Integrated assessment of differentially expressed plasma microRNAs in subtypes of nonsyndromic orofacial clefts.

BACKGROUND: Orofacial clefts include cleft lip only (CLO), cleft palate only (CPO), and cleft lip with palate (CLP). Previously, we reported the expression profile of plasma microRNAs in CLO, CPO, and CLP, respectively. However, the interaction of each subtype remains poorly investigated. METHODS: In this study, we integrated the expression profiles of plasma miRNAs in these 3 subtypes, and assessed the distinct and overlapping dysregulated miRNAs using Venn diagrams. Their respective target genes reported in the literature were further analyzed using pathway analysis. RESULTS AND CONCLUSION

Maize Cytolines Unmask Key Nuclear Genes That Are under the Control of Retrograde Signaling Pathways in Plants.

The genomes of the two plant organelles encode for a relatively small number of proteins. Thus, nuclear genes encode the vast majority of their proteome. Organelle-to-nucleus communication takes place through retrograde signaling (RS) pathways. Signals relayed through RS pathways have an impact on nuclear gene expression but their target-genes remain elusive in a normal state of the cell (considering that only mutants and stress have been used so far). Here, we use maize cytolines as an alternative. The nucleus of a donor line was transferred into two other cytoplasmic environments through at

Strigolactones regulate protonema branching and act as a quorum sensing-like signal in the moss Physcomitrella patens.

Strigolactones are a novel class of plant hormones controlling shoot branching in seed plants. They also signal host root proximity during symbiotic and parasitic interactions. To gain a better understanding of the origin of strigolactone functions, we characterised a moss mutant strongly affected in strigolactone biosynthesis following deletion of the CAROTENOID CLEAVAGE DIOXYGENASE 8 (CCD8) gene. Here, we show that wild-type Physcomitrella patens produces and releases strigolactones into the medium where they control branching of protonemal filaments and colony extension. We further show tha

Novel ideas of gene therapy for atherosclerosis: modulation of cellular signal transduction of TGF-beta family.

Atherosclerosis is a disease characterized by accumulation of lipids and fibrous elements in the innermost layer of the arterial wall. An asymptomatic atherosclerotic plaque is characterized by a lipid core, composed of modified lipids, macrophages and T cells, which were separated from the lumen vessel by a thick fibrous cap, composed of vascular smooth muscle cell-secreted solid collagen matrix. Recently, it has been reported that expressions of TGF-beta family were up regulated in human atherosclerotic plaques. In addition TGF-beta family seems to plays pivotal roles for the development of

Update on MALT lymphomas.

Gastric mucosa associated lymphoid tissue (MALT) lymphoma is a histologically distinct tumour derived from MALT acquired as a result of Helicobacter pylori infection. Eradication of H. pylori causes clinical regression of the lymphoma in 75% of cases. In seeking to identify those cases resistant to this therapy, and in the interests of further understanding the biology of MALT lymphoma, genetic alterations of MALT lymphomas have been investigated. Three translocations, t(11;18)(q21;q21), t(1;14)(p22;q32) and t(14;18)(q32;q21) are specifically associated with MALT lymphoma and the genes involve

Genetically engineered recombinant adenovirus expressing interleukin‑2 for hepatocellular carcinoma therapy.

Regulatory and effector T cells possess immunological cytotoxicity for tumor cells in the tumor microenvironment during tumor progression and are the primary suppressors inhuman cancer therapy. Interleukin‑2 (IL‑2) is an anticancer cytokine, which triggers human innate and adaptive immunity by stimulating T cell propagation and lymphocyte infiltration into tumor sites. IL‑2 has been used successfully for cancer therapy. Recombinant adenovirus expressing IL‑2 (rAd‑IL‑2) injection is a gene therapy agent that may improve prognosis of hepatocellular carcinoma (HCC) patients. In the present study,

Treatment of the mouse model of mucopolysaccharidosis type IIIB with lentiviral-NAGLU vector.

