Preparing Irradiated and Marked Male Aedes aegypti Mosquitoes for Release in an Operational Sterile Insect Technique Program.

The control of such human diseases as dengue, Zika, and chikungunya relies on the control of their vector, the Aedes aegypti mosquito, because there is no prevention. Control of mosquito vectors can rely on chemicals applied to the immature and adult stages, which can contribute to the mortality of non-targets and more importantly, lead to insecticide resistance in the vector. The sterile insect technique (SIT) is a method of controlling populations of pests through the release of sterilized adult males that mate with wild females to produce non-viable offspring. This paper describes the proce…

Ecological drivers of genetic connectivity for African malaria vectors Anopheles gambiae and An. arabiensis.

Anopheles gambiae and An. arabiensis are major malaria vectors in sub-Saharan Africa. Knowledge of how geographical factors drive the dispersal and gene flow of malaria vectors can help in combatting insecticide resistance spread and planning new vector control interventions. Here, we used a landscape genetics approach to investigate population relatedness and genetic connectivity of An. gambiae and An. arabiensis across Kenya and determined the changes in mosquito population genetic diversity after 20 years of intensive malaria control efforts. We found a significant reduction in genetic dive…

Clinical development of placental malaria vaccines and immunoassays harmonization: a workshop report.

Placental malaria caused by Plasmodium falciparum infection constitutes a major health problem manifesting as severe disease and anaemia in the mother, impaired fetal development, low birth weight or spontaneous abortion. Prevention of placental malaria currently relies on two key strategies that are losing efficacy due to spread of resistance: long-lasting insecticide-treated nets and intermittent preventive treatment during pregnancy. A placental malaria vaccine would be an attractive, cost-effective complement to the existing control tools. Two placental malaria vaccine candidates are curre

Lnc40766-TLR6-miR14-3p Regulation Pathway Mediates the Immunity of Plutella xylostella against Entomopathogenic Fungus Isaria cicadae.

The diamondback moth Plutella xylostella is a major agricultural pest with severe insecticide resistance. Entomopathogenic fungi (EPF) serve as eco-friendly biocontrol. Insects rely on immunity to resist fungal invasion, while the regulatory mechanism of long non-coding RNAs (lncRNAs) in insect antifungal immunity remains unclear. Here, lncRNA-mediated immune responses of P. xylostella against EPF Isaria cicadae were explored. RNA-seq identified 33,402 lncRNAs, 97 of which were predicted to cis-regulate 72 immune genes. The lnc40766 targets Toll-like receptor 6 (TLR6); both were upregulated po…

A male-drive female-sterile system for the self-limited control of the malaria mosquito Anopheles gambiae.

Despite great leaps forward in preventing and treating malaria, several challenges, including insecticide resistance, have hindered progress in fighting the disease. Thus, there is a pressing need for new tools to control malaria, including the use of genetically modified mosquitoes (GMMs) in the field. Various genetic strategies for vector control are currently explored, ranging from self-sustaining GMMs with unrestricted geographic and temporal spread to self-limiting alternatives. Here, we describe a self-limiting gene drive strategy called Male Drive Female Sterile (MDFS) targeting Anophel…

CRISPR-based gene drives generate super-Mendelian inheritance in the disease vector Culex quinquefasciatus.

Culex mosquitoes pose a significant public health threat as vectors for a variety of diseases including West Nile virus and lymphatic filariasis, and transmit pathogens threatening livestock, companion animals, and endangered birds. Rampant insecticide resistance makes controlling these mosquitoes challenging and necessitates the development of new control strategies. Gene drive technologies have made significant progress in other mosquito species, although similar advances have been lagging in Culex. Here we test the first CRISPR-based homing gene drive for Culex quinquefasciatus, demonstrati…

The scale insects: Its status, biology, ecology and management in tea plantations.

The scale insects reduce plant photosynthetic ability by sucking sap from leaves and causing significant damage to the tea crop in most tea-producing countries. They suck the sap from stems and tea leaves, which not only prevents further growth but also reduces the nutritional quality of the leaves by promoting the growth of sooty molds. However, due to the widespread use of organosynthetic pesticides in recent decades, most insect pests have developed high levels of pesticide resistance, reducing the effectiveness of insecticide application. Bio-control agents are environmentally safe and pro

Dermatologic aspects of bed bug epidemic: an atlas of differential diagnosis.

Bed bug infestation rate has gone through an unforeseen increase in the past decades worldwide. Their resurgence is a consequence of numerous factors, including growing population density, increased international travel and the spread of insecticide resistance. Bed bug infestation is often revealed by skin symptoms appearing after their bite in sensitive patients. Medical professionals encountering patients with bed bug bites have responsibility for recognizing the condition and for instructing patients about the necessary measures for eradication. Setting the correct diagnosis, however, is no…

[Role of children in the evaluation of the Onchocerciasis Control Program in West Africa].

Incidence of onchocercal infections is studied in children borne since the start of the larvicidal treatments of the Onchocerciasis Control Programme in West Africa (OCP). A total of 8088 children, originating from 155 villages widely distributed throughout the initial area of the OCP were examined for microfilariae of O. volvulus. 37 were found infected. If there had not been control measures, 652 children would normally have become infected in such a sample. It is considered that in the centre of the OCP area which constitutes approximately 90% of the total, transmission has been interrupted

Quantitative trait loci mapping of genome regions controlling permethrin resistance in the mosquito Aedes aegypti.

