O-acetylated glucomannan from Dendrobium officinale alleviates DSS-induced colitis through dual immune-oxidative regulation and gut-microbiota modulation.

Plant-derived polysaccharides are promising phytotherapeutics for mucosal inflammation. We purified an O-acetylated glucomannan (DOP-a) from Dendrobium officinale and investigated its protective effects against dextran sulfate sodium (DSS)-induced colitis and the underlying immune and oxidative-stress mechanisms. DOP-a was purified and structurally characterized, and its monosaccharide composition, molecular weight, and O-acetylation/branching patterns were determined. In a chronic DSS mouse model, disease activity, colon length, histopathology, tight-junction proteins, and inflammatory cytoki

A review on molecular crosstalk: Bioactive polysaccharides and gut microbiota in type 2 diabetes-Pathways, signaling, and therapeutic translation.

Type 2 diabetes mellitus (T2D) is a chronic metabolic disorder characterized by insulin resistance, impaired glucose homeostasis, and low-grade inflammation, in which gut microbiota dysbiosis plays a pivotal role in disease progression. Natural bioactive polysaccharides (NBPs) from diverse sources have emerged as effective microbiota-targeted agents with antidiabetic potential. After colonic fermentation, NBPs are metabolized into key microbial products, including short-chain fatty acids (SCFAs), indole derivatives, and secondary bile acids (SBAs), which reshape microbial ecological compositio

Ultrasound-assisted deep eutectic solvent extraction of green, high-efficient, and high-bioactive polysaccharides from Glycyrrhiza uralensis.

Licorice (Glycyrrhiza uralensis) roots are among the most widely used medicinal raw materials in traditional medicine and in numerous industries, with their polysaccharides serving as key bioactive constituents responsible for immunomodulation, tissue repair, and drug synergy. However, the low extraction yield and quality of the polysaccharides limit their extensive utilization. Presently, a highly efficient method for the extraction of polysaccharides from G. uralensis roots is lacking. Here, we developed an ultrasound-assisted deep eutectic solvent (UDE) method for extracting high-yield and

Mass spectrometric analysis of lipid-linked oligosaccharide heterogeneity in genetically modified E. coli used for glycoconjugate bacterial vaccine production.

Polysaccharide-based glycoconjugate bacterial vaccines can help combat multidrug-resistant pathogens by harnessing the immune system to recognize and neutralize bacteria effectively. Bacterial cells can be engineered to link O-antigen polysaccharides (O-PSs) to a carrier protein, producing a potential bacterial vaccine, e.g., against extraintestinal pathogenic Escherichia coli (ExPEC). Lipid-linked oligosaccharides (LLOs) play a key role in the bioconjugation process, and determining their heterogeneity is essential to evaluate the variability among clinical isolates and the success of the bio

Structure characteristics of a pectic polysaccharide from the leaves of Cornus officinalis Sieb. et Zucc. (Shanzhuyu) and its inhibition on hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) represents a highly aggressive malignancy with limited treatment options. While the traditional herb Cornus officinalis Sieb. et Zucc. (C. officinalis, commonly called Asiatic dogwood or "Shanzhuyu" in Chinese) is known for its hepatoprotective properties, the structural features and anti-HCC potential of polysaccharides from its leaves remain largely unexplored. Therefore, we hypothesized that the leaf-derived polysaccharides with uncharacterized structure may exhibit anti-HCC activity. In this study, a homogeneous anti-HCC polysaccharide, designated PCL-2I (Mw

Cytoplasmic sequestering of a fungal stress-activated MAPK in response to a host plant phenolic acid.

Fungal pathogens employ conserved signaling pathways to survive in the host. The stress-activated MAP kinase Hog1 of the maize pathogen Cochliobolus heterostrophus undergoes dephosphorylation upon exposure to ferulic acid, a phenolic compound abundant in the host plant. Unlike its nuclear localization during osmotic stress, Hog1 forms cytoplasmic foci in response to FA, indicating its sequestering to a compartment or condensate. FA prevents several characteristic responses of the Hog1 pathway to osmotic stress: hyperphosphorylation of Hog1, nuclear localization, and expression of a monosacchar

The emerging importance of the α-keto acid dehydrogenase complexes in serving as intracellular and intercellular signaling platforms for the regulation of metabolism.

The α-keto acid dehydrogenase complex (KDHc) class of mitochondrial enzymes is composed of four members: pyruvate dehydrogenase (PDHc), α-ketoglutarate dehydrogenase (KGDHc), branched-chain keto acid dehydrogenase (BCKDHc), and 2-oxoadipate dehydrogenase (OADHc). These enzyme complexes occupy critical metabolic intersections that connect monosaccharide, amino acid, and fatty acid metabolism to Krebs cycle flux and oxidative phosphorylation (OxPhos). This feature also imbues KDHc enzymes with the heightened capacity to serve as platforms for propagation of intracellular and intercellular signal
