Stem cells exist within a niche, a microenvironment that regulates their activity, but the mechanisms by which niche cells influence stem cell behaviour are poorly understood. In this issue, Stephen DiNardo and colleagues reveal that the shape of the adult Drosophila testes niche, which is dependent on the cytoskeleton of the niche cells, is crucial to maintaining germinal stem cell function. To learn more about this work, we spoke to first author Gabriela Vida and corresponding author Stephen DiNardo, Professor of Cell and Developmental Biology at the University of Pennsylvania, USA.

Talin-1 (TLN1), encoded by the TLN1 gene, is a focal adhesion-related protein capable of binding various proteins in the cytoskeleton. It is also expressed at high levels in many cancers wherein it influences cellular adhesion and the activation of integrins. TLN1 is also capable of promoting tumor cell invasivity, proliferation, and metastatic progression, in addition to being a relevant biomarker and therapeutic target in certain cancers. The present review offers a comprehensive overview of current knowledge regarding TLN1 with respect to its structural properties, functions, and role in tu…

The actin cytoskeleton is critically involved in a variety of cell functions. The Arp2/3 complex mediates branching of filamentous actin. The members of the Wiskott-Aldrich syndrome protein (WASP) family are major regulators of the complex. As such, the family proteins are also involved in numerous aspects of cell biology. In this short review, we first define the expanding WASP family. Next, we compare the domain structure of the members, and explain the known or proposed functions of each domain or region. Finally, we demonstrate the well-characterized roles of the proteins in specific cellu…

A number of unrelated bacterial species as well as vaccinia virus (ab)use the process of actin polymerization to facilitate and enhance their infection cycle. Studies into the mechanism by which these pathogens hijack and control the actin cytoskeleton have provided many interesting insights into the regulation of actin polymerization in migrating cells. This review focuses on what we have learnt from the actin-based motilities of Listeria, Shigella and vaccinia and discusses what we would still like to learn from our nasty friends, including enteropathogenic Escherichia coli and Rickettsia

Glomerular epithelial cells contribute significantly to the hydraulic and macromolecular permeability properties of the glomerulus. Because of their location as the terminal element in the filtration barrier and their elaborate cytoskeleton, they are likely to participate in the regulation of filtration. In disease, alterations of the glomerular epithelial cells may lead to simplification and retraction of foot processes, resulting in a decreased density of slit diaphragms and decreased by hydraulic conductivity. In addition, denudation of epithelial cells from the basement membrane results in…

In the submitted paper the author deals with recent findings on damage of erythrocyte membranes and enzymes in beta-thalassemia. The primary disorder in the formation of beta chains leads to the formation of haemichromes and free oxygen radicals which leads to peroxidative damage of the lipid membrane. The thus generated aldehydes damage important thiol groups of enzymes. Haemichromes interfere with the structure of the protein membrane and the cytoskeleton of the cell. There are differences in membrane damage between alpha and beta thalassaemia. Beta-thalassaemia must be conceived as a compre…

Systemic lupus erythematosus is an autoimmune disease that mainly affects women of reproductive age. Its pathological manifestations directly and indirectly negatively affect the ovarian reserve, the quality of oocytes, and the precise mechanisms of meiosis. This review presents the molecular mechanisms by which lupus damages ovarian tissue and meiosis in oocytes. The role of chronic inflammation, impaired hormonal levels, and the presence of specific autoantibodies are considered. The available data on how oocyte structures (meiotic spindle, actin cytoskeleton, membrane organelles and chromat…

Proximal spinal muscular atrophy, the most frequent genetic cause of childhood lethality, is caused by homozygous loss or mutation of the SMN1 gene on human chromosome 5, which codes for the survival motor neuron (SMN) protein. SMN plays a role in the assembly of small nuclear ribonucleoproteins and, additionally, in synaptic function. SMN deficiency produces defects in motor neuron β-actin mRNA axonal transport, neurofilament dynamics, neurotransmitter release, and synapse maturation. The underlying molecular mechanisms and, in particular, the role of the cytoskeleton on the pathogenesis of t…

LSP1 is an F-actin bundling cytoskeletal protein expressed in hematopoietic lineage and endothelial cells. We investigated the function of this protein by generating and analyzing an LSP1-deficient mouse strain and in this review we describe our findings together with those of other investigators. The results show a complex function of LSP1 in regulating leukocyte recruitment to inflamed sites. Based on current evidence, we propose that the levels of LSP1 on the cytoskeleton and the type of integrin involved are some of the critical elements which affect LSP1 function in modulating the thresho…

