Fitting the pieces together: current research on the genetic basis of attention-deficit/hyperactivity disorder (ADHD).

Attention-deficit/hyperactivity disorder (ADHD) is a highly disruptive childhood-onset disorder that often persists into adolescence and adulthood. Comorbidity with other problems, such as autism, dyslexia and conduct disorder (CD) is very common. Although little is known about the pathophysiology of ADHD, family, twin and adoption studies have shown that it is highly heritable. Whole genome linkage studies suggest there are no common susceptibility genes of moderate effect size. Most published research has been based on functional candidate gene studies. The most consistent evidence for assoc

Extending the ATP9A-Related Phenotypic Spectrum: Indication of Schizophrenia Susceptibility.

ATP9A, which belongs to the P4-ATPase family of proteins, is involved in the efficient transport of vesicles from the Golgi apparatus to the plasma membrane, as well as the release of extracellular vesicles from human cells. In 2021, a loss-of-function variant of this gene was identified as being associated with a recessive neurodevelopmental disorder characterized by severe intellectual disability, motor delay, speech impairment, impaired fine motor skills, and behavioral anomalies, such as attention deficit hyperactivity disorder and aggressive behavior. Furthermore, in 2025, a de novo misse

Tolerability and safety profile of risperidone in a sample of children and adolescents.

The aim of this prospective observational study was to verify the tolerability and safety profile of risperidone in a sample of antipsychotic-naive children/adolescent patients having a different psychiatric diagnosis. Twenty-two (mean age of 12±3.2) antipsychotic-naive patients who started therapy with risperidone were recruited. The assessment involved anthropometric data (weight, height, BMI, BMI z-score and BMI percentile), cardiovascular parameters (blood pressure and QTc interval) and blood tests (levels of glucose, triglycerides, total cholesterol, glutamic oxaloacetic and pyruvic trans

Neural mechanisms of frustration in chronically irritable children.

OBJECTIVE: Irritability is common in children and adolescents and is the cardinal symptom of disruptive mood dysregulation disorder, a new DSM-5 disorder, yet its neural correlates remain largely unexplored. The authors conducted a functional MRI study to examine neural responses to frustration in children with severe mood dysregulation. METHOD: The authors compared emotional responses, behavior, and neural activity between 19 severely irritable children (operationalized using criteria for severe mood dysregulation) and 23 healthy comparison children during a cued-attention task completed und

Environmental tobacco smoke exposure and brain development: the case of attention deficit/hyperactivity disorder.

Environmental tobacco smoke, inhaled by active firsthand smokers and their entourage, is associated with morbidity and mortality. Many children are passively exposed to secondhand smoke worldwide. Infants and young children account for the largest global disease burden associated with prenatal and postnatal secondhand smoke, probably due to underdeveloped neurological, immune, and respiro-circulatory systems. There is an increasingly robust association between tobacco smoke exposure, before and after birth, and executive function problems in children, adding to current and future disease burde

Neurodevelopment as a shared genetic etiology linking sleep-related phenotypes and psychiatric disorders: a genome-wide pleiotropic analysis.

Comorbidity and mutual transformation between psychiatric disorders (including autism spectrum disorder, attention-deficit/hyperactivity disorder, bipolar disorder, post-traumatic stress disorder, major depressive disorder, obsessive-compulsive disorder, schizophrenia, and anxiety disorders) and sleep disorders are common, with circadian rhythm disruption considered a potential biological basis, and neurodevelopmental abnormalities seemingly playing a key role. However, the extent to which shared genetic determinants contribute to these associations remains unclear. Extensive genetic correlati

Comorbidities, Social Impact, and Quality of Life in Tourette Syndrome.

