Identification and structural-functional analysis of cyclin-dependent kinases of the cattle tick Rhipicephalus (Boophilus) microplus.

Cyclin-dependent kinases (CDKs) are a family of serine/threonine kinases essential for cell cycle progression. Herein, we describe the participation of CDKs in the physiology of Rhipicephalus microplus, the southern cattle tick and an important disease vector. Firstly, amino acid sequences homologous with CDKs of other organisms were identified from a R. microplus transcriptome database in silico. The analysis of the deduced amino acid sequences of CDK1 and CDK10 from R. microplus showed that both have caspase-3/7 cleavage motifs despite their differences in motif position and length of encode

Molecular identification of Rickettsia spp. in chigger mites in Taiwan.

The genus Rickettsia is the causative agent of several rickettsial diseases that are primarily transmitted by hard ticks. The occurrence of Rickettsia in chigger mites, which are vectors of scrub typhus in the western Pacific region, has been infrequently investigated. We identified Rickettsia spp. in chiggers collected from small mammals in six counties of Taiwan. Moreover, by capitalising on parallel Rickettsia detections on small mammals and their infested ticks and fleas, we were able to identify Rickettsia spp. that suggested more intimate associations with chigger mites. Rickettsia detec

First molecular evidence of Borrelia burgdorferi sensu lato in goats, sheep, cattle and camels in Tunisia.

Borrelia burgdorferi sensu lato (s.l.) are tick-transmitted spirochaetes of veterinary and human importance. Molecular epidemiology data on ruminants are still lacking in most countries of the world. Therefore, the aim of this study was to estimate the rate of B. burgdorferi s.l. infection in ruminants from Tunisia. A total of 1,021 ruminants (303 goats, 260 sheep, 232 cattle and 226 camels) from different bioclimatic areas in Tunisia were investigated for the presence of B. burgdorferi s.l. DNA in blood by real time PCR. Prevalence rates were 30.4% (92/303) in goats, 6.2% (16/260) in sheep, 1

Seroprevalence estimation and management factors associated with high herd seropositivity for Babesia bovis in commercial dairy farms of Puerto Rico.

A cross-sectional study was conducted to determine individual cow seroprevalence of Babesia bovis in adult lactating dairy cattle of Puerto Rico (PR), to assess the associations of farm management factors on herd seroprevalence, and to document the species of ticks infesting cattle within these farms. Antibody activity against B. bovis was determined using an indirect fluorescent antibody test (IFAT). Serum samples were obtained from 2,414 adult lactating dairy cattle from 76 randomly selected commercial dairy farms. Herd seroprevalence ranged from 0 to 51% with an overall individual cow serop

Control of immature Ixodes scapularis (Acari: Ixodidae) on rodent reservoirs of Borrelia burgdorferi in a residential community of southeastern Connecticut.

A 3-yr community-based study was conducted on residential properties on Mason's Island, Mystic, CT, to determine the efficacy of a rodent-targeted acaricide (fipronil) to control immature Ixodes scapularis (Say) on Peromyscus leucopus. Results indicated that modified commercial bait boxes were effective as an acaricide delivery method for reducing nymphal and larval tick infestations on white-footed mice by 68 and 84%, respectively. Passive application of fipronil significantly reduced the infection rate of Borrelia burgdorferi among white-footed mice by 53%. Moreover, the abundance of questin…

Reciprocal regulation of NF-kB (Relish) and Subolesin in the tick vector, Ixodes scapularis.

BACKGROUND: Tick Subolesin and its ortholog in insects and vertebrates, Akirin, have been suggested to play a role in the immune response through regulation of nuclear factor-kappa B (NF-kB)-dependent and independent gene expression via interaction with intermediate proteins that interact with NF-kB and other regulatory proteins, bind DNA or remodel chromatin to regulate gene expression. The objective of this study was to characterize the structure and regulation of subolesin in Ixodes scapularis. I. scapularis is a vector of emerging pathogens such as Borrelia burgdorferi, Anaplasma phagocyto

A lifelong study of a pack Rhodesian ridgeback dogs reveals subclinical and clinical tick-borne Anaplasma phagocytophilum infections with possible reinfection or persistence.

BACKGROUND: Various tick-borne infections often occur without specific clinical signs and are therefore notoriously hard to diagnose separately in veterinary practice. Longitudinal studies over multiple tick seasons performing clinical, serological and molecular investigations in parallel, may elucidate the relationship between infection and disease. In this regard, six related Rhodesian Ridgeback dogs living as a pack became subject of lifetime studies due to ongoing tick infestations and recurring clinical problems. Blood samples for diagnostic tests were obtained throughout the years 2000 t…

Influence of hosts on the oviposition and hatching of engorged females Amblyomma cajennense sensu stricto (Acari: Ixodidae) with notes on new host records.

