Mechanism of reaction of a suggested superoxide-dismutase mimic, Fe(II)-N,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine.

The interaction of a recently developed intracellular superoxide dismutase analogue, Fe(II)-N,N,N',N'-tetrakis(2- pyridylmethyl)ethylenediamine (Fe(II)-TPEN), with reactive oxygen species was investigated under in vitro conditions. The complex catalyzed the dismutation of enzyme- or radiolysis-generated superoxide with the production of H2O2; under steady-state conditions the equilibrium was strongly shifted toward Fe(III)-TPEN. Fe(II)-TPEN reacted with H2O2 to generate hydroxyl radicals in a Fenton reaction. The oxidized Fe(III)-TPEN was readily reduced by ascorbate or glutathione. Given the

[Preventing iron deficiency in breast-fed infants by suitable supplementary food. A prospective, controlled study].

Iron deficiency may develop in prolonged breast feeding. Introduction of beikost (supplementary nutrition) is recommended in Germany for infants after 4 months of age. In a prospective study 73 exclusively breastfed infants at the age of 16 weeks were assigned to one of two feeding groups: 35 infants received a meat vegetable dinner fortified with iron-2-sulfate (3 mg iron per 100 kcal) as their first supplementary food. At 20 weeks of age a milk based rice cereal (MBRC) without iron fortification was added as a second beikost meal. The other group comprised 38 infants who first received a MBR

Iron management during recombinant human erythropoietin therapy.

Treatment with recombinant human erythropoietin (r-HuEPO; EPOGEN [epoetin alfa], AMGEN Inc, Thousand Oaks, CA) rapidly corrects the anemia associated with end-stage renal disease during the acute phase of therapy and supports hematocrit levels throughout the maintenance phase. However, during the acute phase of therapy, iron deficiency will develop in most patients; it is therefore initially essential to monitor body iron stores monthly. A plasma ferritin level of less than 30 ng/mL or a transferrin saturation level of less than 20% confirms the diagnosis of iron deficiency. Microcytic, hypoch

Efficient clearance of non-transferrin-bound iron by rat liver. Implications for hepatic iron loading in iron overload states.

In hemochromatosis and other disorders associated with iron overload, a significant fraction of the total iron in plasma circulates in the form of low molecular weight complexes not bound to transferrin. Efficient and unregulated clearance of this form of iron by the liver may account for the hepatic iron loading and toxicity that characterize these diseases. We tested this possibility by examining the hepatic removal process for representative iron complexes in the single-pass perfused rat liver. Hepatic uptake of both ferrous and ferric 55Fe from ultrafiltered human serum was found to be hig

Effect of streptococcal lipoteichoic acid on prolyl hydroxylase activity as related to collagen formation in mouse fibroblast monolayers.

Dried and wet mouse fibroblast monolayers with labeled collagenous substrate were used to study the effects of lipoteichoic acid (LTA) on cellular prolyl hydroxylase activity. LTA is a scavenger of cations, and Fe2+ is essential for prolyl hydroxylase activity. Surprisingly, addition of LTA to dried monolayers resulted in increased prolyl hydroxylase activity, whereas preincubation of Fe2+ with LTA only negated this increase. However, significant inhibition of enzyme activity by wet monolayers occurred whether LTA was added directly to the test system or whether it was used after preincubation

Haematological aspects of life with an ileostomy.

The haematological aspects of life with an ileostomy have been studies in 51 patients, of whom 39 had had ulcerative colitis and 12 had Crohn's disease. The findings in these patients have been compared with those in 39 healthy volunteers who were matched for age and sex with the 39 patient who had had ulcerative colitis. There was evidence of a mild degree of iron deficiency in the patients with an ileostomy. This was partly due to a pre-existing iron deficiency resulting from their preceding illness and operation, as the abnormality was less pronounced in the patients in whom the ileostomy h

Enhancers of iron absorption: ascorbic acid and other organic acids.

