Participation of opioid receptors in the modulation of the analgesic response by adrenal and gonadal glands.

The involvement of opioid receptors in the analgesic response was evaluated by the tail-immersion test in simultaneously adrenalectomized and ovariectomized female Wistar rats (210-250 g). The reaction time (mean +/- SEM) for tail withdrawal from hot water decreased significantly 2 weeks after surgery (3.52 +/- 0.20 s) when compared to intact animals (6.09 +/- 0.23 s). Hormonal replacement with dexamethasone (50 micrograms/day) did not affect reaction time (3.38 +/- 0.19 s). However, this response was restored by combined adrenal and gonadal steroid substitution (estradiol 5 micrograms/day and

Antinociception in the rat induced by a cold environment.

Rats placed in a cold environment (4 degrees C) for 2 h had a sustained increase in tail flick latency (TFL) as well as an increase in tail pinch latency (TPch) that was often biphasic with an early peak response at 15 min and a later, often higher, peak at 2 h. Plasma beta-endorphin levels after a modest increase at 5 min (24%) declined throughout the remaining time in the cold. The long-acting opioid antagonist naltrexone had no effect on TFL increases but led to greater increases in TPch (P less than 0.04). In morphine-tolerant rats TFL response was the same as in controls but TPch increase

Paradoxical analgesia produced by low doses of the opiate-antagonist naloxone is mediated by interaction at a site with characteristics of the delta opioid receptor.

This study addressed the opiate receptor subclass which underlies the paradoxical analgesic action of intrathecal administration of low doses of the stereospecific opiate receptor antagonist, naloxone. The analgesic effect of low dose naloxone was abolished in rats that had been pretreated 24 hr earlier with a large intrathecal dose of naloxone or 20 min previously with the delta receptor specific antagonist, ICI-174,864. A large intrathecal dose of naloxone administered 24 hr previously also abolished the analgesic effects of the delta-specific ligands [D-Pen2,5]-enkephalin and [D-Ser2]-Leu e

Modulation of humoral immune responses by endogenous opioids.

The effects of opioid agonists and antagonists were investigated on humoral immune mechanisms in mice and rats. Opioid agonists like morphine, Leu-enkephalin, and Met-enkephalin, enhanced antigen-induced histamine release from mixed peritoneal cells of rats in vitro; this enhancement was effectively antagonized by naloxone, an opioid antagonist. Naloxone, per se, decreased anaphylactic mortality in doses of 10 mg/kg, while it increased mortality in a dose of 1 mg/kg. Reduced IgE antibody titer, measured by passive cutaneous anaphylaxis, decreased hemagglutination titer to sheep red blood cells

Effects of central administration of morphine on renal function in conscious rats.

Systemic administration of morphine in rats produces an anti-natriuretic effect that is at least partially dependent on renal nerves. The present studies were carried out in order to assess the renal response to central administration of morphine. Male Sprague-Dawley rats were surgically prepared with arterial, venous and bladder cannulas. In addition, a guide cannula was placed into the lateral ventrical and secured to the surface of the skull. Experiments were carried out at least 3 days after surgery. Renal clearance measurements were 30 min each. After a basal period, morphine sulfate (4 m

Role of the pineal gland in immunity. III. Melatonin antagonizes the immunosuppressive effect of acute stress via an opiatergic mechanism.

We have recently demonstrated that the pineal neurohormone melatonin exerts important immunoregulatory functions. We now report that exogenous melatonin counteracts completely the effect of acute anxiety-restraint stress on thymus weight and antibody response to sheep red blood cells (SRBC). In addition, administration of melatonin in the evening prevented paralysis and death of mice infected with sublethal doses of encephalomyocarditis virus (EMCV) after acute stress. The anti-stress activity of melatonin was present in mice injected with T-dependent antigens, and it was abolished by the cont

Opioid peptides from milk as a possible cause of sudden infant death syndrome.

Milk from breast or baby formula is the exclusive source of nutrition for newborn infants. Short chain opioid peptides such as beta-casomorphins have been isolated from breast milk as well as baby formula. These biologically active peptides are absorbed from the gastrointestinal tract. In infants predisposed to respiratory apnea because of abnormal autonomic nervous system development and respiratory control mechanisms, opioid peptides derived from milk might be one of the etiological factors for sudden infant death syndrome and near miss sudden infant death syndrome.

Cardiovascular responses to morphiceptin in spontaneously hypertensive and normotensive rats.

In the urethane anesthetized spontaneously hypertensive rats (SHR) intraventricular injections of morphiceptin produced dose-related increase in heart rate and blood pressure. In contrary, intraventricular administration of morphiceptin in Wistar rats induced a fall in blood pressure and heart rate. Yohimbine antagonized pressor responses to morphiceptin in SHR. Naloxone counteracted both the stimulatory effects of morphiceptin in SHR as well as hypotensive responses in Wistar rats. Bilateral vagotomy blocked depressant action of morphiceptin in normotensive but failed to alter the pressor eff

Dynorphin: potent analgesic effect in spinal cord of the rat.

