A testis-specific E3 ubiquitin ligase complex governs spermiogenesis and male fertility.

The ubiquitin-proteasome system (UPS) represents an evolutionarily conserved machinery governing proteostasis through spatiotemporal regulation of protein degradation. While spermatogenesis involves multilayered regulatory mechanisms spanning translation to dynamic post-translational modifications (PTMs), the identity of UPS-associated E3 ligases orchestrating germ cell-specific protein turnover remains elusive. Here, we identify a testis-specific E3 ubiquitin ligase complex comprising elongin B/C, Cullin-2 (CUL2), RING-box protein-1 (RBX1), and SOCS box protein ASB9, designated ECSASB9. Genet

The Small Ubiquitin-Like Modifier SUMO-3 Acts as a Neutrophil Chemoattractant via the Chemokine Receptors CXCR1 and CXCR2.

INTRODUCTION: Small ubiquitin-like modifiers (SUMOs) are small peptides conjugated to proteins during post-translational modification, which have been reported to modulate several aspects of the immune system, notably in autoimmune disorders. METHODS: We used a SUMO-based bacterial expression system to create a recombinant protein putatively expressed by Toxocara canis, which we hypothesised might antagonise responses via the chemokine receptor CXCR1. RESULTS: Although our recombinant T. canis protein was devoid of antagonist activity, we serendipitously observed that recombinant SUMO-3 prot

AlphaFlex: Ensembles of the human proteome representing disordered regions.

Over a third of residues in the canonical human proteome are predicted to fall within intrinsically disordered protein regions (IDRs), which do not adopt stable folded structures. These IDRs play critical roles in biological regulation and organization, including as targets for post-translational modifications, scaffolds and mediators of biomolecular condensates. To address the pressing need for valid structural models providing biological relevance and enabling functional insight, we developed the AlphaFlex workflow, using IDPConformerGenerator or IDPForge to calculate fully atomistic conform

Epitranscriptomic regulation in the progression of cancer and platinum resistance.

Epitranscriptomic regulation has emerged as a critical mechanism in cancer biology, particularly in the development of chemoresistance. RNA modifications including N6-methyladenosine (m6A), 5-methylcytosine (m5C), N1-methyladenosine (m1A), 7-methylguanosine (m7G), pseudouridine (Ψ), and A-to-I editing dynamically control mRNA stability, splicing, translation, and degradation. RNA-modifying proteins called 'writers,' 'erasers,' and 'readers' regulate post-transcriptional networks to enable tumor adaptation and chemoresistance. In platinum-resistant tumors, epitranscriptomic changes modulate DNA

The cellular pathways that maintain the quality control and transport of diverse potassium channels.

Potassium channels are multi-subunit transmembrane proteins that permit the selective passage of potassium and play fundamental roles in physiological processes, such as action potentials in the nervous system and organismal salt and water homeostasis, which is mediated by the kidney. Like all ion channels, newly translated potassium channels enter the endoplasmic reticulum (ER) and undergo the error-prone process of acquiring post-translational modifications, folding into their native conformations, assembling with other subunits, and trafficking through the secretory pathway to reach their f

Atomic mutagenesis at the ribosomal decoding site.

Ribosomal decoding is an essential process in every living cell. During protein synthesis the 30S ribosomal subunit needs to accomplish binding and accurate decoding of mRNAs. From mutational studies and high-resolution crystal structures nucleotides G530, A1492 and A1493 of the 16S rRNA came into focus as important elements for the decoding process. Recent crystallographic data challenged the so far accepted model for the decoding mechanism. To biochemically investigate decoding in greater detail we applied an in vitro reconstitution approach to modulate single chemical groups at A1492 and A1

Conditions of endoplasmic reticulum stress favor the accumulation of cytosolic prion protein.

