π-Delocalized Lipophilic Cations as New Candidates for Antimalarial, Antitrypanosomal and Antileishmanial Agents: Synthesis, Evaluation of Antiprotozoal Potency, and Insight into Their Action Mechanisms.

The search for new drugs that could treat tropical protozoan diseases, such as malaria or neglected tropical diseases (NTDs), motivates many medicinal chemists. New classes of antiprotozoal drugs that act through a novel mechanism of action must be developed. This review presents our efforts toward finding new candidate treatments for malaria, American trypanosomiasis, human African trypanosomiasis and leishmaniasis based on π-delocalized lipophilic cations (DLCs). DLCs, such as rhodacyanines, azarhodacyanines, β-carbolinium salts, and phenoxazinium salts, displayed strong antiprotozoal activi…

Determinants of disease phenotype in trypanosomatid parasites.

Trypanosomatid parasites infect over 21 million people worldwide, with a range of disease phenotypes. Trypanosoma cruzi causes American trypanosomiasis, wherein 30-40% of infected individuals develop disease manifestations, most commonly cardiomyopathy but also digestive megasyndromes. In the case of Trypanosoma brucei, the etiological agent of African trypanosomiasis, disease progression can be rapid or slow, with early or late central nervous system involvement. Finally, Leishmania species cause leishmaniasis, a disease that ranges from self-healing but scarring cutaneous lesions to fatal vi…

Effect of ivermectin on Trypanosoma brucei brucei in experimentally infected mice.

BACKGROUND & OBJECTIVES: Human and livestock African trypanosomiasis, otherwise known as sleeping sickness, is a neglected tropical disease of public health importance in west and central Africa. In view of the adverse side effects of the antitrypanosomal drugs, the relatively few side effects observed in ivermectin use, and because both onchocerciasis and typanosomiasis occur in overlapping foci in Africa, it would be desirable if the ivermectin that has been used successfully on onchocerciasis management could also be used in the control and treatment of trypanosomiasis. METHOD: In this stu…

A substance P antagonist, RP-67,580, ameliorates a mouse meningoencephalitic response to Trypanosoma brucei brucei.

Mice infected with the protozoan parasite Trypanosoma brucei brucei and treated subcuratively with the trypanocidal drug diminazene aceturate develop an acute inflammatory meningoencephalitis with associated astrocytic proliferation. This reaction is very similar to that seen in the fatal posttreatment reactive encephalopathies that can occur in human African trypanosomiasis. The 11-amino acid neuropeptide substance P (SP) has recently been identified as a mediator in many inflammatory responses, and the development of potent, highly specific, nonpeptide SP antagonists has provided a new oppor…

Cations in body fluids of Trypanosoma brucei in infected rabbits.

Serum copper, magnesium, zinc, calcium and ionized calcium (Ca++) concentrations were compared in 6 rabbits infected with Trypanosoma brucei … tion of hypercellularity in the glomeruli and tubular degeneration. Our findings thus indicate that trypanosomiasis causes kidney damage which may be responsible for the depletion of the cations seen in the study. Some of the clinical manifestations associated with African trypanosomiasis such as convulsions, anaemia, electrocardiographic changes and splenomegaly …

In Vitro Mechanism of Action of Acanthospermum hispidum in Trypanosoma brucei.

African trypanosomiasis is a major neglected tropical disease with significant health and economic concerns in sub-Saharan Africa. In the absence of vaccines for African trypanosomiasis, there is a consideration for alternative sources of chemotherapy. Acanthospermum hispidum DC (A. hispidum) is a herbal species of the Asteraceae family that is endowed with rich phytochemicals with unknown mechanisms of antitrypanosomal effects. This study aimed to investigate the cellular mechanisms of antitrypanosomal and antioxidant activities of A. hispidum against Trypanosoma brucei (T. brucei), a causati…

Parasitic diseases of the heart.

The following chapter is one of a series of chapters in the volume entitled Infections of the Myocardium appearing in Frontiers in Bioscience. The full table of contents can be found at http://www.bioscience.org/current/special/tanowitz.htm. In this chapter, we review several parasitic infections involving the myocardium and pericardium. The most widely studied parasitic infection affecting the heart is Chagas' disease or American trypanosomiasis. In this chapter we describe issues relating to Chagas' disease not covered in detail in other chapters. African trypanosomiasis may also cause a myo…

Genetic basis of trypanotolerance in cattle and mice.

Under most circumstances, certain breeds of domestic ruminants show a remarkable resistance to the effects of African trypanosomiasis: they can tolerate the presence of parasites while apparently controlling levels of parasitaemia and, crucially, not showing the severe anaemia and production loss that are characteristic of infection in susceptible hosts. As discussed here by Stephen Kemp and Alan Teale, the genetic control of this phenomenon might finally be yielding to gene mapping studies. Genetic regions determining susceptibility to trypanosomiasis in mice have been identified and parallel…

[Electroencephalographic study of meningoencephalitis from Trypanosoma gambiense before and after treatment with melarsoprol].