Evidences are presented to show a strong and long-lasting analgesic effect after injection of dynorphin into the subarachnoid space of the spinal cord in the rat. Taking the amplitude and time course of the increase of tail flick latency as the indices of analgesia, dynorphin elicited dose-dependent analgesic effect in the range of 2.3-18.6 nmol. Calculating on a molar basis dynorphin was 6-10 times more potent than morphine and 65-100 times more potent than morphiceptin, another mu opiate receptor agonist. Dynorphin analgesia was completely reversed by intrathecal injection of anti-dynorphin

Food intake: opioid/purine interactions.

The exogenous opioids butorphanol tartrate (BT) and ethylketocyclazocine (EKC) have been reported to stimulate feeding in rats. In this study we evaluated the effects of purines (known to suppress feeding) and the adenosine antagonist, caffeine, on opioid induced feeding. Adenosine and inosine significantly suppressed BT and EKC induced feeding at various doses and time points. Caffeine enhanced food consumption was suppressed by various doses of naloxone, but was not suppressed by adenosine or inosine. Although caffeine itself induced further feeding, it did not enhance BT induced food consum

The effect of somatostatin and enkephalin on ion transport in the intestine.

SRIF and enkephalin stimulate net Na and C1 absorption in the rabbit ileum and appear to do so primarily by stimulating the coupled influx of Na and C1 across the brush border membrane. The stimulatory effect of enkephalin on ion transport is blocked by verapamil and Ca-free solution, indirectly indicating that enkephalin may work as a Ca-channel blocker. The resultant lowering in cytosolic calcium level stimulates Na and C1 absorption. SRIF was able to block the effect of cyclic AMP-dependent and cyclic AMP-independent secretogogues in the rat colon without affecting cyclic AMP levels suggest

[Schizophrenia and addiction].

The relationship between schizophrenia and drug abuse has been approached using physiopathological, psychopathological, and cybernetics communicational techniques. The hypotheses which have been proposed with regard to the physiological mechanism involved in schizophrenia and drug abuse all have in common, among other things, the idea that neurotransmitters play a role in the development of these behaviors. Likewise, the psychopathological symptoms presented by schizophrenia and drug abuse offen many similarities. The mechanism by which drug abuse may lead to schizophrenia is discussed. The an

Moderate protective effect of Kyotorphin against the late consequences of intracerebroventricular streptozotocin model of Alzheimer's disease.

The established decrease in the level of endogenous kyotorphin (KTP) into the cerebrospinal fluid of patients with an advanced stage of Alzheimer's disease (AD) and the found neuroprotective activity of KTP suggested its participation in the pathophysiology of the disease. We aimed to study the effects of subchronic intracerebroventricular (ICV) treatment (14 days) with KTP on the behavioral, biochemical and histological changes in rats with streptozotocin (STZ-ICV)-induced model of sporadic AD (sAD). Three months after the administration of STZ-ICV, rats developed increased locomotor activity

Suppressive effect of vagal afferents on the activity of the trigeminal spinal neurons related to the jaw-opening reflex in rats: involvement of the endogenous opioid system.

The purpose of the present study is to test the hypothesis that via the endogenous pain control system, vagal afferent input modulates the activity of the trigeminal spinal nucleus oralis (TSNO) related to the tooth pulp (TP)-evoked jaw-opening reflex (JOR). Extracellular single-unit recordings were made from 36 TSNO units responding to TP electrical stimulation with a constant temporal relationship to a digastric electromyogram (dEMG) signal in 26 pentobarbital-anesthetized rats. The activity of 36 TSNO neurons and the amplitude of the dEMG increased proportionally during 1.0-3.5 times the th

[The effect of short peptides isolated from the brain of a hibernating suslik on the rat EEG and behavior].

The intracerebroventricular injection of the peptides kyotorphin (KT), newkyotorphin (NKT), and Aps-Tyr (BS) changed rat behaviour. There was an increase in the rate of exploratory reactions, rearing, grooming, stereotypic scratching movements, yawning, stretching, hiccupping, sneezing. Similar effects had been earlier observed in rats after administration of the fraction (F, 1-10 kD) from the brain of hibernating ground squirrels in which the studied peptides were found. The EEG effects depended on a peptide dose. As in the case of F injection, KT in the dose of 4 mcg suppressed the theta- an

A naturally occurring opioid peptide from cow's milk, beta-casomorphine-7, is a direct histamine releaser in man.

beta-Casomorphine-7, a naturally occurring product of cow's milk with opiate-like activity, was studied for possible direct histamine liberation activities in humans. It was found to cause concentration-dependent in vitro histamine release from peripheral leukocytes of healthy adult volunteers. Intradermal injection of beta-casomorphine-7 induced a wheal and flare reaction in the skin similar to histamine or codeine. Oral pretreatment with the H1 antagonist terfenadine significantly inhibited the skin responses to beta-casomorphine-7. The intradermal injection of an opiate receptor antagonist,