Ascorbic acid (AA), with its reducing and chelating properties, is the most efficient enhancer of non-heme iron absorption when its stability in the food vehicle is ensured. The number of studies investigating the effect of AA on ferrous sulfate absorption far outweighs that of other iron fortificants. The promotion of iron absorption in the presence of AA is more pronounced in meals containing inhibitors of iron absorption. Meals containing low to medium levels of inhibitors require the addition of AA at a molar ratio of 2:1 (e.g., 20 mg AA: 3 mg iron). To promote absorption in the presence o

Photothermal and immunomodulatory chitosan composite hydrogel remodels the pathological microenvironment to accelerate infected diabetic wound healing.

The recalcitrant nature of chronic diabetic wounds stems from a complex pathological microenvironment characterized by bacterial biofilm formation, elevated reactive oxygen species (ROS) levels, and persistent inflammation. To address this challenge, we developed an L-arginine/ferrocene dual-functionalized chitosan hydrogel architecture (CLF@MZ) integrated with Ti3C2 MXene quantum dots (QDs) and ZIF-8 nanoparticles. This system aims to remodel the wound microenvironment through a multi-synergistic photothermal-biochemical strategy. Upon near-infrared irradiation, the MXene QDs generate localiz

A non-enzymatic sensor based on rGO/Pt NPs/Fc-Tyr/POPD nanocomposite for hydrogen peroxide determination in liver cancer tissues.

Malignant tumors remain a major global health challenge, highlighting the need for rapid and sensitive analytical tools for investigating cancer-associated oxidative stress. In this study, we developed a non-enzymatic electrochemical hydrogen peroxide (H₂O₂) sensor based on a reduced graphene oxide/platinum nanoparticle/ferrocene-tyramine/poly(o-phenylenediamine) (rGO/Pt NPs/Fc-Tyr/POPD) nanocomposite. H₂O₂, although not a tumor-specific biomarker, is an important reactive oxygen species associated with oxidative stress in cancer-related systems. The proposed sensing platform integrates the hi

Integration of dual-output TMSD and CRISPR/Cas12a for crosstalk-free electrochemiluminescence detection of aflD gene and AFB1.

Aflatoxins are among the most toxic mycotoxins and pose a severe threat to food safety and human health. In addition to the direct monitoring of aflatoxin B1 (AFB1), simultaneous detection of its key biosynthesis gene, aflD, can effectively indicate the presence of toxin-producing strains, thereby enhancing the early screening and traceability of AFB1 contamination. Due to the significant functional differences among various biomarkers, performing multi-target analysis on a single detection interface remains challenging. Herein, we constructed a novel dual-target electrochemiluminescence (ECL)

Characterizing ferrous versus ferric iron in Alzheimer's disease using X-ray fluorescence imaging and XANES spectroscopy.

BackgroundThe accumulation of iron, such as ferrous Fe2+, in the Alzheimer's disease (AD) brain may contribute to neurodegeneration by driving oxidative stress. While elevated iron in AD has been shown, the oxidation state of iron and its regional distribution in AD, particularly in the hippocampus, is unclear.ObjectiveTo characterize the oxidation state and spatial distribution of iron in the hippocampus of AD and control brains, and to assess the effect of tissue thawing on ferrous iron measurements.MethodsWe utilized X-ray fluorescence imaging and X-ray absorption near edge structure spectr

A smart polymer-based nanoplatform potentiates cancer ferroptosis-immunotherapy via perturbing metabolic compensation.

Ferroptosis, an iron-dependent form of regulated cell death, has emerged as a promising strategy for antitumor immunotherapy. However, its therapeutic efficacy is hindered by the persistently high intracellular glutathione (GSH) level driven by metabolic compensation. Herein, we report a ferroptosis-inducible polymeric nanovesicle encapsulated with BAY-876 and ferrous ion (Fe2+) (FIPNV/B@Fe2+) to potentiate ferroptosis-immunotherapy via perturbing metabolic compensation. Mechanically, double bonds in the polymeric side chain deplete the intracellular GSH and coordinated Fe2+ facilitates lipid

A Spatial Confinement Effect-Based "Swing Probe" In Situ Entropy-Driven Assembly for the Sensitive Ratiometric Electrochemical Detection of Exosomal miRNA.