One case of transitional cells carcinoma with sarcomatoid pattern, located in the renal pelvis in a 77 year-old male with haematuria. The tumour also expressed a non-neoplastic component of osteoclast-like giant cells which probably represent a stromal reaction of the host to the neoplasia. The tumoral cells co-express keratin and vimentin, two strands of the cytoskeleton specific for epithelial and mesenchymal cells, respectively. We speculate with the possibility of the tumoral cells fusocellular morphology being related to the vimentin expression. A review of the literature is also included…

The wall of intact tissue cysts in the brains of mice with congenital toxoplasmosis was investigated using light and electron microscopy. Impregnation of the wall with reduced silver salts and protargol silver suggested that it was composed, at least in part, of components derived from the neuronal cytoskeleton. Electron microscopy extended these observations and revealed that intact cysts were separated from the extracellular compartment by a layer of neurofibrillae enclosed within the host cell membrane. It is suggested that this may be a means whereby intact Toxoplasma tissue cysts are prot…

Neuronal function relies on the precise coordination between intracellular calcium (Ca2+) signaling and the cytoskeletal architecture that underpins synaptic transmission, plasticity, and structural stability. Disruption of this calcium-cytoskeleton interplay has been noted in numerous neurodegenerative diseases. We discuss how Ca2+-dependent cytoskeletal remodeling governs long-term potentiation and depression, dendritic spine morphology, and presynaptic function, highlighting the functions of end-binding proteins, STIM (Stromal Interaction Molecule)/Orai-mediated store-operated calcium entry…

The continued rise in cancer-related deaths suggests that the precise nature of the disease remains poorly understood. In the field of cellular mechanics, researchers have identified mechanobiological aspects of cancer cells by examining the link between their physical and molecular properties and functions, thereby revealing some of their previously unknown characteristics. Therefore, expanding scientific insight in this area may deepen our understanding of genetic and mechanical interactions and clarify the functional mechanisms at both the cellular and subcellular levels. Accordingly, in th

The actin/spectrin membrane-associated periodic skeleton (MPS) is a cytoskeletal structure that supports axonal integrity and function. Lower spinal motor neurons (MNs) are characterized by exceptionally long axons and are particularly susceptible to degeneration in a wide range of hereditary neuromuscular disorders, including amyotrophic lateral sclerosis. Using confocal and super-resolution imaging, we characterized the spatial distribution of βII-spectrin and the assembly pattern of the MPS in human MN axons derived from induced pluripotent stem cells. We discovered a striking gap-and-patch

Nanovibration, a kilohertz-frequency, nano-amplitude mechanical stimulation, has been shown to drive osteogenesis; however, the mechanisms remain unclear. Mechanotransduction has been proposed with limited cell population-level evidence. We propose the use of a high-throughput mechanical phenotyping technique, Real-Time Deformability Cytometry (RT-DC), to observe mechanical changes in an osteogenic model, MG63s. We have demonstrated that MG63 cells respond to the nanovibrational mechanical stimulation by changing their cytoskeletal morphology and showing higher expression of osteocalcin than r

Morphogenesis requires dynamic changes in cell and tissue mechanics. Because organs are complex multi-cellular structures that develop slowly, understanding how they acquire their unique 3-D forms requires in vivo experimental systems that can bridge the discrete processes that occur at different physical-scales, from cell, to tissue, to multi-organ, and at different time-scales, from minutes, to hours, to days. The challenge of establishing causal relationships across scales has limited exploration of the cell biological processes that collectively produce the changes in tissue material prope

The actin cytoskeleton plays a crucial role in fundamental eukaryotic processes such as morphogenesis, motility, endocytosis, intracellular trafficking, and cell division. However, our understanding of actin and its associated proteins in trypanosomatid parasites like Leishmania remains limited. Over the past two decades, considerable progress has been made in elucidating the structure and functions of Leishmania actin and its core regulators. Notably, these findings are primarily derived from studies of the insect-stage promastigote form, while the roles of the actin machinery during the dise…

Efficient methods for the extraction of features of interest remain one of the biggest challenges for the interpretation of cryo-electron tomograms. Various automated approaches have been proposed, many of which work well for high-contrast datasets where the features of interest can be easily detected and are clearly separated from one another. Our inner ear stereocilia cryo-electron tomographic datasets are characterized by a dense array of hexagonally packed actin filaments that are frequently cross-connected. These features make automated segmentation very challenging, further aggravated by
