Exploring the molecular basis of insecticide resistance in the dengue vector Aedes aegypti: a case study in Martinique Island (French West Indies).

BACKGROUND: The yellow fever mosquito Aedes aegypti is a major vector of dengue and hemorrhagic fevers, causing up to 100 million dengue infections every year. As there is still no medicine and efficient vaccine available, vector control largely based on insecticide treatments remains the only method to reduce dengue virus transmission. Unfortunately, vector control programs are facing operational challenges with mosquitoes becoming resistant to commonly used insecticides. Resistance of Ae. aegypti to chemical insecticides has been reported worldwide and the underlying molecular mechanisms, in…

Quantification of the efficiency of treatment of Anopheles gambiae breeding sites with petroleum products by local communities in areas of insecticide resistance in the Republic of Benin.

BACKGROUND: The emergence of Anopheles populations capable of withstanding lethal doses of insecticides has weakened the efficacy of most insecticide based strategies of vector control and, has highlighted the need for developing new insecticidal molecules or, improving the efficacy of existing insecticides or abandoning those to which resistance has emerged. The use of petroleum products (PP) against mosquito larvae had an immense success during early programmes of malaria control, but these compounds were abandoned and replaced in the 1950s by synthetic insecticides probably because of the h…

A dietary risk assessment of the pyrethroid insecticide resmethrin associated with its use for West Nile Virus mosquito vector control in California.

An outbreak of human illnesses associated with West Nile Virus (WNV) occurred in New York City in 1999. Since then, it has gradually spread westwards, reaching northern California for the first time in 2005. WNV is transmitted by several mosquito species and birds serve as the main reservoir. Several control measures have been used, targeting both the aquatic larvae and the adult mosquitoes. In the latter case, roosting birds in trees are sprayed with pyrethroid insecticides because these are highly toxic to mosquitoes, but have low avian toxicity. A request was made to use a resmethrin-contai…

Insecticide resistance spectra and resistance mechanisms in populations of Japanese encephalitis vector mosquitoes, Culex tritaeniorhynchus and Cx. gelidus, in Sri Lanka.

Culex tritaeniorhynchus Giles and Cx. gelidus Theobald (Diptera: Culicidae), both vectors of Japanese encephalitis, were collected in 1984 and 1998 from two disease endemic localities in Sri Lanka: Anaradhapura and Kandy. Using wild-caught adult mosquitoes from light traps, log dosage-probit mortality curves for insecticide bioassays were obtained for three insecticides: malathion (organophosphate), propoxur (carbamate) and permethrin (pyrethroid). LD50 values showed that, in 1998, Cx. tritaeniorhynchus was -100-fold more resistant to malathion and 10-fold more resistant to propoxur than was C…

Novel insecticidal peptides from Tegenaria agrestis spider venom may have a direct effect on the insect central nervous system.

Fractionation of venom from an agelenid spider, Tegenaria agrestis, resulted in the isolation of a family of three peptides with potent insecticidal activity. These peptide toxins, TaITX-1, -2, and -3, whose sequences were revealed from cloned cDNAs, each consist of 50 amino acid residues, six of which are cysteines. They appear to be amidated at their C-termini and exhibit greater than 90% sequence identity. Unlike other reported spider toxins, the TaI toxins are processed from precursors containing no propeptide sequences. In lepidopteran larvae and corn rootworm beetles, the insecticidal Te

Spider peptides from Brachypelma smithi with slightly different amino acids at their C-terminal loops exert different insecticidal activities.

The insecticidal molecules of spiders persistently evolve to ensure rapid paralysis of their prey, and the best molecules are transmitted to their progeny. Here, we cloned two insecticidal peptides, Bs2 and Bs3, from the venom glands of the theraphosid Brachypelma smithi. Bs2 and Bs3 are 90.2% identical, but they exhibit interesting structural differences at their C-termini, including a connecting disulfide bond (residues Cys15-Cys36 for Bs2 and Cys15-Cys30 for Bs3). The genomic origin of Bs2 and Bs3 may be a cause for gene duplication events. Moreover, Bs2 differs in two residues from Tal1 (9

Evaluation of the insecticide custody chain and its relationship with malaria burden in the Brazilian Amazon: a process and exploratory impact assessment (2019-2023).

