Role of laminin in axonal extension from olfactory receptor cells.

The role of laminin, an extracellular matrix molecule believed to be involved in axon extension, was explored in the outgrowth of olfactory receptor cells and therefore in the maintenance of organization in the olfactory pathway. First, immunocytochemistry was used to examine laminin expression in the olfactory nerve and bulb during development. Laminin immunoreactivity was high in the olfactory nerve and glomerular layers. Although it declined in intensity, laminin expression continued in the nerve and in single glomeruli of adults. Second, the influence of laminin on neurite outgrowth was ex

Toward defining the regenerative potential of olfactory mucosa: establishment of Schwann cell-free adult canine olfactory ensheathing cell preparations suitable for transplantation.

Olfactory mucosa (OM)-derived olfactory ensheathing cells (OECs) are attractive candidates for autologous cell transplantation-based therapy … ating that freshly dissociated OM prior to culturing contained as many Schwann cells as OECs, while olfactory bulb (OB) primary cell suspensions revealed lower levels of Schwann cell contamination. Interestingly, neurite growth of neonatal rat dorsal root ganglion (DRG) neurons cocultured with OM-OECs, OB-OECs, and fibular nerve (FN) Schwann cells used as contro …

Microglial/macrophage cells in mammalian olfactory nerve fascicles.

This is the first description of a population of Iba1- and annexin A3-immunopositive cells residing in the peripheral olfactory nerves of adult rats and adult cats. Based on their ramified appearance, positive immunostaining for the monocytic markers Iba1 and annexin A3, and reactivity to bulbectomy (in adult rats), these cells found within the olfactory nerve fascicles of both mammalian species meet several important criteria for their designation as microglia/macrophages. These Iba1-/annexin A3-immunopositive cells may be uniquely positioned to protect against the potential spread of dangero

Continuous neural plasticity in the olfactory intrabulbar circuitry.

In the mammalian brain each olfactory bulb contains two mirror-symmetric glomerular maps linked through a set of reciprocal intrabulbar projections. These projections connect isofunctional odor columns through synapses in the internal plexiform layer (IPL) to produce an intrabulbar map. Developmental studies show that initially intrabulbar projections broadly target the IPL on the opposite side of the bulb and refine postnatally to their adult precision by 7 weeks of age in an activity-dependent manner (Marks et al., 2006). In this study, we sought to determine the capacity of intrabulbar map …

The BEACH Protein LRBA Promotes the Localization of the Heterotrimeric G-protein Golf to Olfactory Cilia.

BEACH domain proteins are involved in membrane protein traffic and human diseases, but their molecular mechanisms are not understood. The BEACH protein LRBA has been implicated in immune response and cell proliferation, and human LRBA mutations cause severe immune deficiency. Here, we report a first functional and molecular phenotype outside the immune system of LRBA-knockout mice: compromised olfaction, manifesting in reduced electro-olfactogram response amplitude, impaired food-finding efficiency, and smaller olfactory bulbs. LRBA is prominently expressed in olfactory and vomeronasal chemose…

The olfactory system as a puzzle: playing with its pieces.

The mammalian olfactory bulb (OB) has all the features of a whole mammalian brain but in a more reduced space: neuronal lamination, sensory inputs, afferences, or efferences to other centers of the central nervous system, or a contribution of new neural elements. Therefore, it is widely considered as "a brain inside the brain." Although this rostral region has the same origin and general layering as the other cerebral cortices, some distinctive features make it very profitable in experimentation in neurobiology: the sensory inputs are driven directly on its surface, the main output can be acce…

[Neural circuit analysis using isolated whole brain preparation].

Many electrophysiological studies have been conducted on the 'limbic system', which is known to play a crucial role in mnemonic, emotional, … ng voltage-sensitive dyes in order to investigate how olfactory information is transferred from the olfactory bulb to the piriform, entorhinal and amygdaloid cortices. We visualized the propagation pattern of the neural activity after olfactory nerve stimulation and found that piriform and entorhinal activities converge in the amygdaloid cortex. This convergence …

Brain structure is changed in congenital anosmia.

Olfactory function in healthy people correlates with structural features of both the olfactory bulb and higher order olfactory processing areas, but we do not yet know how congenital anosmia affects these latter structures. In order to examine this question closer, we acquired T1 weighted magnetic resonance images from 17 subjects with congenital anosmia and from 17 age- and sex-matched controls. We compared white and gray matter volumes as well as cortical thickness between both groups. We found subjects with congenital anosmia to exhibit larger gray matter volumes in the left entorhinal and …

A reversible rat model of hyposmia affects respiration-linked brain oscillations and behavior.