The purpose of the study was to assess the long-term effects of melarsoprol on awakening electroencephalogram (EEG) in 18 patients at the meningo-encephalic stage of human African gambiense trypanosomiasis. Electroencephalographic data were taken prior to then 1 and 3 months after therapy. Before treatment EEG tracings showed important abnormalities (delta bursts organized in a more or less periodic fashion); 1 and 3 months after treatment a clear improvement was observed in 10 patients; however, it was unusual that the EEG completely returned to the normal pattern and a case of relapse was no…

Maintaining zebu Maure cattle in a tsetse infested area of Mali.

A study was initiated to evaluate two trypanocidal drugs, the prophylactic isometamidium chloride (Trypamidium) and the curative diminazene aceturate (Berenil) as to their potential for long-term maintenance of zebu cattle in a tsetse infested area of Mali. Trypamidium was administered quarterly and Berenil was administered only as animals were found to be positive for trypanosomes. During the 21-month study trypanosomiasis was the most frequently encountered disease in the cattle in the Berenil treatment group with an average of 5.5 reinfections. The predominant trypanosome species encountere…

Multitask learning-driven identification of novel antitrypanosomal compounds.

Background: Chagas disease and human African trypanosomiasis cause substantial death and morbidity, particularly in low- and middle-income countries, making the need for novel drugs urgent. Methodology & results: Therefore, an explainable multitask pipeline to profile the activity of compounds against three trypanosomes (Trypanosoma brucei brucei, Trypanosoma brucei rhodesiense and Trypanosoma cruzi) were created. These models successfully discovered four new experimental hits (LC-3, LC-4, LC-6 and LC-15). Among them, LC-6 showed promising results, with IC50 values ranging 0.01-0.072 μM and se…

Benzimidazole inhibitors of the major cysteine protease of Trypanosoma brucei.

Aim: Limitations in available therapies for trypanosomiases indicate the need for improved medicines. Cysteine proteases cruzain and rhodesain are validated targets for treatment of Chagas disease and human African trypanosomiasis. Previous studies reported a benzimidazole series as potent cruzain inhibitors. Results & methodology: Considering the high similarity between these proteases, we evaluated 40 benzimidazoles against rhodesain. We describe their structure-activity relationships (SAR), revealing trends similar to those observed for cruzain and features that lead to enzyme selectivity. …

Neuropeptides kill African trypanosomes by targeting intracellular compartments and inducing autophagic-like cell death.

Trypanosoma brucei is the causative agent of African sleeping sickness. Available treatments are ineffective, toxic and susceptible to resis … ophagic-like cell death. Neuropeptide-based treatment improved clinical signs in a chronic model of trypanosomiasis by reducing the parasite burden in various target organs. Of physiological importance is the fact that hosts respond to trypanosome infection producing neuropeptides as part of their natural innate defense. From a therapeutic point of view, targetin …

Parasite genome analysis. Progress in the Leishmania genome project.

Genome projects have been established for 7 major groups of human parasitic infections: malaria, leishmaniasis, African trypanosomiasis, American trypanosomiasis, toxoplasmosis, schistosomiasis and filariasis. All except malaria and toxoplasmosis have come under the umbrella of the World Health Organization's Strategic Committee on Parasite Genome Analysis. The focus of this meeting of the Society was to review progress made in the Leishmania and African trypanosome genome projects. This paper introduces the genome projects and reviews briefly progress in pulsed-field gel karyotype mapping and…

Nitric oxide-mediated suppression of T cell responses during Trypanosoma brucei infection: soluble trypanosome products and interferon-gamma are synergistic inducers of nitric oxide synthase.

African trypanosome infections result in lymphocyte unresponsiveness and anemia in the mammalian host. In murine infections, these effects are mediated by suppressor macrophages releasing nitric oxide (NO). We investigated the mechanism of activation of macrophages to produce NO during trypanosomiasis in vitro. A soluble component of trypanosome lysates induced NO synthesis in peritoneal macrophage cultures only when the macrophages were co-stimulated with interferon-gamma (IFN-gamma). The macrophage-activating factor was also released in a soluble form by live bloodstream-form trypanosomes, b…

[Sleeping sickness in German travelers to the tropics].

A brother and sister (the latter having been resident in Ruanda for three years) fell ill with African trypanosomiasis (sleeping sickness) after a two-day safari in the Akagera National Park. Cardinal symptoms were fever, lymphadenopathy and the typical primary lesion (trypanosomal chancre). The diagnosis was confirmed by demonstrating trypanosomes in the peripheral blood. There was no CNS involvement in either case. Administration of suramin, 1 g weekly intravenously for six weeks, quickly brought about regression of the symptoms and the parasitaemia. According to the number of cases reported…
