Causal association between body mass index and temporomandibular disorders: a bidirectional two-sample Mendelian randomization analysis.

BACKGROUND: Observational studies have shown that body mass index (BMI) is highly correlated with the occurrence of temporomandibular disorders (TMDs). However, these studies failed to present a causal relationship. Thus, we aimed to performed a Mendelian randomization (MR) study to investigate causality between BMI and TMDs. METHODS: We performed a two-sample bidirectional MR analysis using large-scale genome-wide association studies (GWAS). Data were obtained from a large-scale BMI dataset (N = 322,154), TMDs dataset (N = 134,280). The causal effects were estimated with inverse-variance wei

Oily fish and raw vegetable consumption can decrease the risk of AQP4-positive neuromyelitis optica spectrum disorders: a Mendelian-randomization study.

Neuromyelitis optica spectrum disorders (NMOSD) are severe inflammatory disorders of the central nervous system targeting aquaporin-4 (AQP4). The risk factors for NMOSD remain to be determined, though they may be related to diet and nutrition. This study aimed to explore the possibility of a causal relationship between specific food intake and AQP4-positive NMOSD risk. The study followed a two-sample Mendelian randomization (MR) design. Genetic instruments and self-reported information on the intake of 29 types of food were obtained from a genome-wide association study (GWAS) on 445,779 UK Bio

Experimental viral evolution to specific host MHC genotypes reveals fitness and virulence trade-offs in alternative MHC types.

The unprecedented genetic diversity found at vertebrate MHC (major histocompatibility complex) loci influences susceptibility to most infectious and autoimmune diseases. The evolutionary explanation for how these polymorphisms are maintained has been controversial. One leading explanation, antagonistic coevolution (also known as the Red Queen), postulates a never-ending molecular arms race where pathogens evolve to evade immune recognition by common MHC alleles, which in turn provides a selective advantage to hosts carrying rare MHC alleles. This cyclical process leads to negative frequency-de

Parallel Genomic Responses to Ecological Gradients in the Phalaenopsis amabilis Complex.

There are multiple mechanisms that can underlie parallel adaptation to similar habitats at the genetic level, including de novo mutations, introgression, and shared standing variation. While evidence for each pathway is emerging, few studies have directly examined how demographic processes, range expansion and introgression interact to shape the fate of adaptive alleles. In the Phalaenopsis amabilis complex, we applied a dimensionality reduction approach to identify environmental extremes and link alleles to ecological gradients. By combining gene-environment association analyses with demograp

Genetic association and potential mediators between age at menarche and cholelithiasis: a two-sample two-step bidirectional mendelian randomization study.

OBJECTIVE: Pubertal reproductive factors, such as age at menarche, have been linked to various long-term health outcomes, but their potential role in the development of cholelithiasis remains insufficiently studied. Although observational studies have suggested a possible association between menarche timing and gallstone risk, inconsistent findings and methodological limitations have left this relationship unclear. To fill this gap, we performed a bidirectional two-sample Mendelian randomization (MR) study to investigate the causal relationship between age at menarche and the risk of cholelith

Natural bioactive compounds targeting the macrophage clock in asthma: evidence, challenges, and perspectives.

BACKGROUND: Nocturnal asthma and airway remodeling are intimately linked to pulmonary circadian dysfunction. The dysregulation of core clock genes (e.g., Bmal1, Rev-erbα) in macrophages drives metabolic reprogramming and pro-fibrotic signaling. Pharmacological modulation of this clock using natural bioactive compounds offers a promising therapeutic strategy. PURPOSE & SCOPE: This review critically evaluates the in vivo evidence of natural chronobiotics (including resveratrol, quercetin, curcumin, honokiol, and nobiletin) in reshaping the asthmatic microenvironment, specifically focusing on su

Oral health status and esophageal diseases: Univariate and multivariate Mendelian randomization analyses.

Emerging evidence suggests a profound connection between oral health status and esophageal diseases. Nevertheless, the causal relationship remains controversial. This Mendelian randomization (MR) study was performed to probe into the genetic causality between oral health status and esophageal diseases. Summary statistics from genome-wide association studies were obtained from the FinnGen project and the UK Biobank consortium. The number of instrumental single nucleotide polymorphisms per trait ranged from approximately 8.97 million to 16.38 million. The analyzed datasets included sample sizes

Metabolic reprogramming-driven neuroimmunoregulation: Key mechanisms and therapeutic opportunities and challenges in central nervous system disorders.

Central nervous system (CNS) disorders are fundamentally linked to metabolic dysregulation within immune and glial cells. This review provides a systematic synthesis of immunometabolic reprogramming-encompassing glucose, lipid, and amino acid metabolism, and oxidative phosphorylation-in CNS-resident microglia, immunomodulatory astrocytes, and peripherally infiltrating immune cells (T cells, B cells, and neutrophils) across Alzheimer's disease, Parkinson's disease, multiple sclerosis, and ischemic stroke. Critically, rather than presenting all reported metabolic alterations as equivalently esta

Gut microbiota, inflammatory cytokines and gastro-esophageal reflux disease: A Mendelian randomization analysis.

Gut microbiota has been recognized as an extrahepatic manifestation of gastro-esophageal reflux disease (GERD) in observational studies. However, the directionality and causality of the association and whether cytokines act as a mediator remain unclear. We aim to estimate the casual relationship between gut microbiota, inflammatory cytokines and GERD using a 2-sample Mendelian randomization method. Gut microbiota, cytokines, and GERD were identified using summary data from the genome-wide association studies and the FinnGen consortium. The primary method for causal estimation was the inverse-v

Investigating the causal association between coffee and migraine using Mendelian randomization analysis.

Background: Migraine is a chronic neurological disorder influenced by various risk factors, including inadequate medication, anxiety, sleep disorders, depression, and stress-inducing events. Coffee is well-known for its diverse bioactive properties and may help relieve acute migraine symptoms; however, stopping long-term coffee consumption can unexpectedly trigger migraines. While some migraine sufferers view caffeine as a potential trigger, research suggests it may also have preventive effects. The complex physiological and pharmacological mechanisms behind coffee and its active components ar