Rhythmic breathing coordinates neural activity across olfactory and limbic circuits by coupling nasal airflow to oscillations in the olfactory bulb (OB) and downstream networks. To test how reduced peripheral olfactory input reshapes this respiration-linked coordination, we developed a reversible hyposmia model in rats using intranasal gadolinium chloride. This manipulation impaired hidden cookie performance for around 1 week and was accompanied by nasal epithelial damage. Hyposmia robustly decreased OB respiration-linked (1-10 Hz) and gamma (30-90 Hz) power, with smaller reductions in prefron…

Coding odor modality in piriform cortex efficiently with low-dimensional subspaces: a shared covariance decoding approach.

A fundamental question in neuroscience is how sensory signals are decoded from noisy cortical activity. We address this question in the olfactory system, decoding the route by which odorants arrive into the nasal cavity: through the nostrils (orthonasal), or through the back of the throat (retronasal). We recently showed with modeling and novel experiments on anesthetized rats that orthonasal versus retronasal modality information is encoded in the olfactory bulb (OB, a pre-cortical region). However, key questions remain: is modality information transmitted from OB to anterior piriform cortex …

Olfactory ensheathing cells represent an optimal substrate for hippocampal neurons: an in vitro study.

Olfactory ensheathing cells (OECs) are cells that display Schwann cell or astrocyte-like properties. They are a source of growth factors and adhesion molecules which play a very important role as neuronal support enhancing cellular survival. Over the past 10 years, OECs have emerged as a leading reparative candidate, when transplanted into the injured spinal cord, having shown significant promise in the regeneration of spinal cord lesions. In this study we assessed the efficacy of OECs on the survival and neurite outgrowth of hippocampal neurons in vitro. Co-cultures of OECs and hippocampal of

Terminal nerve in the mouse-eared bat (Myotis myotis): ontogenetic aspects.

As in other mammals, ontogenesis of the terminal nerve (TN) in the mouse-eared bat (Myotis myotis) starts shortly after the formation of the … er of the nasal roof before reaching the central ganglion (M) located in the meninges medial to the olfactory bulb. From the mid-fetal period onward, a thin TN fiber bundle with some intermingled perikarya connects M to the brain by penetrating its wall rostral to the olfactory tubercle. Additional smaller ganglia may occur in this region. The TN and its ganglia …

Different olfactory system deficits affect the antigonadal action of light deprivation differently in male rats.

Twenty-eight day-old male rats were subjected to: Blinding-olfactory bulbectomy, Blinding-peripheral anosmia, Blinding-accessory olfactory bulbectomy and Blinding-sham olfactory operation. A set of rats remained intact. Six weeks later, their pituitary-gonads-accessory sex organs were studied. Bulbectomy as well as peripheral anosmia exaggerated the antigonadal effects of blindness, while the accessory olfactory system impairment did not. It is suggested that olfactory bulbectomy potentiation of the antigonadal effects of light deprivation is due to a lack of sensory function rather than to bu…

Expression of CD36 by Olfactory Receptor Cells and Its Abundance on the Epithelial Surface in Mice.

CD36 is a transmembrane protein that is involved in the recognition of certain amphiphilic molecules such as polar lipids in various tissues and body fluids. So far, CD36 homologues in insects have been demonstrated to be present on the surface of olfactory dendrites and to participate in the perception of exogenous compounds. However, little is known about the relationship between CD36 and mammalian olfaction. Indeed, the detection of only CD36 mRNA in the mouse olfactory epithelium has been reported to date. In the present study, to provide potential pieces of evidence for the involvement of

Olfactory system measurements in COVID-19: a systematic review and meta-analysis.

PURPOSE: The neurotropism of SARS-CoV-2 and the consequential damage to the olfactory system have been proposed as one of the possible underlying causes of olfactory dysfunction in COVID-19. We aimed to aggregate the results of the studies which reported imaging of the olfactory system of patients with COVID-19 versus controls. METHODS: PubMed and EMBASE were searched to identify relevant literature reporting the structural imaging characteristics of the olfactory bulb (OB), olfactory cleft, olfactory sulcus (OS), or olfactory tract in COVID-19 patients. Hedge's g and weighted mean difference…

Human cord blood CD133+ stem cells transplanted to nod-scid mice provide conditions for regeneration of olfactory neuroepithelium after permanent damage induced by dichlobenil.

The herbicide dichlobenil selectively causes necrosis of the dorsomedial part of olfactory neuroepithelium (NE) with permanent damage to the underlying mucosa, whereas the lateral part of the olfactory region and the nasal respiratory mucosa remain undamaged. We investigated here whether human umbilical cord blood CD133(+) stem cells (HSC) injected intravenously to nod-scid mice pretreated with dichlobenil may engraft the olfactory mucosa and contribute to the regeneration of the damaged NE. We tested HLA-DQalpha1 DNA and three human microsatellites (Combined DNA Index System) as indicators of
