Glucocorticoids suppress early lung inflammation and impair control of SARS-CoV-2 in non-human primates.

In severe cases of COVID-19, glucocorticoid treatment improves clinical outcomes. However, in non-hospitalized patients, glucocorticoids have limited benefit and may impair viral clearance. Here, we used the rhesus macaque model of acute SARS-CoV-2 infection to investigate the impact of glucocorticoids on host responses and viral control in a setting of mild disease. Rhesus macaques were pre-treated with intravenous methylprednisolone for 5 days prior to SARS-CoV-2 (Delta) infection and maintained on a daily oral prednisolone until necropsy at day 13 post infection. Glucocorticoid (GC) treatme

Zoonotic and Anthroponotic Plasmodium spp. Circulation between Wild Primates and Indigenous Community, Peruvian Amazon, 2007-2020.

Malaria transmission at the human-wildlife interface remains poorly characterized in the Amazon. We conducted a molecular survey of Plasmodium spp. in an Indigenous community (n = 141) and sympatric nonhuman primates (NHPs) (n = 341; 10 species) in the Peruvian Amazon during 2007-2020. By using nested or quantitative PCR (targeting cytb, cox3, and 18S rRNA genes) and sequencing, we estimated prevalence, parasite load, and genetic similarity. We detected Plasmodium in 43.3% of humans and 51.9% of NHPs. P. vivax/simium predominated in humans (42.1%), whereas P. brasilianum/malariae predominated …

Surveillance Service of Yellow Fever in Non-Human Primates in the Federal District, Brazil, 2008-2022.

BACKGROUND: Surveillance of non-human primate (NHP) deaths is vital for the early detection of yellow fever (YF) and prevention of its spread to the human population. This study assessed the YF surveillance system for NHPs in the Brazilian Federal District (FD) from 2008 to 2022. METHODS: A retrospective analysis of the aggregated data from 15 years of outbreak surveillance involving NHP deaths was conducted. The analyzed variables included spatiotemporal distribution, species, sex, age, sample collection, cause of death, and YF test results. RESULTS: In total, 1,175 outbreaks involving 1,35

Rapid and robust immune response boosting with potent, next-generation adjuvant in viral vector-primed primates.

To identify strategies for augmenting vaccine immunogenicity, we compared a pox-protein prime-boost regimen comprising recombinant modified vaccinia virus Ankara and multimeric HIV-1 Env gp145, adjuvanted with Army Liposomal Formulation (ALF) either adsorbed to aluminum salt (ALFA) or formulated with the QS-21 saponin (ALFQ), in rhesus macaques. ALFQ promoted greater magnitude and more durable humoral and cellular immune responses than ALFA, which exhibits similar immunogenicity to aluminum-based adjuvants. Peak Env-specific CD4+ T cell responses assessed by intracellular cytokine staining wer

Molecular evidence of host specificity in Buxtonella-like ciliates in mandrills (Mandrillus sphinx) and related primates.

Balantioides coli (formerly Balantidium coli) is a ciliate protozoan causing balantidiasis in humans and non-human primates (NHPs), yet it remains underrecognized in zoological settings. Morphologically similar ciliates described as Buxtonella-like organisms have been reported in NHPs and are generally considered asymptomatic. In this study, we report symptomatic infection in mandrills (Mandrillus sphinx) and investigate the prevalence and genetic diversity of Balantioides spp. and Buxtonella-like ciliates across multiple NHP species housed in Polish zoological gardens. Faecal samples were col…

Strategies to decipher neuron identity from extracellular recordings in the cerebellum of behaving non-human primates.

Identification of neuron type is critical to understand computation in neural circuits through extracellular recordings in awake, behaving animal subjects. Yet, modern recording probes have limited power to resolve neuron type. Here, we leverage the well-characterized architecture of the cerebellar circuit to perform expert identification of neuron type from extracellular recordings in behaving non-human primates. Using deep-learning classifiers we evaluate the information contained in readily accessible extracellular features for neuron identification. Waveform, discharge statistics, anatomic…

Immune-driven gene expression loss following intramuscular AAV delivery to non-human primates is only transient.

