Tryptophan residues in alpha-galactosidase from Trichoderma reesei.

Tryptophan residues in alpha-galactosidase were modified with bromosuccinimide. The fact that galactose, a specific inhibitor of alpha-galactosidase, does not prevent this modification demonstrates that tryptophan residues are not located in galactose binding sites. Analysis of the inactivation kinetics revealed two groups of Trp residues (8.5 and 7.5 residues) with different accessibility for N-bromosuccinimide. We studied specific quenching of alpha-galactosidase fluorescence resulting from modification of an sulfhydryl group in the active site of the enzyme with Hg2+ and Ag+ ions. The speci

Potency of anti-trematode in inhibiting cathepsin-2L and cathepsin-5L of Fasciola gigantica using homology, docking, and molecular dynamics.

BACKGROUND: Cathepsin-L (FhCL) is a group of enzymes that most flukes express and secreted significantly in parasite-host interactions. Researches are focusing on antigens released by Fasciola gigantica as one of the keys to understanding immunologic pathways in parasite infection and targets for anthelmintics. Efforts to suppress FhCL function through vaccination or therapy using anthelmintic drugs are key factors in controlling field-level trematode infections. A molecular docking method can be used to observe the interaction between FhCL and some anthelmintic drugs to better understand the

[Pyloroplasty and motile functions of the stomach and duodenum].

Mongrel dogs were prepared by attaching 2 and 1 silver needle bipolar electrodes on the gastric antrum and bulbus of the duodenum, respectively. Among the obtained active potential from these dogs at chronic stage, investigations were performed mainly on the discharge frequency of spikes, propagation pattern of electrical excitation, and relationship of the stomach and duodenum. In addition, barium was loaded as the stomach contents and extraction dynamics of the loads were observed radiologically. The following was these investigations concerning about the pyloroplasty and motile functions of

A six-week balance training intervention with older inmates improves static balance in a German prison.

INTRODUCTION: Given the accelerated aging processes and elevated fall risk associated with functional decline in correctional settings, developing effective exercise programs for older inmates is increasingly important. METHODS: This study examined the potential effects of a six‑week balance‑training program on static and dynamic balance performance, well‑being, and enjoyment of physical activity among older inmates in an open German prison. Ten male inmates (mean age = 62.1 ± 4.4 years) participated, with five assigned to an intervention group and five to a control group. The intervention co

A retrospective cohort study of three wound catheter infusion analgesia regimes after midline laparotomy.

BACKGROUND: Local anesthetic wound infusion catheters are increasingly used postmidline laparotomy to reduce pain and opioid use, however there is little evidence to support any particular infusion regime. METHODS: A retrospective cohort study was undertaken of patients after midline laparotomy who had bilateral local anesthetic wound infusion catheters surgically placed. Patients were recruited into 3 cohorts: ropivacaine 0.2% 5-8 mL/hour continuous wound infusion, 10 mL programmed intermittent bolus 2 hourly, 20 mL programmed intermittent bolus 4 hourly. The primary outcome was the maximum

Clinical and hemodynamic significance of innominate artery lesions evaluated by ultrasonography and digital angiography.

To determine the hemodynamic and clinical consequences of an atherosclerotic obstructive lesion of the innominate artery on the cerebral circulation, 20 patients with an innominate artery lesion underwent neurologic examination and ultrasonic duplex scanning before and after right arm exercise. The patients were divided into two groups: Group 1, 12 patients with 40-80% stenosis and Group 2, eight patients with 80-100% stenosis. A significant difference between the groups was noted in both the hemodynamic and clinical manifestations. All 12 Group 1 patients compensated for the increased demand

Abnormal intrinsic functional network connectivity in adolescent major depressive disorder related to severity of insomnia.