The parasite-host relationships of Amblyomma cajennense sensu stricto, one of the species of the Amblyomma cajennense complex that occurs in the state of Maranhão, are poorly known and are restricted to its association with horses and humans. This study evaluated, for the first time, the effect of hosts on the oviposition of A. cajennense engorged females, naturally collected from wild animals and humans, as well as those experimentally fed on cattle and highlights the importance of the relationship between A. cajennense and different hosts in maintaining tick populations in the Amazon biome.

Ornithodoros hermsi.

Ornithodoros hermsi is a small, multi-host soft tick that feeds rapidly on blood in all life stages, including larva, 2 to 5 nymphal instars (primarily 3), and male and female. As adults, they can feed repeatedly and reproduce for many years. O. hermsi is nidicolous and primarily inhabits nest cavities and burrows of small rodents and birds, with chipmunks and tree squirrels serving as its primary hosts. When no hosts are available, they can fast for several years. O. hermsi is found in restricted habitats where their hosts live in coniferous pine and Douglass fir forests at elevations of 1,50

[Occurrence of Francicella tularensis subsp. holarctica in haematophagous arthropods in the Breclav district in 2022].

Tularemia is a zoonosis caused by Francisella tularensis, a gram-negative aerobic bacterium belonging to the class of Gammaproteobacteria and the family Francisellaceae. Despite its undeniable importance for human health, there is little data on the current distribution of F. tularensis in various hematophagous arthropods. The aim of this study was to perform a mass molecular screening of different possible hematophagous vectors: ticks (4348 ticks of the species Ixodes ricinus, Dermacentor reticulatus, and Haemaphysalis concinna), mosquitoes (4100 specimens of Aedes vexans), and blackflies (69

Epidemiology of bovine anaplasmosis and babesiosis in Latin America and the Caribbean.

The various parameters which interact in the epizootiology of babesiosis and anaplasmosis and which it is necessary to obtain in a survey are analysed and the two diseases compared. Where no data are available from Latin America and the Caribbean, the references from Australia and the United States of America are discussed. The two principal objectives of a study on the epidemiology of babesiosis and anaplasmosis are: to determine the risk of occurrence to understand the relevant factors leading to outbreaks. The first objective is relatively simple to achieve for both diseases by calculating

Crimean-Congo Haemorrhagic Fever Virus Exposure in Horses and Donkeys in Border Livestock Markets of Nigeria.

INTRODUCTION: Crimean-Congo haemorrhagic fever virus (CCHFV) is a tick-borne zoonotic pathogen of major public health importance in Africa. Although CCHFV exposure has been reported in several livestock species in Nigeria, data on equids remain limited despite their extensive involvement in livestock movement and trade. This study assessed CCHFV seroprevalence in horses and donkeys in major international livestock markets in northern Nigeria and examined factors associated with exposure. METHODS: From June 2023 to July 2024, a longitudinal sero-surveillance study was conducted in three high-v

Biotin-avidin amplified ELISA for detection of antibodies to Sarcoptes scabiei in chamois (Rupicapra spp.).

Scabies is a major threat to the well being of mountain-dwelling Bovid hosts, Rupicapra rupicapra and Rupicapra pyrenaica. Severe outbreaks are in progress over a significant part of their distribution area and resource managers demand improved methods to monitor, analyse and possibly forecast the spread and effects of scabies at the population level. An amplified capture enzyme-linked immunosorbent assay was developed to detect antibodies to Sarcoptes scabiei in chamois (Rupicapra spp.) serum. The method used the biotin-avidin amplification system and was validated on a panel of 144 serum sam

Interspecies co-feeding transmission of Powassan virus between a native tick, Ixodes scapularis, and the invasive East Asian tick, Haemaphysalis longicornis.

BACKGROUND: Powassan virus, a North American tick-borne flavivirus, can cause severe neuroinvasive disease in humans. While Ixodes scapularis are the primary vectors of Powassan virus lineage II (POWV II), also known as deer tick virus, recent laboratory vector competence studies showed that other genera of ticks can horizontally and vertically transmit POWV II. One such tick is the Haemaphysalis longicornis, an invasive species from East Asia that recently established populations in the eastern USA and already shares overlapping geographic range with native vector species such as I. scapulari