After signal sequence-dependent targeting to the endoplasmic reticulum (ER), prion protein (PrP) undergoes several post-translational modifications, including glycosylation, disulfide bond formation, and the addition of a glycosylphosphatidylinositol anchor. As a result, multiple isoforms are generated. Because of the intrinsic weakness of the PrP signal sequence, a fraction of newly synthesized molecules fails to translocate and localizes to the cytosol. The physiopathologic role of this cytosolic isoform is still being debated. Here we have shown that, in both cultured cell lines and primary

Identification of the most damaging nsSNPs in the human CFL1 gene and their functional and structural impacts on cofilin-1 protein.

The cofilin-1 protein, encoded by CFL1, is an actin-binding protein that regulates F-actin depolymerization and nucleation activity through phosphorylation and dephosphorylation. CFL1 has been implicated in the development of neurodegenerative diseases (Alzheimer's disease and Huntington's disease), neuronal migration disorders (lissencephaly, epilepsy, and schizophrenia), and neural tube closure defects. Mutations in CFL1 have been associated with impaired neural crest cell migration and neural tube closure defects. In our study, various computational approaches were utilized to explore singl

METTL3-mediated N6-methyladenosine modification of Dnajb1 modulates cardiomyocyte ferroptosis during myocardial infarction.

Myocardial infarction (MI) affects millions of individuals worldwide, with ferroptosis recognized as a pivotal regulated cell death pathway in this context. N6-methyladenosine (m6A), the most prevalent mRNA modification, is essential in modulating RNA splicing, export, stability, and translation during MI progression. Nonetheless, the involvement of m6A modification in cardiomyocyte ferroptosis has yet to be elucidated. This study aimed to elucidate the regulatory mechanisms of m6A modification in cardiomyocyte ferroptosis by conducting an integrated analysis of methylated RNA immunoprecipitat

PRMT5 promotes psoriatic inflammation through mediating dimethylation of p65 in keratinocytes.

BACKGROUND: Psoriasis is a chronic inflammatory skin disease driven by aberrant keratinocyte activation and immune dysregulation. Protein arginine methyltransferase 5 (PRMT5) was identified as a candidate regulator in psoriatic keratinocytes, yet its function and mechanistic role in psoriasis pathogenesis remain unclear. METHODS: We analyzed multiple public bulk RNA-sequencing and single-cell RNA-sequencing (scRNA-seq) transcriptomic datasets from psoriasis patients to assess PRMT5 mRNA expression and distribution. Protein expression was further validated by immunofluorescence staining of pso

Molecular basis for the regulation of the SUMOylation activity of the Smc5/6 complex by Esc2.

Post-translational modification by SUMO regulates a wide array of cellular processes. Despite the large number of SUMOylated substrates, only a limited set of SUMO E3 ligases have been identified, raising important questions about how substrate specificity is achieved. In budding yeast, the nuclear protein Esc2 functions as a cofactor in the SUMO pathway through its interaction with the E2 enzyme Ubc9. Esc2 is relevant under conditions of replicative stress, promoting the removal of recombination intermediates during DNA replication. This role has been largely attributed to its ability to enha

Research progress in the role and mechanism of GALNT3 in human diseases (Review).

Protein glycosylation is a crucial post-translational modification. Polypeptide N-acetylgalactosaminyltransferase 3 (GALNT3) is a glycosyltransferase that plays an essential role in various human diseases by modifying proteins, including fibroblast growth factor 23 and mucins. In non-tumor conditions, mutations in GALNT3 result in hyperphosphatemia in familial tumoral calcinosis, and its dysregulation has been linked to coronary artery disease. Notably, GALNT3 plays a seemingly opposing role in influenza A virus infection by potentially aiding early viral replication, and later exerting antivi

Molecular characterization of palmitoylation in prostate cancer reveals KIFC2 as a prognostic biomarker and potential therapeutic target.

Protein palmitoylation, a reversible post-translational lipid modification, has been implicated in regulating cancer cell signaling and progression; however, its role in prostate cancer (PCa) remains unclear. In this study, we comprehensively analyzed palmitoylation-related genes (PRGs) in PCa by integrating bulk transcriptomic, single-cell RNA sequencing, and spatial transcriptomic datasets. Unsupervised consensus clustering based on PRG expression identified two molecular subtypes with distinct prognoses, immune infiltration profiles, and pathway activities. A Novel Strategy for Highly Efficient Heterologous Expression of Carbonic Anhydrase in Yarrowia lipolytica.