BACKGROUNDS: Insomnia is closely associated with depression and plays a critical role in its development. Investigating insomnia-related neuroimaging changes in depression could enhance understanding of the neural mechanisms underlying depression insomnia. However, the brain network mechanism underlying the bidirectional relationship between depression and insomnia in adolescents remains unknown. METHODS: We recruited 83 drug-naïve adolescents with MDD and classified them into high- (HI-aMDD) and low-insomnia (LI-aMDD) groups based on the Hamilton Depression Rating Scale sleep subscale. Aberrant dynamic functional network connectivity in progressive supranuclear palsy.

BACKGROUND: The clinical symptoms of progressive supranuclear palsy (PSP) may be mediated by aberrant dynamic functional network connectivity (dFNC). While earlier research has found altered functional network connections in PSP patients, the majority of those studies have concentrated on static functional connectivity. Nevertheless, in this study, we sought to evaluate the modifications in dynamic characteristics and establish the correlation between these disease-related changes and clinical variables. METHODS: In our study, we conducted a study on 53 PSP patients and 65 normal controls. In

Altered Dynamic Functional Network Connectivity in Healthy Adults with Acute Pain: Findings from the Human Connectome Project.

Characterizing the neural signature of pain and its modulation is critical for assessing treatment efficacy and conducting translational clinical research. However, the dynamics of pain processing in the brain have remained largely unknown. In this study, we employed independent component analysis (ICA) as a data-driven clustering method on resting-state functional magnetic resonance imaging (fMRI) to obtain intrinsic connectivity networks (ICNs) in a cohort of healthy adults from the Human Connectome Project (HCP) who were identified as having acute pain. We examined the temporal dynamic func

Aberrant global and local dynamic properties in schizophrenia with instantaneous phase method based on Hilbert transform.

BACKGROUND: Emerging functional imaging studies suggest that schizophrenia is associated with aberrant spatiotemporal interaction which may result in aberrant global and local dynamic properties. METHODS: We investigated the dynamic functional connectivity (FC) by using instantaneous phase method based on Hilbert transform to detect abnormal spatiotemporal interaction in schizophrenia. Based on resting-state functional magnetic resonance imaging, two independent datasets were included, with 114 subjects from COBRE [51 schizophrenia patients (SZ) and 63 healthy controls (HCs)] and 96 from Open

Abnormal early dynamic individual patterns of functional networks in low gamma band for depression recognition.

BACKGROUND: The functional networks are associated with emotional processing in depression. The mapping of dynamic spatio-temporal brain networks is used to explore individual performance during early negative emotional processing. However, the dysfunctions of functional networks in low gamma band and their discriminative potentialities during early period of emotional face processing remain to be explored. METHODS: Functional brain networks were constructed from the MEG recordings of 54 depressed patients and 54 controls in low gamma band (30-48 Hz). Dynamic connectivity regression (DCR) alg

Clinical Implication of Mid-Range Dynamic Instability in Lumbar Degenerative Spondylolisthesis.

STUDY DESIGN: Retrospective evaluation. PURPOSE: To determine the prevalence of mid-range dynamic instability in patients with degenerative spondylolisthesis (DS) and to evaluate the clinical implication of mid-range instability (MI). OVERVIEW OF LITERATURE: Instability is identified by measuring vertebral body anterior-posterior translation on static end-range flexion and extension lateral radiographs. Mid-range kinematics could evince occult dynamic instability in which motion is not appreciated at the terminal-range of motion. METHODS: In this study, 30 patients with DS with checked stan

Unilateral cochlear implant use promotes normal-like loudness perception in adolescents with childhood deafness.

OBJECTIVES: Behavioral measures of cochlear implant (CI) device stimulation levels can be difficult to obtain in individuals with limited or no hearing experience. Loudness measures are particularly challenging. It would therefore be useful to have a battery of objective and behavioral measures to determine CI stimulation levels in listeners with childhood deafness. In the present study, the authors characterized loudness growth in 20 adolescents: 8 with normal hearing and 12 CI participants with pre/perilingual bilateral sensorineural hearing loss. They asked (1) do adolescent CI users with c

Radiological adjacent-segment degeneration in L4-5 spondylolisthesis: comparison between dynamic stabilization and minimally invasive transforaminal lumbar interbody fusion.