The Sanfilippo syndrome type B (mucopolysaccharidosis IIIB) is an autosomal recessive disorder due to mutations in the gene encoding NAGLU (alpha-N-acetylglucosaminidase), one of the enzymes required for the degradation of the GAG (glycosaminoglycan) heparan sulphate. No therapy exists for affected patients. We have shown previously the efficacy of lentiviral-NAGLU-mediated gene transfer in correcting in vitro the defect on fibroblasts of patients. In the present study, we tested the therapy in vivo on a knockout mouse model using intravenous injections. Mice (8-10 weeks old) were injected wit

Macrophage plasticity in thyroid eye disease: dual effects of inflammation and fibrosis.

Thyroid Eye Disease (TED), also known as Graves' ophthalmopathy, is an organ-specific inflammatory disorder associated with autoimmune thyroid dysfunction. Its primary pathological features include immune cell infiltration of orbital tissues, fat hyperplasia, and fibrotic remodeling. The pathogenesis centers on abnormal expression of TSHR on orbital fibroblasts and immune attacks mediated by autoantibodies. Recent studies increasingly reveal that infiltrating immune cells, particularly highly plastic macrophages, do not simply divide into static M1/M2 phenotypes. Instead, they exist within a f

Hesperidin and Hesperetin: Epigenetic-Stemness Crosstalk, Antitumor Mechanisms, Preclinical Data and Translation Barriers.

Hesperidin is a natural flavonoid derived from citrus plants, which can be hydrolyzed into hesperetin in vivo. Both compounds have anti-inflammatory, antioxidant and antitumor activities. At present, there is a lack of reviews focusing on the epigenetic regulation of cancer stem cells (CSCs) mediated by hesperidin and hesperetin. This review summarizes the molecular crosstalk between hesperidin/hesperetin and CSCs mediated via three major epigenetic pathways, including direct regulatory effects, indirect modulatory actions, and mechanistic relationships proposed based on scientific hypotheses.

The hormetic landscape: from evolutionary origination to mechanistic pathways in therapeutics.

Hormesis is a biphasic dose-response, where mild stress triggers adaptive defense responses that increase cellular resilience. The hormesis is now recognized as a core concept with applications in toxicology, metabolism, aging, and evolutionary biology. A historical review, evolutionary explanations, and recent molecular insights led to the concept of hormesis as an anticipatory, "predict-and-prepare" strategy that allows organisms to adapt to changing conditions are addressed. The mitohormesis, autophagy, xenohormesis, epigenetic remodeling, and their interdependence with signaling pathways (

Molecular Mechanisms of APOL1-Associated Kidney Disease.

The discovery of apolipoprotein L1 (APOL1) risk polymorphisms has significantly changed our knowledge of kidney disease susceptibility and development in African American populations. Several non-diabetic kidney disorders, such as focal segmental glomerulosclerosis (FSGS), collapsing glomerulopathy, HIV-associated nephropathy (HIVAN), and accelerated chronic kidney disease (CKD) development, are significantly more likely to occur in people with two coding variations, G1 and G2. The significance of context-dependent pathogenic processes is highlighted by the poor penetrance and remarkable pheno

TGF-β: The Molecular Mechanisms of Atherosclerosis - insights into SMAD Pathways and Gene Therapy Prospects.

Atherosclerosis, a leading cause of global morbidity and mortality, is characterized by plaque formation resulting from the accumulation of fibrous elements, lipids, and calcification in arteries, leading to complications such as ischemic stroke, coronary artery disease, and myocardial infarction. Traditional treatments primarily address symptoms but fail to target underlying causes, prompting exploration of novel approaches like gene therapy. The TGF-β family, encompassing TGF-β1, TGF-β2, and TGF-β3, plays a critical role in cellular processes including proliferation, apoptosis, and migration

Molecular Mechanisms in Seborrheic Dermatitis-Systematic Review.

Seborrheic dermatitis (SD) is a chronic inflammatory skin disorder with a multifactorial pathogenesis involving immune dysregulation, oxidative stress, neuroendocrine signaling, and alterations of the epidermal barrier-lipid axis. Increasing molecular evidence indicates that SD is associated with both systemic and cutaneous abnormalities, including elevated β-endorphin levels, disturbed redox homeostasis, enhanced lipid peroxidation, dysregulated cytokine signaling, and genetic and epigenetic susceptibility factors. This systematic review was conducted in accordance with PRISMA guidelines. Com

Molecular requirements of pathogenic Th2 cell differentiation in allergic airway disease.

