Connectome of a human foveal retina.

The fovea is a unique specialization of the primate retina and is a promising site for obtaining the first complete connectome of a human central nervous system (CNS) structure. Within the fovea, neural cells and circuits have been miniaturized and compressed during evolution to sample the visual image at highest spatial resolution and begin the neural processing that serves human form, color, and motion perception. Here we present a comprehensive analysis of a sample of human foveal retina using deep learning-based segmentation to reconstruct all cells and synaptic connections at nanoscale re

High-Content Genome-Wide RNAi Screen Reveals CCR3 as a Key Mediator of Neuronal Cell Death.

Neuronal loss caused by ischemic injury, trauma, or disease can lead to devastating consequences for the individual. With the goal of limiting neuronal loss, a number of cell death pathways have been studied, but there may be additional contributors to neuronal death that are yet unknown. To identify previously unknown cell death mediators, we performed a high-content genome-wide screening of short, interfering RNA (siRNA) with an siRNA library in murine neural stem cells after exposure to N-methyl-N-nitroso-N'-nitroguanidine (MNNG), which leads to DNA damage and cell death. Eighty genes were

Mapping structural and functional brain networks associated with migraine.

BACKGROUND: Although numerous neuroimaging studies have reported structural and functional abnormalities in migraine, results have remained heterogeneous. Emerging evidence suggests that neuropsychiatric symptoms may arise from dysfunction within distributed brain networks rather than isolated brain regions; however, migraine-related network abnormalities remain insufficiently explored. METHODS: We integrated findings from 51 neuroimaging publications and extracted reported locations showing structural and functional abnormalities associated with migraine. Using a functional connectivity netw

From Contrasts to Representations: Rethinking fMRI Approaches for Cortico-Cerebellar Networks.

The relationship between the cerebellum and cerebrum remains poorly understood in the neuroimaging literature, despite decades of functional magnetic resonance (fMRI) research. In this opinion article, we argue that this gap reflects fundamental methodological constraints rather than merely a lack of scientific interest. We first discuss challenges in acquiring fMRI data and analysis, with an emphasis on the cerebellum. We then critically evaluate the most common analysis approaches including functional connectivity, univariate contrasts, multivariate pattern analysis, and encoding models, exa

Exercise-Induced modulation of molecular-enriched functional connectivity in Parkinson's disease.

Parkinson's disease (PD) involves degeneration of dopaminergic neurons and dysfunction across multiple neurotransmitter systems, contributing to both motor and cognitive impairments. Aerobic exercise improves clinical outcomes; however, its underlying neural mechanisms remain unclear. Using conventional resting-state fMRI combined with Receptor-Enriched Analysis of functional Connectivity by Targets (REACT), we examined molecular-enriched motor network changes following six months of supervised aerobic training in PD. Exercise-related connectivity changes were inversely correlated with baselin

Disrupting the Frontal Aslant Tract: Network Evidence for Speech Initiation Deficits in a Non-Anglophone Stroke Survivor.

BACKGROUND: The frontal aslant tract (FAT) is a direct cortico-cortical intra-frontal pathway linking the inferior frontal gyrus with the pre-supplementary and supplementary motor areas and is implicated in speech initiation and verbal fluency. However, much of the available evidence regarding its role in speech production derives from studies in English-speaking populations, which may limit the generalizability of these findings to underrepresented languages and linguistic contexts. CASE PRESENTATION: We report a 74-year-old Arabic-speaking man who developed acute non-fluent aphasia followin

Homologous specialization of arcuate fasciculus ventrolateral frontal connectivity in marmosets and humans.

The arcuate fasciculus (af) is a crucial dorsal pathway underpinning human language, yet its weak frontal connectivity in macaques-the standard primate model-creates an evolutionary puzzle. Here, we investigate the common marmoset, a distantly related platyrrhine with high vocal complexity, to test for convergent neural adaptations. By integrating retrograde and anterograde tracing with ultra-high-resolution diffusion MRI, we identified a robust af homolog in marmosets that is anatomically distinct from the superior longitudinal fasciculus. Comparative mapping across marmosets, macaques, chimp

Propagation effects of abnormal beta oscillations on sleep rhythms in Parkinson's disease: A computational study.