Carbonic anhydrases (CAs) efficiently catalyze CO2 reversible hydration, critical for carbon capture and sequestration (CCS), but naturally low yield limits industrial use. Yarrowia lipolytica, an unconventional yeast, is an ideal heterologous expression host with robust adaptability, post-translational modification capacity, and versatile genetic tools. In this study, 10 α-, β-, and γ-class CAs were successfully expressed in Y. lipolytica, and two top-performing candidates were identified: Methanosarcina mazei γ-CA (MmaCA) and Sulfurihydrogenibium azorense α-CA (SazCA). PAD4 deletion in synovial macrophages exacerbates pathology in inflammatory arthritis.

Citrullination by peptidylarginine deiminase (PAD) enzymes is a post-translational protein modification implicated in the etiopathogensis of rheumatoid arthritis. Of the five known PAD isoforms, PAD4 is expressed in the nucleus of innate immune cells including synovial macrophages, and studies have shown citrullination of intracellular proteins such as transcription factors and histones can polarize cell phenotype. Here, we report how systemic and local deletion of PAD4 in synovial macrophages from PAD4-/- or PAD4f/fCreLysM mice results in activation of both shared and distinct gene modules in

Drosophila eye development is compromised by dTet overexpression through altered retinal determination gene expression.

Epigenetic regulation via DNA demethylation and histone modification is essential for tissue development. We investigated the role of the Drosophila ten-eleven translocation protein (dTet) in eye development and its interaction with the oncohistone H3.3K27M. dTet overexpression in eye imaginal discs caused strong downregulation of retinal determination genes (toy, ey, so, eya, dac) and developmental arrest, leading to partial or complete eye loss. Enhanced dTet knockdown using sensitized conditions also resulted in eye developmental defects, supporting a dosage-dependent role for dTet during r

Phosphoproteomics of WHO-Priority Fungal Pathogens: Conserved Signaling Architecture, Pathogen-Specific Outputs, and Therapeutic Vulnerabilities.

Protein phosphorylation is a central post-translational modification. In pathogenic fungi, it dynamically governs morphogenesis, stress adaptation, and antifungal drug resistance. Using high-resolution mass spectrometry-based phosphoproteomics, researchers have systematically mapped phosphorylation dynamics in WHO-priority pathogens-Candida albicans, Aspergillus fumigatus, Cryptococcus neoformans, and the multidrug-resistant Candidozyma auris (formerly Candida auris). These studies reveal that thousands of phosphorylation events are coordinately reprogrammed in response to antifungal drug expo

[Histone deacetylases and alcohol-related liver disease].

Alcohol-related liver disease (ALD) is a common chronic liver disease caused by prolonged excessive alcohol consumption. It may initially be asymptomatic or manifest as mild alcoholic fatty liver, then progress to subclinical steatohepatitis characterized by hepatic inflammation, and eventually develop into fibrosis, cirrhosis, liver failure, or even hepatocellular carcinoma. Although alcohol abstinence remains the primary strategy for ALD management, its effectiveness is often limited by patient adherence and social factors, making combined pharmacological intervention necessary in many cases

Expanding the Arsenal of Nanocarriers Against Parkinson Disease: Success so Far Roads Ahead.

Parkinson's disease (PD) is a common progressive neurodegenerative condition involving the degeneration of dopaminergic neurons and the accumulation of misfolded α-synuclein protein. Although standard pharmacotherapies such as levodopa provide symptomatic improvement, their effectiveness is usually compromised by issues with low bioavailability, short half-life, and limited penetration of the blood-brain barrier (BBB). Nanotechnology provides a revolutionary answer by allowing more targeted drug delivery to the brain. A broad range of nanocarriers such as liposomes, polymeric nanoparticles, an
