Engineering Escherichia coli for Perillyl Alcohol Production with Reduced Endogenous Dehydrogenation.

Perillyl alcohol is a widely utilized antitumor agent in cancer therapy. Due to the limitations associated with chemical synthesis and plant extraction methods, bioengineering microorganisms for the production of perillyl alcohol and its precursor limonene offers a more scalable and industrially compatible approach. In this study, we present the design of an Escherichia coli cell factory capable of de novo synthesis of limonene and perillyl alcohol from glucose. A comprehensive systems engineering approach was employed to increase precursor availability, enhance electron transfer efficiency, a…

Engineering CO2-Fixing Carboxysome into Saccharomyces cerevisiae to Improve Ethanol Production.

Bacterial microcompartments (BMCs) are intracellular structures for compartmentalizing specific metabolic pathways in bacteria. As a unique type of BMCs, carboxysomes utilize protein shells to sequester ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and carbonic anhydrase for efficient carbon dioxide (CO2) fixation. This study aims to reconstruct an α-carboxysome in Saccharomyces cerevisiae and investigate its metabolic effects. Here, genes of the cso operon from Halothiobacillus neapolitanus, Calvin cycle-related enzyme phosphoribulokinase (PRK) from Spinacia oleracea, and two S. c

Promoter engineering for enhanced 3-hydroxypropionic acid production in Pichia pastoris.

Enabling tools are essential for facilitating the methanol bioconversion in Pichia pastoris. However, there is still a relative lack of prom … yl-CoA pathways for the production of 3-hydroxypropionic acid. Further combining rational metabolic engineering strategies, such as optimizing gene copy numbers and blocking byproduct synthesis pathways, the engineered strains CHP9 and CHP20 achieved 3-HP titers of 23 g/L and 22 g/L by using methanol as the sole carbon source. These results indicate that adap …

Non-human primate: the new frontier model of female reproductive engineering.

Reproductive engineering encompasses a range of advanced tissue engineering techniques aimed at addressing infertility that is non-curable with current assisted reproductive technology (ART). The use of animal models has been crucial for these advancements, with a notable preference for non-human primates (NHPs) given their genetic, anatomical, and physiological similarities to humans. Therefore, NHPs are invaluable for studying reproductive engineering. Thus, in reproductive studies, NHPs bridge the anatomical and physiological gaps between rodent models and humans. Their shared features with…

Engineering endoplasmic reticulum targeted metal-polyphenol curcumin nanomicelles for melanoma therapy.

Melanoma has a high incidence and mortality, and current therapies are limited. Here, we developed TCPC, an ER-targeted nanomedicine guided by omics analysis. It consists of curcumin-loaded PEG-PCL nanomicelles coated with a Tannic acid-Cu metal-polyphenol network. Guided by clinical transcriptome mining, we identified ER-stress vulnerabilities to inform material design. RNA-seq and imaging confirmed that TCPC preferentially accumulates in melanoma cells and localizes to the ER. TCPC markedly elevates CHOP expression, which triggers ER Ca²⁺ release, mitochondrial dysfunction, ROS accumulation,

Surface-Adsorbed Oxygen-Triggered Luminescence-Guided Defect Engineering of Oxygen Vacancy.

Oxygen vacancies (Ov) in metal oxide-based materials can regulate their surface chemical and electronic properties, and thus the amount of Ov is crucial to exert their photo-, electro-, and thermocatalytic performances. Defect engineering has been introduced to prepare metal oxide-based materials with sufficient Ov. However, the current efforts have been hampered due to the failure to quantify the absolute values of Ov in metal oxide-based materials. In this work, we have prepared ZnFe oxides with varied Ov through the calcination of a ZnFe hydroxide precursor under the guidance of reaction-tr…

Engineering a macromolecular JAK inhibitor for treating acute inflammation and endotoxaemia.

Uncontrolled and sustained inflammation is inextricably associated with the pathogenesis of numerous diseases. However, there is still demand for effective and safe anti-inflammatory therapies. Here we report a potent anti-inflammatory macromolecular therapy named HPL, created by conjugating polyethylene glycol and luminol onto a multivalent and hydrolysable cyclic structure. Leveraging its amphiphilic nature, HPL can spontaneously self-assemble into micelles capable of targeting inflamed tissues and localizing in inflammatory cells. In mice with acute lung, kidney and liver injuries, as well

Engineering single-layered poly(amidoamine) dendrimer microcapsules for enhanced cellular uptake.

Microcapsules are considered as one of the most promising drug carriers due to their exceptional characteristics. The cellular uptake of microcapsules is determined by physicochemical properties, yet comprehensive studies on thickness and shape effects are limited. In this study, we developed single-layered poly(amidoamine) (PAMAM) dendrimer microcapsules with tunable thicknesses and morphologies to systematically investigate their cellular internalization. By varying PAMAM generations (G2-G8), the wall thickness can be tuned within the range of 19.3-53.7 nm, while the microcapsule shapes can

Engineering a placenta-inspired biomimicking fibrous patch for chondrogenesis through immunomodulation.

The frequent onset of hyper-inflammatory responses after implantation impairs the availability of tissue transplants, impedes cellular renewal, and ultimately leads to the failure of scaffold-based cartilage regeneration. Existing scaffold modification strategies show limited efficacy in controlling immune cell infiltration and modulating inflammatory responses. Inspired by the placenta's dual protective and immunomodulatory attributes-through its physical barrier and biofactors release-this study develops a biomimetic surface-engineered aligned poly (L-lactic acid) (PLLA) fibrous patch barrie

The Regulation of Pericellular Matrix Synthesis During Articular Cartilage Tissue Engineering.

Articular cartilage, vital to the health and functioning of joints, remains challenging to regenerate. The pericellular matrix (PCM) is critical for transducing biophysical stimuli to the articular chondrocytes (ACs) that it envelops. Given the mechanobiological sensitivity of ACs, it is pivotal in maintaining the chondrogenic phenotype and the production of extracellular matrix (ECM) during articular cartilage tissue engineering. While the maintenance of the native PCM significantly improves the quality of neocartilage, current isolation methods are limited. A solution to this challenge is fa…