The mosquito Aedes aegypti is the principal vector of dengue and yellow fever flaviviruses. Permethrin is an insecticide used to suppress Ae. aegypti adult populations but metabolic and target site resistance to pyrethroids has evolved in many locations worldwide. Quantitative trait loci (QTL) controlling permethrin survival in Ae. aegypti were mapped in an F(3) advanced intercross line. Parents came from a collection of mosquitoes from Isla Mujeres, México, that had been selected for permethrin resistance for two generations and a reference permethrin-susceptible strain originally from New Or

Development of 1,2,4-oxadiazole-containing isoxazoline insecticides with improved honeybee compatibility.

BACKGROUND: Spodoptera frugiperda (S. frugiperda) and Plutella xylostella (P. xylostella) are major constraints to staple and cruciferous crops, respectively, and have evolved widespread resistance to anthranilic amide insecticides. Isoxazolines, acting as γ-aminobutyric acid (GABA) receptor modulators, are promising alternatives for controlling resistant populations. However, many members of this class exhibit high toxicity toward ecologically important pollinators. Here, we report a series of new isoxazoline analogues featuring a 1,2,4-oxadiazole motif designed by a ring-closure strategy to

Malaria: a review on its current epidemiological status and management strategies.

Malaria remains a global health concern, with Africa bearing the highest global burden of malaria, as Plasmodium falciparum malaria remains the leading cause of malaria-related mortality on the continent. The transmission dynamics of malaria are shaped by a combination of factors, including climate conditions, economic constraints, geographic variability, human activities, and security instability. Owing to repeated infections and widespread implementation of early interventions, there has been a notable rise in cases of clinically atypical malaria and asymptomatic Plasmodium carriers, which i

Characterization of ATP-binding cassette transporters associated with emamectin benzoate tolerance: from the model insect Drosophila melanogaster to the agricultural pest Spodoptera frugiperda.

BACKGROUND: Multiple families of detoxification genes, including the increasingly recognized family of ATP-binding cassette (ABC) transporters, work together to influence the toxicity of synthetic insecticides and thus their resistance. Effective management of insecticide resistance requires identification of all toxicity-affecting members from each family of toxicity-related genes. RESULTS: Here, we used emamectin benzoate (EB), ABC transporters and Spodoptera frugiperda as a working case to test whether the strategy of 'from the model insect Drosophila melanogaster to agricultural pests' ca…

Activity of tea tree oil and nerolidol alone or in combination against Pediculus capitis (head lice) and its eggs.

Head lice infestation is an emerging social problem in undeveloped and developed countries. Because of louse resistance increasing, several long-used insecticidal compounds have lost their efficacy, and alternatives, such as essential oils, have been proposed to treat this parasitic infestation. The present study investigated the efficacy of two natural substances: tea tree (Melaleuca alternifolia) oil and nerolidol (3,7,11-trimethyl-1,6,10-dodecatrien-3-ol) against lice and its eggs. Products were used alone and in combination (ratio 1:1 and 1:2) from 8 % dilution. The in vitro effect of natu

Vector control with driving Y chromosomes: modelling the evolution of resistance.

BACKGROUND: The introduction of new malaria control interventions has often led to the evolution of resistance, both of the parasite to new drugs and of the mosquito vector to new insecticides, compromising the efficacy of the interventions. Recent progress in molecular and population biology raises the possibility of new genetic-based interventions, and the potential for resistance to evolve against these should be considered. Here, population modelling is used to determine the main factors affecting the likelihood that resistance will evolve against a synthetic, nuclease-based driving Y chro

An orally available PfPKG inhibitor blocks Plasmodium's infection of the liver.

Malaria remains a global health threat exacerbated by emerging resistance to antimalarial therapies and insecticides, climate-driven outbreaks, and limited chemoprotective options. Here, we report the characterization of RUPB-61, the first orally bioavailable inhibitor of Plasmodium falciparum cGMP-dependent protein kinase (PfPKG). RUPB-61 prevents infection by P. falciparum and P. cynomolgi sporozoites, including the formation of hypnozoites by the latter. A single oral dose blocks liver infection by P. berghei sporozoites in vivo, demonstrating efficacy consistent with further development as

Inheritance and fitness costs of field-evolved resistance to Gpp34/Tpp35Ab1 corn by western corn rootworm, Diabrotica virgifera virgifera (Coleoptera: Chrysomelidae).

BACKGROUND: The western corn rootworm, Diabrotica virgifera virgifera LeConte, is among the most serious pests of corn in the United States, and an invasive pest in parts of Europe. In the United States, feeding injury to roots by western corn rootworm larvae is managed, in part, with genetically modified corn hybrids that produce insecticidal proteins derived from the bacterium Bacillus thuringiensis (Bt). The widespread use of Bt corn has contributed to field-evolved resistance to all commercially available Bt traits by some western corn rootworm populations. Insect resistance management (IR

Essential Oils from Neolamarckia cadamba: Methyl Salicylate-Rich Stem Bark Oil as a Multi-Functional Biopesticide with Insecticidal and Antifungal Efficacy.

The escalating challenges of insecticide resistance and environmental pollution underscore the urgent need for sustainable and multi-functional biopesticides. This study reveals the chemical diversity and potent bioactivity of essential oils (EOs) from Neolamarckia cadamba, highlighting their potential as a valuable source of bioactive agents. Gas chromatography-mass spectrometry analysis revealed a striking contrast between the essential oils: the stem bark EO was dominated by methyl salicylate (MeSA, 97.61%), representing the first report of MeSA as a major constituent in this species, while…
