The Clinical and Genetic Characteristics of Streptococcus agalactiae Meningitis in Neonates.

Streptococcus agalactiae (Group B Streptococcus, GBS) is an important pathogen of bacterial meningitis in neonates. We aimed to investigate the clinical and genetic characteristics of neonatal GBS meningitis. All neonates with GBS meningitis at a tertiary level medical center in Taiwan between 2003 and 2020 were analyzed. Capsule serotyping, multilocus sequence typing, antimicrobial resistance, and whole-genome sequencing (WGS) were performed on the GBS isolates. We identified 48 neonates with GBS meningitis and 140 neonates with GBS sepsis. Neonates with GBS meningitis had significantly more

Interleukin-13-Overexpressing Mice Represent an Advanced Preclinical Model for Detecting the Distribution of Antimycobacterial Drugs within Centrally Necrotizing Granulomas.

The Mycobacterium tuberculosis-harboring granuloma with a necrotic center surrounded by a fibrous capsule is the hallmark of tuberculosis (TB). For a successful treatment, antibiotics need to penetrate these complex structures to reach their bacterial targets. Hence, animal models reflecting the pulmonary pathology of TB patients are of particular importance to improve the preclinical validation of novel drug candidates. M. tuberculosis-infected interleukin-13-overexpressing (IL-13tg) mice develop a TB pathology very similar to patients and, in contrast to other mouse models, also share pathog

A Prospective Evaluation of Ileocecal Valve Dysfunction and Intestinal Motility Derangements in Small Intestinal Bacterial Overgrowth.

BACKGROUND: Small intestinal bacterial overgrowth (SIBO) is an increasingly recognized clinical syndrome; however, its etiophathogenesis is poorly understood. We hypothesized that loss of gastric acid, a delayed intestinal transit, and ileocecal valve dysfunction may contribute to the genesis of this syndrome. AIMS: Our primary aim was to assess these parameters using wireless motility capsule (WMC) testing and to correlate them with the presence or absence of SIBO. METHODS: We prospectively evaluated 30 consecutive patients at a tertiary care center with suspected SIBO, diagnosed by lactulo

The effect of DNA extraction methodology on gut microbiota research applications.

BACKGROUND: The effect that traditional and modern DNA extraction methods have on applications to study the role of gut microbiota in health and disease is a topic of current interest. Genomic DNA was extracted from three faecal samples and one probiotic capsule using three popular methods; chaotropic (CHAO) method, phenol/chloroform (PHEC) extraction, proprietary kit (QIAG). The performance of each of these methods on DNA yield and quality, microbiota composition using quantitative PCR, deep sequencing of the 16S rRNA gene, and sequencing analysis pipeline was evaluated. RESULTS: The CHAO yi

Different sample types in pigs challenged with Haemophilus parasuis following two treatment schemes with tulathromycin.

This study aimed to test the efficacy of samplings for the detection of Haemophilus parasuis after metaphylactic treatment and subsequent challenge using an established model for Glässer's disease. In this model, 36 piglets were equally assigned to a negative control, a positive control, and two trial groups receiving tulathromycin 7 or 4 days prior to challenge. The piglets of three groups were challenged intratracheally with H. parasuis serovar 5. As a result, four pigs in each challenged group died or had to be euthanised within 10 days post challenge. The remaining 15 pigs of these challen

Direct Invasion of the Optic Nerves, Chiasm, and Tracts by Cryptococcus neoformans in an Immunocompetent Host.

Cryptococcus spp is a common fungal infection and frequent cause of meningitis in immunocompromised patients; however, immunocompetent patients are also at risk of infection. Visual loss often occurs via elevated intracranial hypertension but can rarely occur through direct optic nerve, chiasm, or tract invasion. We report a case of a 38-year-old woman who presented with decreased acuity in both eyes. She had generalized visual field constriction in the right eye and temporal hemianopsia in the left eye. Magnetic resonance imaging of the brain and orbits showed multiple areas of ill-defined en

[An Early Neonatal Sepsis Case Due to Streptococcus pneumoniae Serotype 7F].

Streptococcus pneumoniae infections which are relatively unusual in the neonates, are associated with substantial morbidity and mortality. Although S.pneumoniae has more than 90 serotypes, invasive disease is more common with certain serotypes. Pneumococcal polysaccharide and conjugate vaccines are developed against the serotypes that cause invasive disease. Until now invasive pneumococcal diseases have been defined due to various serotypes in the neonatal period. Here we present a neonate with an invasive pneumococcal disease due to S.pneumoniae serotype 7F which has not yet been reported in

Proteasome particle-rich structures are widely present in human epithelial neoplasms: correlative light, confocal and electron microscopy study.