Recombinant adeno-associated virus (rAAV) vectors stand out as highly promising for in vivo gene transfer, particularly in targeting the skeletal muscle for treating muscular genetic diseases or secreting therapeutic factors. Despite the simplicity and efficacy of the established intramuscular (IM) route, it has been often associated with an immune-induced rapid loss of transgene expression, in particular in large animal models, and generally considered irreversible as a consequence of a cytotoxic elimination of transduced cells. Here, we report in a non-human primate model that transgene expr

The morphology of the oval window in Paranthropus robustus compared to humans and other modern primates.

The oval window (OW) is an opening connecting the inner and middle ear. Its area has been shown to consistently scale with body mass (BM) in primates, and has been used alongside semi-circular canal (SCC) size to differentiate Homo sapiens and fossil hominins, including Paranthropus robustus. However, while the morphology of other inner ear elements, such as cochlea and SCCs, has been extensively studied in primates, OW shape has received little attention. In this study, we assess OW morphological variability in extant primates, and compare P. robustus to extant hominids. The potential of OW s…

Peptide-Bound Aflibercept Eye Drops for Treatment of Neovascular Age-Related Macular Degeneration in Nonhuman Primates.

The advent of biomacromolecules antagonizing vascular endothelial growth factor (VEGF) has revolutionized the treatment of neovascular age-related macular degeneration (nAMD). However, frequent intravitreal injections of these biomacromolecules impose an enormous burden on patients and create a massive workload for healthcare providers. This causes patients to abandon therapy, ultimately leading to progressive and irreversible vision loss. In order to address this unmet clinical need, a noninvasive treatment for nAMD is developed. An optimized cell-penetrating peptide derivative, bxyPenetratin

Suppression of the neutrophil-derived S100A8/A9 complex ameliorates doxorubicin-induced cardiomyopathy in non-human primates.

Doxorubicin (DOX) is used for treatment of various cancers worldwide, but has a cumulative dose-dependent risk of irreversible cardiomyopathy. The mechanism of DOX-induced cardiomyopathy has not been fully elucidated because animal models such as rodents used in previous studies are largely different from humans in their physiological characteristics. In this study, we generated a DOX-induced cardiomyopathy model using non-human primates, cynomolgus monkeys, which are closely related to humans. After DOX was intravenously injected into the monkeys using the same protocol as that for humans, ca…

Pharmacokinetic and Metabolomic Studies with BBT-059 in Nonhuman Primates Exposed to Total-Body Gamma Radiation.

BBT-059 is a long-acting PEGylated interleukin-11 analog that has been shown to have hematopoiesis-promoting and anti-apoptotic attributes, and is being studied as a radiation countermeasure for the hematopoietic acute radiation syndrome (H-ARS). This potential countermeasure has been demonstrated to enhance survival in irradiated mice. To investigate the toxicity and safety profile of this agent, 14 nonhuman primates (NHPs, rhesus macaques) were administered two different doses of BBT-059 subcutaneously 24 h after 4 Gy total-body irradiation and were monitored for the next 60 days postirradia…

Cardiac allograft tolerance can be achieved in nonhuman primates by donor bone marrow and kidney cotransplantation.

Long-term, immunosuppression-free allograft survival has been induced in human and nonhuman primate (NHP) kidney recipients after nonmyeloablative conditioning and donor bone marrow transplantation (DBMT), resulting in transient mixed hematopoietic chimerism. However, the same strategy has consistently failed in NHP heart transplant recipients. Here, we investigated whether long-term heart allograft survival could be achieved by cotransplanting kidneys from the same donor. Cynomolgus monkeys were transplanted with heart allografts alone or heart and kidney allografts from the same major histoc

Edonerpic maleate enhances functional recovery from spinal cord injury with cortical reorganization in non-human primates.

