The PI3K/Akt inhibitor LY294002 reverses BCRP-mediated drug resistance without affecting BCRP translocation.

Cellular responses toward cytotoxic drugs are influenced by crosstalk between oncogenic signals and resistance mechanisms. Inhibition of the PI3K/Akt pathway is effective in sensitizing cancer cells of various organs, although the mechanisms largely remain to be elucidated. Breast cancer resistance protein (BCRP)/ABCG2, a drug efflux pump, confers resistance to multiple anticancer agents such as SN-38 and topotecan. Previous studies reported that inhibition of the PI3K/Akt pathway, by gene knockout or PI3K inhibitors, modulated BCRP-mediated drug transport via BCRP translocation in hematopoiet

Molecular targeting of breast and colon cancer cells by PAR1 mediated apoptosis through a novel pro-apoptotic peptide.

A novel activating peptide was designed and synthesized from V. cholerae hemagglutinine protease (HAP) mediated cleavage site of mouse PAR1. The peptide "PFISED" interacts with PAR1 in a new site which is different from its thrombin mediated conventional activation site and induced a series of new downstream signaling pathways. The peptide showed apoptosis in human and mouse breast (MCF-7 and EAC) and colon (HT29 and CT26) cancer cells where as in the same peptide concentration in normal human breast epithelial cells (MCF-10A), normal human fibroblast cells (MRC-5), normal mouse peritoneal mac

Hyaluronic Acid-Targeted Stimuli-Sensitive Nanomicelles Co-Encapsulating Paclitaxel and Ritonavir to Overcome Multi-Drug Resistance in Metastatic Breast Cancer and Triple-Negative Breast Cancer Cells.

Active targeting and overcoming multi-drug resistance (MDR) can be some of the important attributes of targeted therapy for metastatic breast cancer (MBC) and triple-negative breast cancer (TNBC) treatment. In this study, we constructed a hyaluronic acid (HA)-decorated mixed nanomicelles-encapsulating chemotherapeutic agent paclitaxel (PTX) and P-glycoprotein inhibitor ritonavir (RTV). HA was conjugated to poly (lactide) co-(glycolide) (PLGA) polymer by disulfide bonds (HA-ss-PLGA). HA is a natural ligand for CD44 receptors overexpressed in breast cancer cells. Disulfide bonds undergo rapid re

Sakuranetin modulates estrogen receptor signaling in breast cancer cells.

Identification of natural, bioactive compounds has been of interest to the biomedical field, in particular for the treatment of cancer. Flavonoids are a family of compounds, present across many plant species, known for their polyphenolic structure that results in cellular bioactivity. The flavonoid, sakuranetin, is produced due to external stressors, through the naringenin biosynthetic pathway. Binding affinity and docking simulations confirmed sakuranetin binding to the estrogen receptor alpha (ERα) pocket in a manner similar to, but with weaker affinity than 17β-estradiol (E2). Due to sakura

A Light-Controlled DNA Nanoclaw Machine: Integrated Multiplexed Recognition, Spatiotemporally Programmable Therapy, and Real-Time Electrochemical Signaling for Precision Receptor Regulation.

Cell membrane receptors are pivotal targets in precise therapeutics, yet their ubiquitous expression across tissues remains a fundamental barrier to achieving cell-specific intervention. To overcome the limitations of conventional monotargeting approaches, we developed a light-gated DNA nanoclaw machine (L-DNM) that integrates high-specificity recognition, spatiotemporally controlled therapy, and real-time monitoring of molecular activation within a unified nanoplatform. The L-DNM employed a multivalent targeting mechanism directed against a triple-marker signature, epithelial cell adhesion mo

Unraveling the therapeutic potential of a thiosemicarbazone-based Zn(II) metal-organic triangle complex.

Demand for new and effective chemotherapeutic agents continues to drive the search for innovative metal-based drug candidates. We introduce here a metal-organic triangle complex as a novel candidate against breast cancer cells. The self-assembly of a new thiosemicarbazone building block, N4-cyclohexyl-2-(2-hydroxy-3,5-diiodobenzylidene)hydrazinecarbothioamide (H2L), with zinc(II) ions resulted in the formation of a novel metallosupramolecular complex, [Zn3(µ-L)3·3DMF] (1), where DMF acts as an ancillary ligand stabilizing the coordination environment. The unique structure of the trinuclear com

Functional evidence of hsa-miR-21-5p and hsa-miR-3158-3p in regulating immune pathways relevant to cerebral malaria.

BACKGROUND: Cerebral malaria (CM), the most severe manifestation of Plasmodium falciparum infection, is characterized by immune dysregulation, endothelial dysfunction, and brain injury. MicroRNAs (miRNAs) have emerged as promising biomarkers and regulators of malaria pathogenesis. Previous clinical studies identified elevated plasma levels of hsa-miR-21-5p and hsa-miR-3158-3p as correlates of CM severity and poor outcomes. However, their mechanistic roles in modulating host immune responses remain poorly defined. METHODS: To elucidate the regulatory effects of these miRNAs, MCF7 and HepG2 cel

Bioactive silver nanoparticles derived from Carica papaya floral extract and its dual-functioning biomedical application.

Replacing synthetic phytochemicals with natural plant extracts for metal nanoparticle synthesis enable cost-effective, large-scale production with reduced environmental and health risks while enhancing biomedical efficacy. This study presents the green synthesis of silver nanoparticles (AgNPs) using a flavonol-enriched extract from male papaya flowers (KQE), an underutilized agricultural waste. Using 20% (v/v) KQE, highly stable, spherical KQ-AgNPs (12.3 ± 3.0 nm) were synthesized via in-situ generation of free radicals, such as ortho-quinones, which reduced Ag+ ions. KQ-AgNPs exhibit superior

Development of PEGylated PLGA Nanoparticles Co-Loaded with Bioactive Compounds: Potential Anticancer Effect on Breast Cancer Cell Lines.

OBJECTIVE: The incidence of breast cancer continues to rise despite decades of laboratory, epidemiological and clinical research. Breast cancer is still the leading cause of cancer death in women. Cyclin D1 is one of the most important oncoproteins associated with cancer cell proliferation and is overexpressed in more than 50% of cases. Curcumin and chrysin are plant-derived components that are believed to assist in inhibiting the viability of breast cancer cells. These agents are involved in cancer cells' growth and reducing cyclin D1 expression. In this study, the hypothesis of combining cur

The palladacycle, BTC2, exhibits anti-breast cancer and breast cancer stem cell activity.

In women globally, breast cancer is responsible for most cancer-related deaths and thus, new effective therapeutic strategies are required to treat this malignancy. Platinum-based compounds like cisplatin are widely used to treat breast cancer, however, they come with limitations such as poor solubility, adverse effects, and drug resistance. To overcome these limitations, complexes containing other platinum group metals such as palladium have been studied and some have already entered clinical trials. Here we investigated the anti-cancer activity of a palladium complex, BTC2, in MCF-7 oestroge

Limitations of an ex vivo breast cancer model for studying the mechanism of action of the anticancer drug paclitaxel.

Paclitaxel is widely used in the treatment of breast, ovarian, lung, and other cancers. Its primary mechanism is to prevent microtubule depolymerization causing loss of dynamic instability crucial for normal microtubule function leading to mitotic arrest. Prolonged mitotic arrest results in cell death as a secondary response. The effects of paclitaxel are typically studied in cell lines which precludes assessment of the possible influence of tumor-associated cells. We therefore examined paclitaxel action ex vivo in fresh explant cultures of human breast tumors. Surprisingly, we found that pacl