A novel cytoplasmic structure has been recently characterized by confocal and electron microscopy in H. pylori-infected human gastric epithelium, as an accumulation of barrel-like proteasome reactive particles colocalized with polyubiquitinated proteins, H. pylori toxins and the NOD1 receptor. This proteasome particle-rich cytoplasmic structure (PaCS), a sort of focal proteasome hyperplasia, was also detected in dysplastic cells and was found to be enriched in SHP2 and ERK proteins, known to play a role in H. pylori-mediated gastric carcinogenesis. However, no information is available on its o

Structure of a MenB de-N-acetyl polysialic acid antibody and mechanism of immune cell inhibition.

The Neisseria meningitidis serogroup B (MenB) polysaccharide, α(2-8) polysialic acid (polySia), is essential for resistance to complement-mediated bacteriolysis and inhibition of opsonophagocytosis. The monoclonal antibody SEAM 3 binds de-N-acetyl polySia (dPSA) but not polySia, yet still recognizes encapsulated MenB, indicating the capsule contains dPSA derivatives. Since SEAM 3 also recognizes other pathogens as well as human cancers, we have explored its biological role and mapped the SEAM 3 epitope. We compared binding of N-acetylated and de-N-acetylated polySia and ganglioside GD3 derivat

Patterning bacterial nanocellulose at cell-size scale using UV laser technology to create immunomodulatory interfaces.

The development of immune-instructive biomaterials that actively guide host immune responses toward healing and homeostasis is a key step for advancing the next generation of biomedical implants and regenerative medicine. UV laser technology offers a non-contact, scalable, and versatile approach for directly patterning bacterial nanocellulose (BNC) topography at a cell-size scale. Distinct 40 μm-period topographies were generated on the BNC surface, including anisotropic patterns with distinct morphologies and an isotropic pattern of symmetrically arranged bumps. Scanning electron microscopy a

Contribution of Proteins to Membrane and Cell Wall Structures in Mycobacterium tuberculosis.

The cell wall of Mycobacterium tuberculosis (Mtb) is an exceptionally complex, multilayered structure that is fundamental to the bacterium's survival, pathogenicity, and resistance to both host immune defenses and antibiotics. This cell envelope consists of several distinct components: a surface capsule, an outer membrane, and a peptidoglycan layer that is covalently attached to arabinogalactan. The arabinogalactan is further esterified with mycolic acids, forming the characteristic mycolyl-arabinogalactan-peptidoglycan (mAGP) complex, which is anchored to the underlying plasma membrane. The a

High-fat and low-fiber diet elevates the gut resistome: a comparative metagenomic study.

Antimicrobial resistance (AMR) is a global health challenge. The gut microbiome, a major reservoir for AMR, is influenced by dietary habits. However, the specific impact of dietary patterns on gut resistome remains poorly understood. This study aimed to assess the effects of high-fat/low-fiber and high-fiber/low-fat diets on the development of AMR in the gut microbiome. The shift from a normal diet to a high-fat/low-fiber or a high-fiber/low-fat diet in mice resulted in corresponding increases and decreases in the relative abundance of the resistome (0.14 to 0.25, p < 0.001 vs. 0.14 to 0.09 p

A critical review of diagnostic strategies for bacterial pathogens in opium poppy (Papaver somniferum) cultivation.

Opium poppy (Papaver somniferum) is a pharmaceutically and economically strategic crop in which bacterial diseases cause measurable yield and quality losses. Yet the evidence base for diagnostics in poppy is fragmented, taxonomically inconsistent, and constrained by the crop's narcotic status. This review synthesizes historical records and updates taxonomy for priority taxa-most notably seed-borne Xanthomonas campestris pv. papavericola and soft-rot agents within Pectobacterium spp.-and compares conventional, molecular, immunological, point-of-care, and non-invasive optical/spectral approaches

Therapeutic efficacy of a probiotic preparation on idiopathic halitosis: a retrospective observational study.

Idiopathic halitosis is an unusual condition of unclear causes, which has never been thoroughly investigated. We aimed to explore the role of small intestinal bacterial overgrowth (SIBO) in the pathogenesis of idiopathic halitosis, and to evaluate the therapeutic efficacy of a probiotic preparation on this condition. This retrospective observational study included 162 idiopathic halitosis patients and 198 healthy controls (HCs). Halitosis was diagnosed using the organoleptic test, and idiopathic halitosis was diagnosed by excluding known causes. SIBO was identified through the hydrogen/methane