Parkinson's disease (PD) is characterized by abnormal beta oscillations (13-30 Hz) within the basal ganglia, which contribute not only to motor symptoms but also to sleep disturbances. In this study, we developed a computational model of the basal ganglia-thalamocortical (BGTC) network that includes the pedunculopontine nucleus (PPN), to investigate the mechanism by which the abnormal oscillations disrupt sleep. The model incorporates key nuclei, neurotransmitter systems, and neural pathways to simulate sleep-related brain activity in Parkinsonian conditions. Our simulations show that abnormal…

The Role of the Ventral Tegmental Area in Reward and Addiction: A Comprehensive Review.

The ventral tegmental area (VTA) is a central hub in reward and motivation, integrating heterogeneous neuronal populations and projection-defined circuits to regulate learning, decision-making, and addiction. This review synthesizes anatomical organization and cellular diversity (dopaminergic, GABAergic, and glutamatergic neurons, including multitransmitter co-release), and maps major efferent pathways constituting the mesolimbic and mesocortical systems. We highlight computational roles of VTA activity, including reward prediction errors, incentive salience, and context memory integration, an

The basolateral amygdala decodes visceral pain and itch via distinct circuits and molecules.

As aversive sensations, visceral pain and itch are highly refractory to conventional treatments. Accumulating evidence suggests that the basolateral amygdala (BLA) is involved in pain and itch; however, how the BLA segregates visceral pain and itch remains unclear. Here, we identify two distinct neuronal populations within the BLA that selectively respond to visceral pain and itch in mice. These populations project to distinct downstream brain regions-the ventromedial hypothalamus (VMH) and dorsomedial striatum (DMS)-forming separate circuits that specifically regulate visceral pain and itch.

Biologically grounded on-chip model identifies selective topographic reorganization within hyperexcitable corticostriatal networks.

The mammalian cerebral cortex projects to the striatum in a precise, hierarchical topography, forming parallel loops that underlie sensorimotor, associative, and limbic processing. Despite the striatum's lack of clear anatomical boundaries, these projections remain functionally segregated, suggesting the existence of intrinsic organizing principles. Disruptions in corticostriatal connectivity and excitability are common in neurodevelopmental disorders, but it remains unclear whether such abnormalities are a cause or a consequence of circuit dysfunction. Here, we hypothesized that the excitabil

Medial Pulvinar Nucleus as a Causal Hub for Heteromodal Naming.

Our ability to retrieve the names of objects in our environment is a fundamental aspect of everyday life. This process requires a complex, dynamic network of cortical and subcortical interactions. While the cortical constituents of this network have been extensively studied with intracranial recordings, the subcortical nodes of the naming network are unclear. We probed the role of the left medial pulvinar nucleus in naming with direct intracranial recordings and stimulation in eight humans (three male, five female) as they named objects using pictures and auditory and written descriptions. We

The neuroanatomy and connectivity of the ventral striatum and ventral capsule area in the Göttingen minipig.

A considerable proportion of patients with major depressive disorder (MDD) do not respond to current treatments. Deep brain stimulation has shown promise as an intervention but has yielded inconsistent outcomes, potentially due to the complex biopsychosocial nature of MDD and the lack of standardized large animal models. The ventral striatum/ventral capsule brain area (VS/VC), involved in emotional and behavioral regulation, is a proposed DBS target. The Göttingen minipig (GM) is an emerging non-primate large animal model; however, the anatomy and connectivity of the GM VS/VC remain poorly cha

Abnormal speech production in veterans with bipolar disorder: Relationship to the arcuate fasciculus.

