Imatinib mesylate and dermatology part 2: a review of the cutaneous side effects of imatinib mesylate.

Cutaneous reactions to imatinib are common and occur in 9.5% to 69% of patients depending on the series reported. Maculopapular eruptions, erythematous eruptions, edema, and periorbital edema are the most common adverse events observed. Imatinib can also induce severe skin eruptions and generalized skin eruptions. Toxic epidermal necrolysis and Stevens Johnson syndrome has been linked to the use of imatinib. Imatinib has caused acute generalized exanthematous pustulosis. Purpuric vasculitis and mycosis fungoides-like reactions has occurred after imatinib use. Rarer side effects include: hypopi

Azimilide dihydrochloride: a unique class III antiarrhythmic agent.

The introduction of class III (Vaughn Williams classification) antiarrhythmic agents has improved the available drug treatment modalities for managing cardiac supraventricular and ventricular tachyarrhythmia. An appreciation of the importance of the delayed rectifier potassium current in the pathogenesis of various cardiac arrhythmias has led to the development of azimilide, an oral type III potassium channel blocker agent that blocks both the rapid activating component (I(kr), common to sotalol, amiodarone, and ibutilide) and the slow activating component (I(ks), a unique action of azimilide)

Rapid and long duration tolerance to the vagal bradycardic effects of 5-HT1A receptor agonists.

Administration of a single dose of the 5-HT1A receptor high intrinsic agonist U-93385E (either 0.3, 1.0 or 3.0 mg/kg, i.v.) results in a 20-30% decrease in heart rate. In contrast, cumulative dosing of U-93385E (0.01-3.0 mg/kg, i.v.) failed to lower heart rate in the spinal cat. Similarly, infusion of 1 mg/kg of U-93385E over a 2 h period failed to lower heart rate and prevented a bradycardic effect of a single bolus dose of U-93385E or flesinoxan. In contrast, the alpha 2-receptor agonist clonidine decreased heart rate in animals receiving the U-93385E infusion. Single bolus doses of flesinox

Nefazodone: its place among antidepressants.

OBJECTIVE: To review the pharmacology, pharmacokinetics, efficacy, adverse effects, and drug interactions of nefazodone as well as to determine its place among currently available antidepressants. DATA SOURCES: A search of European and American literature using EMBASE and MEDLINE was completed. Nefazodone was the search term. DATA SYNTHESIS: Nefazodone is an antidepressant that blocks serotonin type 2 (5-HT2) receptors in addition to inhibiting the reuptake of serotonin and norepinephrine. In double-blind, placebo-controlled studies, nefazodone demonstrates antidepressant activity at dosages

Nefazodone in the treatment of premenstrual syndrome: a preliminary study.

Nefazodone, a new phenylpiperazine antidepressant agent with serotonin type 2 antagonism and serotonin reuptake inhibition, was evaluated in two patient groups to determine its effectiveness in reducing the symptoms of premenstrual syndrome (PMS). The two studied groups were PMS patients with no coexisting major depression or dysthymia (N = 23) and PMS patients with current major depression or dysthymia, termed the premenstrual exacerbation group (N = 24). The two patient groups received open-label nefazodone for 8 weeks, with optional maintenance at the same dose for up to 1 year. The initial

In vitro activity of OPC-17116.

The in vitro activity of OPC-17116, a new C-5 methyl fluoroquinolone, was compared with the activities of other fluoroquinolones. OPC-17116 inhibited 50% of the members of the family Enterobacteriaceae tested and 90% of Haemophilus influenzae, Neisseria species, and Moraxella catarrhalis isolates at less than or equal to 0.25 microgram/ml. At less than or equal to 2 micrograms/ml, 90% of the Enterobacteriaceae were inhibited, which was comparable to or better than the activities of fleroxacin, ofloxacin, and lomefloxacin but less than the activity of ciprofloxacin. OPC-17116 inhibited 90% of t

Effect of almitrine on renal sodium excretion in chemoreceptor denervated spontaneously hypertensive rats.

The effect of almitrine on urinary sodium excretion was investigated in conscious carotid body-, sino-aortic- and sham-denervated SHR. The animals were kept singly in metabolic cages and had free access to food and tap water. Oral application of almitrine (0.5 mg/kg bwt) during five days induced a twofold natriuresis in sham-operated rats. The first natriuretic response on the fourth day of almitrine treatment was observed in animals of all groups indicating that this effect is not chemoreceptor mediated. The second natriuresis seen on the sixth day of the post-treatment period in both sino-ao

Esaprazole in the treatment of gastric ulcer: preliminary clinical trial.

The activity and tolerability of esaprazole administered once a day at doses of 900 and 1350 mg/day in the treatment of gastric ulcer were evaluated in a preliminary study carried out in 47 patients. The results failed to show any difference between the two doses in terms of percentage of ulcer healing after 4 weeks (43% and 45% respectively) or 8 weeks (86% and 82%), or in the course of symptoms and signs, which improved rapidly. The safety of esaprazole was good, no modifications of laboratory parameters being observed. Although the study involved only a small number of patients, the finding

Effects of the two new anthelmintic agents CGP 6140 and CGP 20376 on adult Paragonimus uterobilateralis in the experimental host Sigmodon hispidus.

The new compounds CGP 6140 and CGP 20376 were tested for their anthelmintic activity against adult Paragonimus uterobilateralis in experimentally infected Sigmodon hispidus. Both drugs were effective at a dosage of 2 x 100 mg/kg b.w. given orally with an interval of six hours between the doses. This regimen killed 80% of the flukes collected on Days 7 and 11 after treatment with CGP 6140 and 100% of the flukes collected on Days 5, 7, and 11 after therapy with CGP 20376. Both compounds showed no effect at single doses of 25, 50 and 100 mg/kg or with 12.5 and 25 mg/kg/day given on four consecuti

Ventilatory effects of almitrine bismesylate in congenital central hypoventilation syndrome.

Patients with congenital central hypoventilation syndrome (CCHS) lack hypercapnic and hypoxic stimulation of ventilation but have demonstrated carotid body function in response to hyperoxia and to pharmacological stimulation with doxapram. This study investigated the ventilatory effects of almitrine bismesylate, a carotid body stimulant, in 12 patients with CCHS. Measurements of minute ventilation, tidal volume (VT), respiratory rate (RR) and transcutaneous PO2 (TCPO2) were taken before and after administration of 4.5 mg/kg and 6 mg/kg of almitrine. Twenty-four hour pharmacokinetic studies wer

[Histydyl-proline diketopiperazine (HPD), a metabolite of thyrotropin-releasing hormone (TRH), improves the ataxic gait in 3-acetylpyridine (3-AP) treated rats].

Histidyl-proline diketopiperazine (HPD) is the active metabolite deriving from thyrotropin-releasing hormone (TRH). However, its regional distribution in the central nervous system is quite different from TRH. Above all, it is to be noted that cerebellum is one of the richest region of HPD. In rat cerebellum, for example, the concentration of HPD is 20 times higher than that of TRH. This indicates the probable action of HPD within cerebellar system. Recently, a fascinating hypothesis has been proposed by Prasad et al that some activities of TRH are based on its conversion to HPD. The purpose o