Th2 cells were originally described as a homogeneous population capable of simultaneously producing interleukin (IL)-4, IL-5, and IL-13, thereby playing a central role in allergic asthma and related conditions. Subsequent studies have revealed substantial heterogeneity within the Th2 lineage, with distinct subpopulations defined by unique surface markers and cytokine profiles. Of particular interest are pathogenic Th2 subsets, referred to as peTh2, Tpath2, or Th2A, that exhibit specialised effector functions and actively drive allergic disease. These pathogenic Th2 (pTh2) cells have been ident

Vascular dysfunction at the crossroads of gestational diabetes mellitus and cancer.

Gestational diabetes mellitus (GDM) and cancer develop in distinct clinical and biological contexts; however, both conditions expose tissues to chronic metabolic stress, hypoxia, inflammation, and vascular remodelling. In this review, we advance the hypothesis that GDM and cancer share conserved endothelial adaptive response mechanisms, particularly within the placental and tumour-associated vasculature, without implying malignant transformation or direct oncogenic causation. Common alterations in nitric oxide signalling, mitochondrial metabolic programming, intracellular pH regulation, and re

Molecular Basis of Impaired Decidualization in the Eutopic Endometrium of Endometriosis Patients.

Endometriosis is a chronic gynecological disorder characterized by the presence of endometrial tissue outside the uterine cavity. A common feature of this pathology is the impaired decidualization of endometrial stromal cells, a critical process that prepares the uterus for embryo implantation. This decidualization defect has been mechanistically linked to progesterone resistance in endometriotic lesions. However, the presence and underlying mechanisms of decidualization defects in the eutopic endometrium of women with endometriosis remain controversial. The aim of the present study is to inte

Characteristic analysis of N6-methyladenine in different parts of yak epididymis.

BACKGROUND: The epididymis is essential for sperm maturation. During sperm maturation, markable alterations of the payload of small noncoding RNAs are observed in the epididymis, which indicated the role of epigenetic alterations in sperm maturation. However, the N6-Methyladenosine (m6A) modification profile of the epididymis remains unelucidated. Therefore, in this study, we assessed the m6A modification levels in the caput, corpus, and cauda of the yak epididymis using a combination of methylated RNA immunoprecipitation and RNA sequencing. RESULTS: The m6A levels were significantly increase

Molecular Genetics of Acquired Temporal Lobe Epilepsy.

An epilepsy diagnosis reduces a patient's quality of life tremendously, and it is a fate shared by over 50 million people worldwide. Temporal lobe epilepsy (TLE) is largely considered a nongenetic or acquired form of epilepsy that develops in consequence of neuronal trauma by injury, malformations, inflammation, or a prolonged (febrile) seizure. Although extensive research has been conducted to understand the process of epileptogenesis, a therapeutic approach to stop its manifestation or to reliably cure the disease has yet to be developed. In this review, we briefly summarize the current lite

[Rare Autoimmune Diseases Role of Genetics - Example of Systemic Lupus Erythematosus].

Systemic lupus erythematosus (SLE) presents a complex clinical landscape with diverse manifestations, suggesting a multifactorial etiology. However, the identification of rare monogenic forms of the disease has shed light on specific genetic defects underlying SLE pathogenesis, offering valuable insights into its underlying mechanisms and clinical heterogeneity. By categorizing these monogenic forms based on the implicated signaling pathways, such as apoptotic body clearance, type I interferon signaling, JAK-STAT pathway dysregulation, innate immune receptor dysfunction and lymphocytic abnorma

A Review of Proposed Mechanisms in Rheumatoid Arthritis and Therapeutic Strategies for the Disease.

Rheumatoid arthritis (RA) is characterized by synovial edema, inflammation, bone and cartilage loss, and joint degradation. Patients experience swelling, stiffness, pain, limited joint movement, and decreased mobility as the condition worsens. RA treatment regimens often come with various side effects, including an increased risk of developing cancer and organ failure, potentially leading to mortality. However, researchers have proposed mechanistic hypotheses to explain the underlying causes of synovitis and joint damage in RA patients. This review article focuses on the role of synoviocytes a
