims at integrating and critically analyzing the current evidence regarding the relationship between endometriosis and EDs, as well as highlighting the psychosocial and neurobiological vulnerabilities of women with endometriosis to DEBs. A large-scale genetic study showed a nearly threefold increase in the odds of EDs in women with endometriosis, and a significant genetic correlation. Although the preva