OBJECTIVE Pedicle screw-based dynamic stabilization has been an alternative to conventional lumbar fusion for the surgical management of low-grade spondylolisthesis. However, the true effect of dynamic stabilization on adjacent-segment degeneration (ASD) remains undetermined. Authors of this study aimed to investigate the incidence of ASD and to compare the clinical outcomes of dynamic stabilization and minimally invasive transforaminal lumbar interbody fusion (MI-TLIF). METHODS The records of consecutive patients with Meyerding grade I degenerative spondylolisthesis who had undergone surgical

Effect of Transcranial Direct Current Stimulation with Balance Training in a Middle-Aged Population: Randomized Double-Blind Sham-Controlled Trial.

Introduction: The first signs of deteriorated balance impairment begin during middle age. Early intervention could delay the fall risk in older populations; hence, addressing balance deficits during this age is crucial. The authors aimed to determine the effects of transcranial direct current stimulation (tDCS) combined with balance training (BT) on the improvement of static and dynamic balance in a middle-aged population, along with the participants' safety and satisfaction. Methods: Participants (n = 28) were randomized into two groups: active tDCS (active tDCS + BT) and sham tDCS (sham tDCS

Therapeutic efficacy of intravenous lidocaine infusion compared with thoracic epidural analgesia in major abdominal surgery: a noninferiority randomised clinical trial.

BACKGROUND: Open major abdominal surgery is one of the most risky surgical procedures for acute postoperative pain. Thoracic epidural analgesia (TEA) has been considered the standard analgesic approach. In different reports, lidocaine i.v. has been shown to have an analgesic efficacy comparable with TEA. We compared the analgesic efficacy of i.v. lidocaine with thoracic epidural analgesia using bupivacaine in patients undergoing major abdominal surgery. METHODS: In this noninferiority clinical trial, 210 patients were randomised to thoracic epidural bupivacaine with morphine or i.v. lidocaine

Impaired recognition of facial emotions from low-spatial frequencies in Asperger syndrome.

The theory of 'weak central coherence' [Happe, F., & Frith, U. (2006). The weak coherence account: Detail-focused cognitive style in autism spectrum disorders. Journal of Autism and Developmental Disorders, 36(1), 5-25] implies that persons with autism spectrum disorders (ASDs) have a perceptual bias for local but not for global stimulus features. The recognition of emotional facial expressions representing various different levels of detail has not been studied previously in ASDs. We analyzed the recognition of four basic emotional facial expressions (anger, disgust, fear and happiness) from

Mutagenic properties of oxidized GTP and ATP in in vitro transcription-reverse transcription.

Ankylosing spondylitis (AS) is known as a microbiome-driven disease; however, the current understanding of microbiota dynamics in AS is limited. In the present study, we conducted a 16S rDNA sequence-based microbiota survey of 97 fecal samples from healthy subjects and AS patients at baseline, 1, 3 and 6 months after anti-TNF-α treatment to demonstrate the dynamic characteristic variations of gut microbiota in AS patients. The goal of this experiment is to explore the values of gut microbiota as biomarkers of disease activity and therapeutic responses to anti-TNF-α. We found that the relative

Incidence and simple management of priapism following dynamic infusion cavernosometry-cavernosography.

Of 239 patients with erectile dysfunction (aged 36 to 70 years) who were evaluated with dynamic infusion cavernosometry-cavernosography, 32 (13.4%) developed priapism after the procedure and were successfully managed with immediate intracorporal injection of phenylephrine. No single risk factor for the development of priapism was identified in this group. Early pharmacologic intervention for priapism induced by dynamic infusion cavernosometry-cavernosography is a simple, safe, and time-saving measure to achieve detumescence and prevent potential sequelae such as corporal ischemia or fibrosis.