Electrochemical biosensors based on DNA assembly signal amplification have emerged as powerful tools for exosomal miRNA detection, demonstrating broad application potential in accurate early cancer diagnosis. However, due to the passivation of the electrode by DNA nanostructures and inherent limitations in assembly reaction efficiency, electrochemical biosensors still face challenges in the sensitive and rapid detection of exosomal miRNA. Herein, we propose a novel spatial confinement effect based "swing probe" in situ entropy-driven assembly strategy. By constraining two auxiliary probes in a

Synthesis, characterization, and microbiological evaluation of new triazolopyrimidine-based ferrocenes as potent antimicrobial prospects.

Antimicrobial metallodrugs have gained considerable attention for their potent inhibitory activity and clinical success, driving the development of novel metallodrug candidates. These efforts have uncovered new bioactive scaffolds and mechanisms of action. However, the global challenge of antimicrobial resistance (AMR), fueled by the genetic adaptability of microbes and resistance to nearly all antibiotic classes, highlights the urgent need for innovative antibiotics. In this study, we expand the repertoire of metallodrugs by designing, synthesizing, and biologically evaluating triazolopyrimid

Impact of pH and salinity fluctuations on oxidation of Fe(II) by nitrate-reducing microorganisms enriched from the reduced tidal sediment of an extreme acidic river (Río Tinto, Spain).

Nitrate reduction coupled to Fe(II) oxidation (NRFeOx) contributes to Fe cycling in the estuarian sediments of the Río Tinto river (Huelva, Spain). However, it is not yet known (i) whether and which NRFeOx microorganisms can be enriched from the reduced sediment layer and (ii) how in situ pH and salinity fluctuations affect NRFeOx. Therefore, we (i) used two different approaches, such as microcosm experiments (sediment amended with either NO3-/Fe2+aq or acetate/NO3-/Fe2+aq) and enrichment cultures (medium amended with acetate/NO3-/Fe2+aq) to enrich NRFeOx microorganisms to (ii) test their sali

Haem iron versus ferrous iron salts to treat iron deficiency anaemia in Gambian children: protocol for randomised controlled trial {1}.

BACKGROUND: The World Health Organization recommends universal iron supplementation for children aged 6-23 months in countries where anaemia is seen in over 40% of the population. Conventional ferrous salts have low efficacy due to low oral absorption in children with inflammation. Haem iron is more bioavailable, and its absorption may not be decreased by inflammation. This study aims to compare daily supplementation with haem iron versus ferrous sulphate on haemoglobin concentration and serum ferritin concentration after 12 weeks of supplementation. METHODS: This will be a two-arm, randomise

Fluorinated 1,7-DO2A-Based Iron(II) Complexes as Sensitive 19F MRI Molecular Probes for Visualizing Renal Dysfunction in Living Mice.

Kidney diseases have become an important global health concern due to their high incidence, inefficient diagnosis, and poor prognosis. Devising direct methods, especially imaging means, to assess renal function is the key for better understanding the mechanisms of various kidney diseases and subsequent development of effective treatment. Herein, we developed a fluorinated ferrous chelate-based sensitive probe, 1,7-DO2A-Fe(II)-F18 (Probe 1), for 19F magnetic resonance imaging (MRI). This highly fluorinated probe (containing 18 chemically equivalent 19F atoms with a fluorine content at 35 wt %)

Assessing the Protein Quality, In Vitro Intestinal Iron Absorption and Human Faecal Microbiota Impacts of Plant-Based Mince.

The nutritional quality of plant-based meat analogues compared to traditional meat products has been questioned in recent commentary, particularly in relation to protein quality and micronutrient bioavailability. However, the attributes of specific products within this category are unclear. We therefore undertook a comprehensive assessment of the compositional and functional attributes of v2food® (Sydney, Australia) plant-based mince, including an assessment of the effects of reformulation, including the addition of amino acids, ascorbic acid, and different forms of elemental iron. The protein
