Tirzepatide on ingestive behavior in adults with overweight or obesity: a randomized 6-week phase 1 trial.

Tirzepatide induces weight reduction but the underlying mechanisms are unknown. This 6-week phase 1 study investigated early effects of tirzepatide on energy intake. Male and female adults without diabetes (n = 114) and a body mass index from 27 to 50 kg per m2 were randomized 1:1:1 to blinded once-weekly tirzepatide or placebo, or open-label once-daily liraglutide. The primary outcome was change from baseline to week 3 in energy intake during an ad libitum lunch with tirzepatide versus placebo. Secondary outcomes assessed self-reported ingestive behavior and blood-oxygenation-level-dependent

Multimodal machine learning for analysing multifactorial causes of disease-The case of childhood overweight and obesity in Mexico.

BACKGROUND: Mexico has one of the highest global incidences of paediatric overweight and obesity. Public health interventions have shown only moderate success, possibly from relying on knowledge extracted using limited types of statistical data analysis methods. PURPOSE: To explore if multimodal machine learning can enhance identifying predictive features from obesogenic environments and investigating complex disease or social patterns, using the Mexican National Health and Nutrition Survey. METHODS: We grouped features into five data modalities corresponding to paediatric population exogeno…

Research on the characteristics of gut microbiota in overweight patients with early-onset type 2 diabetes mellitus.

BACKGROUND: Early-onset type 2 diabetes mellitus (EOD), defined as a metabolic disorder diagnosed before the age of 40, with higher risks of complications, vascular events, and mortality, poses a significant threat to public health. Its prevalence has risen markedly in overweight populations in recent years, yet the underlying mechanisms remain elusive. Emerging evidence highlights a close association between gut microbiota (GM) and metabolic disorders, suggesting that modulating gut microbial dysbiosis may serve as a potential strategy for preventing and managing overweight EOD. METHODS: In …

Overweight and Obesity in Pediatric Kidney Transplant Recipients Are Not Associated With Higher Risks of Early Postoperative Complications.

BACKGROUND: Childhood overweight and obesity have been hypothesized to pose challenges in pediatric kidney transplantation due to potential complications and resultant worse outcomes. Herein, we aimed to determine if pediatric patients with an elevated body mass index (BMI) (> 85th percentile) are at higher risk for short-term complications following kidney transplantation. METHODS: Following a review of our transplant database (2010-2020), patients > 2-18 years were assigned to groups based on BMI percentiles at the time of surgery: normal (5th-85th percentile; n = 120) and obese/overweight …

Association between Food Insecurity and Overweight: a Systematic Review based on Intersectionality of Gender and Race/Color.

CONTEXT: The relationship between Food Insecurity (FI) and health conditions such as chronic diseases, including obesity, has been discussed in the literature. However, discourse on the association of FI with other social vulnerabilities is a relatively new and less elucidated area. OBJECTIVE: Considering the evidence that gender and race/color are factors of interest in the relationship between FI and overweight, in this study we aimed to carry out a systematic review to analyze the state of knowledge about this relationship and how the intersectionality of these factors permeates the associ…

Hormonal regulation of human adipose tissue lipolysis: impact of adipose GIP system in overweight and obesity.

OBJECTIVE: Given the promising effects of GLP-1/GIP/glucagon receptor triagonists on weight loss in animals and humans, improved understanding of underlying mechanism is required. We investigated a direct lipolytic effect of a specific GLP-1/GIP/glucagon receptor triagonist on human adipose tissue to disentangle central and peripheral effects as potential drivers of weight loss. DESIGN AND METHODS: Isolated primary adipocytes from subcutaneous adipose tissue biopsies of 22 non-diabetic subjects [63.0 (57.0-69.5) years] were incubated with increasing concentrations of isoprenaline, GLP-1, GIP,

High-sensitivity CRP is elevated in pregnant women with overweight and obesity and modulated by gestational weight gain.

INTRODUCTION: The global obesity epidemic presents a growing challenge in perinatal medicine and obstetrics, as it is associated with a higher risk of adverse maternal and perinatal outcomes. In addition to metabolic disturbances, obesity contributes to chronic low-grade inflammation. This study aims to investigate the relationship between maternal overweight, obesity, and excessive weight gain during pregnancy and high-sensitivity C-reactive protein (hs-CRP), an acute-phase reactant of inflammation and infection in maternal serum. MATERIAL AND METHODS: The PRINCE (Prenatal Identification of …

The efficacy of sleep lifestyle interventions for the management of overweight or obesity in children: a systematic review and meta-analysis.

BACKGROUND: Childhood obesity remains a significant public health concern. Sleep duration and quality among children and youth are suboptimal worldwide. Accumulating evidence suggests an association between inadequate sleep and obesity risk, yet it is unclear whether this relationship is causal. This systematic review examines the efficacy of sleep interventions alone or as a part of lifestyle interventions for the management of overweight or obesity among children and adolescents. METHODS: A keyword/reference search was performed twice, in January 2021 and May 2022 in MEDLINE/PubMed, EMBASE/…
