Intervertebral disc puncture predisposes the disc to Cutibacterium acnes infection via hematogenous and contiguous routes: a rat model study.

BACKGROUND: Intervertebral disc degeneration (IVDD) is a primary cause of chronic low back pain. Recent evidence suggests a potential association between low-virulence infections, particularly by Cutibacterium acnes, and IVDD. However, the pathways and mechanisms by which this commensal skin bacterium infects intervertebral discs remain poorly understood. OBJECTIVE: The purpose of this study was to determine whether Cutibacterium acnes can enter the intervertebral disc through hematogenous or contiguous routes, and whether injury-induced damage to the annulus fibrosus and IVDD increases susce

Conservative laparoscopic approach to a perforated pyomyoma after uterine artery embolization.

We describe a conservative laparoscopic approach to treatment of a perforated pyomyoma after uterine artery embolization. A 36-year-old woman came to our emergency department 8 weeks after undergoing uterine artery embolization. She reported painful abdominal cramps. Physical examination revealed fever, tachycardia, hypotension, abdominal pain with rebound tenderness and pain during bimanual examination. Ultrasonography showed a fundal/subserosal leiomyoma and a moderate amount of fluid in the abdominal cavity. Because the patient desired preservation of the uterus, a laparoscopic approach was

Microbiology of chronic hyperplastic sinusitis.

BACKGROUND: Patients with chronic hyperplastic sinusitis (CHS) form a heterogeneous group with similar symptoms and similar treatment despite of possible different mechanisms behind the disease. In the present study we focused on the microbiological findings in CHS and compared these results to the patient history in order to find out a possible explanation for the aetiology and chronicity of CHS. METHODS: In 30 patients the sinus mucus was collected under endoscopic sinus surgery. Samples from 20 healthy volunteers were collected by nasal lavage. Eosinophil staining, bacterial culturing and

Effect of Corynebacterium parvum on the course of a Listeria monocytogenes infection in normal and congenitally athymic (nude) mice.

Spleen counts of Listeria monocytogenes in normal mice challenged with various numbers of organisms were generally lower in mice injected 3 days previously with killed Corynebacterium parvum. This increased resistance was unexpected since C. parvum generally inhibits T cell-mediated immunity, the functional immunity against Listeria being T cell-dependent. The inhibitory effect on T cells seemed to be compensated by the stimulatory effect of C. parvum on macrophages. Therefore, the experiments were repeated in congenitally athymic (nude) mice (B10LP) lacking functional T cells. C. parvum pretr

Changes in oral health during aging in a novel non-human primate model.

Oral health plays a significant role in the quality of life and overall well-being of the aging population. However, age-related changes in oral health are not well understood due to challenges with current animal models. In this study, we analyzed the oral health and microbiota of a short-lived non-human primate (i.e., marmoset), as a step towards establishing a surrogate for studying the changes that occur in oral health during human aging. We investigated the oral health of marmosets using cadaveric tissues in three different cohorts: young (aged ≤6 years), middle-aged, and older (>10 years

Development of an Effective Acne Treatment Based on CBD and Herbal Extracts: Preliminary In Vitro, Ex Vivo, and Clinical Evaluation.

Acne vulgaris, the most common form of acne, is characterized by a mixed eruption of inflammatory and noninflammatory skin lesions primarily affecting the face, upper arms, and trunk. The pathogenesis of acne is multifactorial and includes abnormal keratinization and plugging of the hair follicles, increased sebum production, proliferation and activation of Cutibacterium acnes (C. acnes; formerly Propionibacterium acnes, P. acnes), and finally inflammation. Recent studies have found that cannabidiol (CBD) may be beneficial in the treatment of acne. The aim of this study was to explore natural

[Doxycycline].

Doxycyclin is a semi-synthetic structural isomer of the tetracycline family. It exhibits good intra-cellular penetration, with bacteriostatic activity on many bacteria. Different types or bacterial resistance are known. Acquired resistance has a ribosomal or a plasmidic mechanism. Resistance of Propionibacterium acnes is secondary to a mutation of ARNr. Doxycyclin also has an anti-inflammatory activity, via numerous pathways. Doxycyclin is rapidly and almost completely absorbed by the digestive tract. Food has no incidence on the absorption. It has a high but labile affinity for proteins with

The role of probiotic cultures in the control of gastrointestinal health.

The use of probiotics to enhance intestinal health has been proposed for many years. Probiotics are traditionally defined as viable microorganisms that have a beneficial effect in the prevention and treatment of specific pathologic conditions when they are ingested. There is a relatively large volume of literature that supports the use of probiotics to prevent or treat intestinal disorders. However, the scientific basis of probiotic use has been firmly established only recently, and sound clinical studies have begun to be published. Currently, the best-studied probiotics are the lactic acid ba

The biosynthetic gene cluster landscape of the oral microbiome across health and dental caries.

Specialized metabolites encoded by biosynthetic gene clusters (BGCs) in the oral microbiome remain largely unexplored in the context of oral health and disease. Previous genome-centric surveys have identified hundreds of uncharacterized BGCs in the oral cavity associated with health and disease, but these studies relied on reference genomes and did not capture strain-level variation or the native distribution of BGCs. Here, we assembled three independently sourced metagenomic datasets from healthy and dental caries samples, extracted BGCs and quantified their metagenomic abundance and transcri

Dual-frequency AMCW concurrent ranging and velocimetry for LiDAR imaging.

BACKGROUND: α-Mangostin (α-M) is a natural antimicrobial and anti-inflammatory compound with promising anti-acne potential; however, its poor solubility and instability limit its topical use. This study developed and evaluated chitosan-alginate hydrogel films incorporating nanoencapsulated α-M (HF α-M NPs) to enhance stability, skin penetration, and therapeutic efficacy against Propionibacterium acnes. METHODS: α-M NPs were produced by ionic gelation using chitosan and sodium tripolyphosphate, followed by alginate coating, and subsequently incorporated into chitosan-alginate hydrogel films. N

An antibody to IL-1 receptor 7 protects mice from LPS-induced tissue and systemic inflammation.

INTRODUCTION: Interleukin-18 (IL-18), a pro-inflammatory cytokine belonging to the IL-1 Family, is a key mediator ofautoinflammatory diseases associated with the development of macrophage activation syndrome (MAS).High levels of IL-18 correlate with MAS and COVID-19 severity and mortality, particularly in COVID-19patients with MAS. As an inflammation inducer, IL-18 binds its receptor IL-1 Receptor 5 (IL-1R5), leadingto the recruitment of the co-receptor, IL-1 Receptor 7 (IL-1R7). This heterotrimeric complex subsequentlyinitiates downstream signaling, resulting in local and systemic inflammatio

Unveiling the skin microbial guardians and assailants in psoriasis subtypes: a Mendelian randomization study.

Previous studies have shown an association between skin microbiota and psoriasis, but the causal relationship between skin microbiota and different psoriasis subtypes remains unexplored. Our study employed the Mendelian randomization (MR) method, using summary statistics from Genome-Wide Association Studies, to investigate the causal relationships between skin microbiota and different subtypes of psoriasis. The inverse variance weighted (IVW) method served as the primary method in our MR study. The MR-Egger intercept test, Cochran's Q test, MR-PRESSO test, and leave-one-out analysis were condu