Tourette syndrome (TS) is more than having motor and vocal tics, and this review will examine the varied comorbidities as well as the social impact and quality of life (QoL) in individuals with TS. The relationship between any individual and his/her environment is complex, and this is further exaggerated in the case of a person with TS. For example, tics may play a significant role in shaping the person's experiences, perceptions, and interactions with the environment. Furthermore, associated clinical features, comorbidities, and coexisting psychopathologies may compound or alter this relation

Sex, hormones and cerebrovascular function: from development to disorder.

Proper cerebrovascular development and neurogliovascular unit assembly are essential for brain growth and function throughout life, ensuring the continuous supply of nutrients and oxygen. This involves crucial events during pre- and postnatal stages through key pathways, including vascular endothelial growth factor (VEGF) and Wnt signaling. These pathways are pivotal for brain vascular growth, expansion, and blood-brain barrier (BBB) maturation. Interestingly, during fetal and neonatal life, cerebrovascular formation coincides with the early peak activity of the hypothalamic-pituitary-gonadal

Commonality and diversity of impairments in inhibitory tasks after total sleep deprivation: evidence from drift diffusion model.

STUDY OBJECTIVES: This study investigated the effects of total sleep deprivation (TSD) on two key sub-components of inhibitory control-interference inhibition and response inhibition-and examined whether sleep deprivation affects these components through shared or distinct cognitive mechanisms. METHODS: Thirty-six healthy adults (mean age ± SD: 24.83 ± 2.16 years) completed the Flanker and Go/No-Go tasks, which respectively measure interference inhibition and response inhibition, under both normal sleep and TSD conditions. Two variants of the Drift Diffusion Model (DDM) were applied to decomp

The Staggered Spondaic Word Test. A ten-minute look at the central nervous system through the ears.

We have described three major groupings that encompass most auditory processing difficulties. While the problems may be superimposed upon one another in any individual client, each diagnostic sign is closely associated with particular communication and learning disorders. In addition, these behaviors may be related back to the functional anatomy of the regions that are implicated by the SSW test. The auditory-decoding group is deficient in rapid analysis of speech. The vagueness of speech sound knowledge is thought to lead to auditory misunderstanding and confusion. In early life, this may be

A Painful Beginning: Early Life Surgery Produces Long-Term Behavioral Disruption in the Rat.

Early life surgery produces peripheral nociceptive activation, inflammation, and stress. Early life nociceptive input and inflammation have been shown to produce long-term processing changes that are not restricted to the dermatome of injury. Additionally stress has shown long-term effects on anxiety, depression, learning, and maladaptive behaviors including substance abuse disorder and we hypothesized that early life surgery would have long-term effects on theses complex behaviors in later life. In this study surgery in the rat hindpaw was performed to determine if there are long-term effects

Chronobiological disruption in ADHD: Developmental mechanisms linking sleep, behavior, and comorbid conditions.

BACKGROUND: Attention-Deficit/Hyperactivity Disorder (ADHD) is increasingly recognized as a neurodevelopmental condition involving both dopaminergic dysfunction and circadian misalignment. While these mechanisms have traditionally been studied independently, their combined impact on symptom expression and comorbidity-particularly with mood disorders, substance use disorders, and obesity-remains insufficiently understood. This review aims to integrate current evidence on the interaction between circadian and dopaminergic dysfunction in ADHD and to evaluate the therapeutic potential of chronobio

CK1δ over-expressing mice display ADHD-like behaviors, frontostriatal neuronal abnormalities and altered expressions of ADHD-candidate genes.

The cognitive mechanisms underlying attention-deficit hyperactivity disorder (ADHD), a highly heritable disorder with an array of candidate genes and unclear genetic architecture, remain poorly understood. We previously demonstrated that mice overexpressing CK1δ (CK1δ OE) in the forebrain show hyperactivity and ADHD-like pharmacological responses to D-amphetamine. Here, we demonstrate that CK1δ OE mice exhibit impaired visual attention and a lack of D-amphetamine-induced place preference, indicating a disruption of the dopamine-dependent reward pathway. We also demonstrate the presence of abno

Effects of Family Structure on Mental Health of Children: A Preliminary Study.