BACKGROUND: Malaria remains a major public health concern in Brazil, with the Amazon region accounting for 99.9% of the country's cases. Indoor residual spraying (IRS) using Etofenprox 20% PM is a core vector control strategy. However, inefficiencies in the insecticide custody chain, including planning, storage, and distribution, may compromise intervention effectiveness. OBJECTIVES: This study aimed to evaluate the insecticide custody chain from a process perspective, identifying logistical bottlenecks, while also exploring potential impact by examining associations between insecticide alloc

Effects of cypermethrin insecticide on the growth and plant growth-promoting activity of Burkholderia-like strain (UAGC867) and Pseudomonas-affiliated strain (UAGC97).

Brazil is a global leader in grain production, with corn reaching 122 million tons in the 2024/2025 harvest. Pest control, particularly of Spodoptera frugiperda, depends heavily on cypermethrin, a Class II hazardous synthetic insecticide with significant environmental risks. This study assessed the tolerance and degradation capacity of Pseudomonas-affiliated strain and Burkholderia-like strain in cypermethrin-contaminated environments, along with their indole-3-acetic acid (IAA) production and impact on early maize development. Both strains tolerated the insecticide and grew at varying concent

Dietary grape seed extract mitigated growth retardation, hormonal delay, and gastrointestinal toxicity induced by insecticide imidacloprid in Nile tilapia.

Overusing the insecticide imidacloprid (IMD) in agriculture has led to its presence in water bodies, causing serious environmental issues and fish toxicity. This study explored the potential benefits of grape seed extract (GSE) in mitigating IMD-induced growth and hormonal and gastrointestinal toxicity in Nile tilapia. A total of 240 healthy juvenile tilapias Oreochromis niloticus (O. niloticus), weighing an average of 11.44 ± 2.01 g at 2 weeks of age, were divided equally into four groups, each with three replicates. The control group received no special treatment, while the second group was

Effectiveness and Efficacy of Long-Lasting Insecticidal Nets for Malaria Control in Africa: Systematic Review and Meta-Analysis of Randomized Controlled Trials.

Background: Long-lasting insecticidal nets (LLINs) have significantly reduced the malaria burden in recent decades, and this malaria prevalence reduction has been achieved through the upgrading of pyrethroid long-lasting insecticidal nets. However, this reduction has stalled due to many factors, including rapidly developing pyrethroid resistance. Method: The protocol was registered in PROSPERO, and we used Cochrane methodology to assess bias and evidence quality. Three reviewers extracted data from individual studies, and a meta-analysis was performed using Excel and STATA version 17, expressi

Hazard assessment of insecticidal proteins representing different structural classes using an in vitro experimental platform of human intestinal epithelial monolayers.

Hazard assessment of proteins using human-derived intestinal epithelial cell (IEC) lines has been investigated previously. The purpose of this work was to a) reduce occurrences of false positives by including a non-hazardous protein (BSA) negative control while redesigning statistical analyses; and b) consider the potential of the IEC assay as a component of a weight of evidence approach in the hazard assessment of four different structural/functional classes of insecticidal proteins. The protein classes evaluated included: Mpf (Mpf2Ba1); Cyt (IPD084Aa); Cry (IP3-H9); and uncharacterized (IPD0

Determinants of access to insecticide-treated nets in Sub-Saharan Africa: A multilevel cross-country analysis using 29 DHS data.

BACKGROUND: Having access to Insecticide-Treated Nets (ITNs) is crucial for avoiding malaria in Sub-Saharan Africa (SSA), where the disease burden is disproportionately high. Despite their efficacy, socioeconomic, demographic, and geographic factors continue to cause notable differences in ITN access within and between nations. By employing a multilevel analysis of data from 29 Demographic and Health Surveys (DHS) throughout SSA, this study seeks to fill knowledge gaps about the factors that influence access at the individual and community levels. METHODS: The study utilized data from 29 DHS
