Altered Cav1.2 function in the Timothy syndrome mouse model produces ascending serotonergic abnormalities.

Polymorphism in the gene CACNA1C, encoding the pore-forming subunit of Cav1.2 L-type calcium channels, has one of the strongest genetic linkages to schizophrenia, bipolar disorder and major depressive disorder: psychopathologies in which serotonin signaling has been implicated. Additionally, a gain-of-function mutation in CACNA1C is responsible for the neurodevelopmental disorder Timothy syndrome that presents with prominent behavioral features on the autism spectrum. Given an emerging role for serotonin in the etiology of autism spectrum disorders (ASD), we investigate the relationship betwee

Plasmacytoid dendritic cell-mediated L-glutamate catabolism links gut microbiota to male infertility.

Emerging evidence suggests that gut microbiota composition influences male reproductive health; however, the immunometabolic mechanisms underlying this association remain insufficiently characterized. We investigated whether specific immune cell-mediated metabolic pathways, particularly plasmacytoid dendritic cell (pDC)-driven L-glutamate catabolism via the hydroxyglutarate pathway, contribute to the causal link between gut microbiota and male infertility. We conducted a 2-sample, 2-step Mendelian randomization (MR) analysis using inverse-variance weighting as the primary estimator and Bayesia

Mendelian randomization analysis reveals causal links between specific immune cell phenotypes and Parkinson's disease.

OBJECTIVE: Immunity and neuroinflammation are key factors in Parkinson's Disease (PD); however, detailed studies on how they are causally related to each other are lacking. This study aimed to explore the potential causal relationship between immune cells and PD. METHODS: A comprehensive two-sample Mendelian Randomization (MR) analysis was conducted based on publicly available genetic data to explore the causal relationship between 731 immune cells and PD risk. Inverse Variance Weighted (IVW) served as the main technique to explore the causal relationship, along with methods such as weighted

Nabais Sá-de Vries Syndrome Type 1 in a Mexican Girl: A Case Report.

Nabais Sá-de Vries syndrome (NSDVS) is a rare disease caused by a heterozygous mutation in the SPOP gene (17q21), which is involved in protein degradation via the ubiquitin-proteasome pathway. Different germline variants within this gene cause two distinct phenotypes (allelic heterogeneity) that affect multiple organs and systems (pleiotropy). These variants are clinically associated with intellectual disability, neurological disorders, and dysmorphic features that exhibit variable expressivity. Here, we present the case of a five-year-old girl who was conceived through assisted reproductive t

Causal associations between antibody-mediated immune responses to infectious agents and cardiovascular inflammatory diseases: A Mendelian randomization study based on UK Biobank and FinnGen.

A two-sample Mendelian randomization study was performed to evaluate causal associations between 46 pathogen-specific antibodies and six cardiovascular inflammatory diseases: myocarditis, pericarditis, endocarditis, coronary atherosclerosis (CAS), polyarteritis nodosa (PAN), and giant cell arteritis (GCA). Antibody data came from UK Biobank, diseases data came from FinnGen. Instrumental variables were selected using strict (p < 5 × 10-8) and lenient threshold (p < 5 × 10-6). Benjamini-Hochberg method was used to control FDR; FDR‑adjusted q < 0.05 defined "robust associations." Under strict thr

Immune cells, inflammatory proteins, and sepsis: A mediation Mendelian randomization study.

Immune cells are known to be associated with sepsis. However, whether these associations represent a causal relationship and whether inflammatory proteins act as mediators remain unclear. A mediation Mendelian Randomization (MR) approach was employed to assess the correlation between immune cells and sepsis, along with the mediating effects of inflammatory proteins in this relationship. Inverse variance weighting (IVW) was used as the main statistical method, with MR-Egger and weighted median serving as supplements in the preliminary MR. Sensitivity analyses were implemented using Cochrane Q t

Comprehensive genotype-phenotype analysis in POLR3-related disorders.

RNA polymerase III (RNA Pol III)-related disorders (POLR3-RDs) are a group of clinical entities characterized by causal variants in genes encoding RNA Pol III subunits, including POLR3A, POLR3B, POLR1C, POLR1D, POLR3D, POLR3E, POLR3F, POLR3GL, POLR3H, and POLR3K. These typically cause developmental phenotypes affecting the central nervous system; the eyes; connective tissues including bones, teeth, and endocrine axes; and the reproductive system. Similar phenotypes can be caused by variants in separate subunit genes (multigenic). In contrast, variants in the same gene can cause different pheno

Mendelian randomization analysis does not reveal a causal association between migraine and Meniere's disease.

BACKGROUND: According to observational research, migraine may increase the risk of Meniere's disease (MD). The two have not, however, been proven to be causally related. METHODS: Using Mendelian random (MR) analysis, we aimed to evaluate any potential causal relationship between migraine and MD. We extracted single-nucleotide polymorphisms (SNPs) from large-scale genome-wide association studies (GWAS) involving European individuals, focusing on migraine and MD. The main technique used to evaluate effect estimates was inverse-variance weighting (IVW). To assess heterogeneity and pleiotropy, se

Association of dietary intake with pneumothorax: A Mendelian randomization study.

BACKGROUND: An association between dietary habits and lung disease has been demonstrated in previous studies. Employing Mendelian randomization, we aimed to explore how different dietary intakes relate to pneumothorax, shedding light on the interplay among gut flora, the lung-gut axis, and pneumothorax. METHODS: Employing both two-sample and multi-sample Mendelian randomization (MR) analyses, we investigated 24 dietary intake variables to establish a strong association with pneumothorax. Causal inferences were drawn using the inverse variance weighted (IVW) method. To fortify our findings, we