While spinal cord injury (SCI) aggravates the quality of life in humans by severe paralysis, clinical intervention to promote functional recovery from SCI is limited. We recently identified a small compound, edonerpic maleate (edonerpic MA), which accelerates training-dependent motor functional recovery from brain damage in rodents (cryo-genic cortical injury) and non-human primates (internal capsule haemorrhage) by the facilitation of experience-dependent synaptic trafficking of glutamate α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors. In the present study, we investigated whe…

Cell Type Distribution of Intrathecal Antisense Oligonucleotide Activity in Deep Brain Regions of Non-Human Primates.

Intrathecally administered RNase H1-active gapmer antisense oligonucleotides (ASOs) are promising therapeutics for brain diseases where lowering the expression of one target gene is expected to be therapeutically beneficial. Such ASOs are active, to varying degrees, across most or all cell types in the cortex and cerebellum of mouse and non-human primate (NHP) brain regions with substantial drug accumulation. Intrathecally delivered ASOs, however, exhibit a gradient of exposure across the brain, with more limited drug accumulation and weaker target engagement in deep brain regions of NHP. Here

Enhanced Costimulation Blockade With αCD154, αCD2, and αCD28 to Promote Heart Allograft Tolerance in Nonhuman Primates.

BACKGROUND: Long-term renal allograft acceptance has been achieved in macaques using a transient mixed hematopoetic chimerism protocol, but similar regimens have proven unsuccessful in heart allograft recipients unless a kidney transplant was performed simultaneously. Here, we test whether a modified protocol based on targeting CD154, CD2, and CD28 is sufficient to prolong heart allograft acceptance or promote the expansion of regulatory T cells. METHODS: Eight macaques underwent heterotopic allo-heart transplantation from major histocompatibility complex-mismatched donors. Induction treatmen

Assessing concordance between RNA-Seq and NanoString technologies in Ebola-infected nonhuman primates using machine learning.

This study evaluates the concordance between RNA sequencing (RNA-Seq) and NanoString technologies for gene expression analysis in non-human primates (NHPs) infected with Ebola virus (EBOV). A detailed comparison of both platforms revealed a strong correlation, with Spearman coefficients for 56 out of 62 samples ranging from 0.78 to 0.88. The mean and median coefficients were 0.83 and 0.85, respectively. Bland-Altman analysis confirmed high consistency across most measurements, with values falling within the 95% limits of agreement. Using a machine learning approach with the Supervised Magnitud…

High infection prevalences by Trypanosoma minasense in non-human primates from the Southeast region of Brazil.

Non-human primates (NHPs) are reported as exclusive hosts of Trypanosoma minasense, whose transmission cycle remains unknown. We investigated trypanosomatid infections in 194 NHPs of nine species from the Brazilian Southeast region. We evaluated the impact of landscape composition and forest fragmentation on T. minasense infection. In the Atlantic Forest areas, Sapajus nigritus presented a higher positivity for T. minasense on the blood slides (60.9%) compared to Callithrix spp. (24.6%) and Alouatta guariba clamitans (20.7%); molecular analysis revealed higher infection prevalences in S. nigri…

Novel insight into the genetic diversity of strongylid nematodes infecting South-East and East Asian primates.

With many non-human primates (NHPs) showing continued population decline, there is an ongoing need to better understand their ecology and conservation threats. One such threat is the risk of disease, with various bacterial, viral and parasitic infections previously reported to have damaging consequences for NHP hosts. Strongylid nematodes are one of the most commonly reported parasitic infections in NHPs. Current knowledge of NHP strongylid infections is restricted by their typical occurrence as mixed infections of multiple genera, which are indistinguishable through traditional microscopic ap…

Genomic insights into ESBL-producing Escherichia coli isolated from non-human primates in the Peruvian Amazon.

INTRODUCTION: Extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae are on the WHO priority pathogens list because they are associated with high mortality, health-care burden, and antimicrobial resistance (AMR), a serious problem that threatens global public health and should be addressed through the One Health approach. Non-human primates (NHP) have a high risk of acquiring these antibiotic-resistant bacteria due to their close phylogenetic relationship with humans and increased anthropogenic activities in their natural environments. This study aimed to detect and analyze the g…