BACKGROUND: Individuals with bipolar disorder (BD) often present with language abnormalities, but few studies have quantified these features or examined their relationship with white matter microstructure. METHODS: We analyzed language samples from 17 Veterans with BD and 15 healthy Veteran controls elicited with an open-ended prompt about a stressful interpersonal experience. We applied singular value decomposition (SVD) to 275 linguistic features to derive low-dimensional component scores capturing the dominant covariance structure. In parallel, we conducted feature-level between-group test

Thalamocortical connectivity and nucleus-specific thalamic dysfunction in temporal lobe epilepsy: an integrative review.

Temporal lobe epilepsy (TLE) is increasingly recognized as a distributed network disorder extending beyond mesial temporal structures. Among the brain regions implicated in epileptic network organization, the thalamus has emerged as a key hub due to its extensive structural and functional connections with cortical, limbic, and subcortical systems. This integrative review aimed to synthesize current evidence regarding nucleus-specific thalamic abnormalities and thalamocortical connectivity alterations in TLE.A literature search was conducted following PRISMA guidelines using PubMed and Google Sc

The Fusiform Imagery Node: Where vision meets concepts in the left temporal lobe.

We present a perspective that integrates clinical and neuroimaging evidence to argue that voluntary visual mental imagery is supported by a distributed network organized around the Fusiform Imagery Node (FIN) of the left fusiform gyrus. Lesion evidence shows that imagery is consistently impaired when the FIN is damaged or disconnected, while neuroimaging confirms robust, domain-independent activation of this cortical area. Recent 7 T fMRI data further clarify the putative functional attributes of the FIN. First, the FIN appears to subserve domain-general processes, supporting imagery for objec

Whole-Brain Neural Connectivity to Cholinergic Neurons in the Lower Thoracic Intermediolateral Column.

OBJECTIVE: The intermediolateral column (IML) serves as a crucial hub for sympathetic information processing between the brain and peripheral organs, with its defining hallmark being the presence of cholinergic neurons expressing choline acetyltransferase (ChAT). Specifically, the IML of the lower thoracic cord plays a pivotal role in regulating abdominal metabolic and digestive viscera. However, little is known about the whole-brain neural connectivity targeting these lower thoracic IML cholinergic neurons, necessitating further investigation. METHODS: Specific retrograde tracing virus was i

Computational evidence for an inverse relationship between retinal and brain complexity.

Visual neuroscientists have long observed an inverse relationship between brain and retinal complexity: As brain complexity increases across species, retinas adapt to simpler visual processing. Lindsey et al. previously provided a computational explanation for this pattern, showing that shallow networks encode complex features in their first stage of processing, whereas deep networks encode simpler features. Here, these findings are extended to a suite of representational analyses and show that shallow networks generate high-dimensional representations with linear decision boundaries and speci

Altered Default Mode Network Connectivity in Mild Cognitive Impairment: Insights from Resting-State fMRI Studies.

Mild Cognitive Impairment (MCI) is marked by a measurable decline in cognitive function that exceeds typical age-related changes but does not yet qualify as dementia. The brain's Default Mode Network (DMN) remains active during rest and plays a crucial role in introspective processes, such as memory retrieval and self-referential thinking. Resting-state functional magnetic resonance imaging (rs-fMRI) is a non-invasive neuroimaging technique that measures spontaneous fluctuations in blood oxygenation, providing insights into functional connectivity within brain networks. Investigating the DMN u

Brain networks activated when aggravating baseline chronic pain of an individual with new daily persistent headache: a case study.

In this study we examined neuronal mechanisms associated with chronic headache pain by recruiting a participant diagnosed with New Daily Persistent Headache (NDPH), who had the reliable characteristic that his baseline chronic pain could be immediately and consistently modulated (partially relieved or aggravated) by applying deep pressure to specific locations along his face and head. We sought to map brain regions responsive to modulations of his baseline pain using functional magnetic resonance imaging (fMRI). We constructed customized MRI-compatible 3D-printed face masks and skull plates af
