Genetic analysis of the HPRT mutation of Lesch-Nyhan syndrome in a Chinese family.

BACKGROUND: The Lesch-Nyhan syndrome is an X-linked recessive inherited disease caused by a complete deficiency of hypoxanthine guanine phosphoribosyl-transferase (HPRT) activity. Many different mutations throughout the HPRT coding region of Lesch-Nyhan patients have been described, including single base substitutions, partial or entire gene deletions, gene insertions or endoduplication of exons. However, study of gene mutation in Chinese patients has rarely been reported in Taiwan. METHODS: Polymerase chain reaction (PCR) and nucleotide sequence analysis were used to identify the location an

Screening for molecular pathologies in Lesch-Nyhan syndrome.

Heteroduplex detection by hydrolink gel electrophoresis was performed to screen for small mutations in 12 Lesch-Nyhan syndrome families with characterised molecular pathology which included nine point mutations, two small deletions, and a 1-bp insertion. This modified protocol for heteroduplex detection by hydrolink gel electrophoresis detected all 12 of these mutations and was utilised to rapidly determine the carrier status of females from affected families. On the basis of these results this approach appears to be a rapid and reliable screening method for point mutations in addition to smal

Dendrobium officinale polysaccharide-spermidine composite hydrogels: Tunable rheological properties and improved in vitro colonic fermentation.

Dendrobium officinale polysaccharide (DOP) and spermidine (Spd) exhibit synergistic anti-aging effects in Caenorhabditis elegans, suggesting potential functional complementarity. Here, a DOP-Spd composite hydrogel (DS) was constructed to integrate structural tunability with fermentation-related functionality. Three weight ratios (18:2, 18:4, and 18:6, DOP:Spd) successfully formed hydrogels. Increasing Spd content enhanced gel strength and thermal stability, as reflected by higher hardness (149.62-353.75 g), chewiness (41.04-119.07), and DTG peak temperature (331.7-334.0 °C). All DS hydrogels e

Xin-Zi-Sheng-Wan decoction alleviates hyperuricemia-associated renal injury by inhibiting RIPK1/RIPK3/MLKL-mediated necroptosis.

ETHNOPHARMACOLOGICAL RELEVANCE: Xin-Zi-Sheng-Wan Decoction (XZSWD) is a modified formulation derived from the classical prescription Zi-Sheng Pill, which was recorded in Xian Xing Zhai Yi Xue Guang Bi Ji from the Ming Dynasty. Traditionally, this formula is used to eliminate "turbid stagnation" and restore metabolic balance. Contemporary ethnopharmacological evidence indicates that XZSWD can reduce uric acid (UA) levels and ameliorate renal injury, although the molecular mechanism underlying these effects remains incompletely understood. AIM OF THE STUDY: The aim of this study was to determin

Urinary metabolomic signatures of muscle injury and recovery in elite football players.

INTRODUCTION: Muscle injuries are a leading cause of time-loss in elite football, highlighting the need for objective biomarkers capable of reflecting the athlete's physiological state. Monitoring strategies rely predominantly on external load metrics and may not fully capture internal physiological responses to training and competition. Urinary metabolomics offers a non-invasive approach to characterize injury related physiological processes. OBJECTIVES: To identify urinary metabolomic signatures associated with muscle injury in elite football players and to explore their potential utility f

Establishment and phenotypic characterization of a rat model with a clinically relevant phenotype for chronic gouty arthritis.

BACKGROUND: The study of chronic gouty arthritis (CGA) is limited by the absence of animal models that faithfully mimic its chronic progression. Existing models fail to capture the transition from acute flares to chronic joint damage, highlighting the need for a more representative model. METHODS: A novel CGA model was established in SD rats using a combined strategy. Chronic hyperuricemia was induced via a diet containing 2% potassium oxonate and 12% yeast. This was supplemented with twice-weekly gavage of hypoxanthine to simulate acute uric acid fluctuations, followed by intra-articular inj

Metabolomic signature reveals dysregulated lipoprotein profile in m.3243A>G carriers: a case-control study.

INTRODUCTION: The pathogenic mitochondrial gene variant m.3243A>G disrupts oxidative phosphorylation and is associated with insulin resistance, both of which may be linked to unfavorable lipid metabolism. However, the metabolic alterations in m.3243A>G carriers, including what differentiates those with and without diabetes, remain incompletely understood. OBJECTIVES: To investigate metabolomic profiles in fasting serum and urine samples from m.3243A>G carriers compared to healthy controls. METHODS: Metabolomic profiling of serum and urine samples using nuclear magnetic resonance-based metabo

Exploring the relationship between XDH gene variants and serum urate and xanthine dehydrogenase levels in gout.

BACKGROUND: Gout is a type of arthritis caused by high levels of urate in the body, leading to inflammation and joint pain. Xanthine dehydrogenase (XDH) is a key enzyme in urate biosynthesis, and genetic variations in its XDH gene can influence the amount or activity of the enzyme, as well as the levels of urate produced. OBJECTIVES: This study aimed to explore the association of two single nucleotide variants (SNVs) in the XDH gene with gout susceptibility in the Mexican population, as well as their correlation with urate and XDH levels. METHODS: Genotyping of rs17323225 and rs17011368 XDH

Mass spectrometry-based untargeted metabolomics study of non-obese individuals with non-alcoholic fatty liver disease.

OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is a disease characterized by the accumulation of excessive fat in the liver, which can lead to fibrosis and has an increasing prevalence. NAFLD requires non-invasive diagnostic biomarkers. While typically observed in overweight individuals, it can also occur in non-obese/non-overweight individuals. Comparative studies on non-obese NAFLD patients are scarce. This study aimed to conduct a using liquid chromatography-high resolution mass spectrometry (LC-MS/MS)-based metabolic profiling of non-obese NAFLD patients and healthy controls. MATER

Macrophages undergo functionally significant reprograming of nucleotide metabolism upon classical activation.

During an immune response, macrophages systematically rewire their metabolism in specific ways to support their diversve functions. However, current knowledge of macrophage metabolism is largely concentrated on central carbon metabolism. Using multi-omics analysis, we identified nucleotide metabolism as one of the most significantly rewired pathways upon classical activation. Further isotopic tracing studies revealed several major changes underlying the substantial metabolomic alterations: 1) de novo synthesis of both purines and pyrimidines is shut down at several specific steps; 2) nucleotid

In vitro piperaquine susceptibility is not associated with the Plasmodium falciparum chloroquine resistance transporter gene.

BACKGROUND: Dihydroartemisinin-piperaquine is a new ACT that is administered as single daily dose for three days and has been demonstrated to be tolerated and highly effective for the treatment of uncomplicated Plasmodium falciparum malaria. Piperaquine was used alone to replace chloroquine as the first-line treatment for uncomplicated malaria in China in response to increasing chloroquine resistance in the 1970s. However, the rapid emergence of piperaquine-resistant strains that resulted in the cessation of its use in China in the 1980s, suggests that there is cross-resistance between piperaq