BACKGROUND: To find any association between family structure and rates of hospitalization as an indicator for behavior problems in children. METHODS: Retrospective chart review of 154 patients who were admitted to the preadolescent unit at Lincoln Prairie Behavioral Health Center between July and December 2012. RESULTS: We found that only 11% of children came from intact families living with biological parents while 89% had some kind of disruption in their family structure. Two-third of the children in the study population had been exposed to trauma with physical abuse seen in 36% of cases.

Childhood adversities and adult psychopathology in the National Comorbidity Survey Replication (NCS-R) III: associations with functional impairment related to DSM-IV disorders.

BACKGROUND: Despite evidence that childhood adversities (CAs) are associated with increased risk of mental disorders, little is known about their associations with disorder-related impairment. We report the associations between CAs and functional impairment associated with 12-month DSM-IV disorders in a national sample. METHOD: We used data from the US National Comorbidity Survey Replication (NCS-R). Respondents completed diagnostic interviews that assessed 12-month DSM-IV disorder prevalence and impairment. Associations of 12 retrospectively reported CAs with impairment among cases (n=2242)

CLINICAL NOTE: CEREBELLAR COGNITIVE AFFECTIVE SYNDROME IMPROVEMENT WITH SELECTIVE INHIBITOR OF SEROTONIN RECAPTATION.

Cerebellar cognitive affective syndrome (CCAS) is characterized by alterations at the cognitive level (dysexecutive syndrome, visuospatial deficit, language ...), associated with affective / emotional changes. Its pathophysiology is not well known and there is currently no specific treatment. We describe a 64-year-old man with a rare condition of cognitive-behavioral disorder after an infarction in the left middle cerebral artery, dominated by executive dysfunctions, predominantly oral apraxia, interrupted divided attention, disturbed visuospatial organization and affective abnormalities with

Kappa opioid receptor antagonism restores phosphorylation, trafficking and behavior induced by a disease-associated dopamine transporter variant.

Aberrant dopamine (DA) signaling is implicated in schizophrenia, bipolar disorder (BPD), autism spectrum disorder (ASD), substance use disorder, and attention-deficit/hyperactivity disorder (ADHD). Treatment of these disorders remains inadequate, as exemplified by the therapeutic use of d-amphetamine and methylphenidate for the treatment of ADHD, agents with high abuse liability. In search for an improved and non-addictive therapeutic approach for the treatment of DA-linked disorders, we utilized a preclinical mouse model expressing the human DA transporter (DAT) coding variant DAT Val559, pre

Neurobehavioral phenotype of autism spectrum disorder associated with germline heterozygous mutations in PTEN.

Germline mutations in PTEN, the gene that encodes phosphatase and tensin homolog, have been identified in up to 20% of children with autism spectrum disorder (ASD) and macrocephaly and are associated with marked abnormalities in the white matter of the brain. This study sought to characterize the neurobehavioral phenotype of PTEN-ASD. Comprehensive neurobehavioral evaluations were conducted in 36 participants (ages 3-21 years) with PTEN-ASD and compared to two groups of controls: non-syndromic ASD with macrocephaly (Macro-ASD, n = 25) and those with PTEN mutations without ASD (PTEN-no ASD, n =

Disruptive behaviour disorders: a systematic review of environmental antenatal and early years risk factors.

Disruptive behaviour disorders (DBDs), including attention-deficit/hyperactivity disorder (ADHD), oppositional defiant disorder (ODD) and conduct disorder (CD) are chronic disorders with significant overlap in aetiology and presentation. An integrative examination of environmental risk factors is lacking. Six literature searches of web-based bibliographic databases were completed to identify literature on DBDs in general and five disorders in particular: CD, ODD, ADHD, deficits of attention, motor control and perception, and reactive attachment disorder. Searches were filtered to focus on stud
