A highly selective fluorescence sensor for Tin (Sn4+) and its application in imaging live cells.

A naphthalimide-rhodamine B derivative was synthesized as a fluorescence turn-ON chemodosimeter for Sn(4+). A colour change and marked enhancement of fluorescence was found in the presence of Sn(4+), Cu(2+) and Cr(3+) due to the ring open reaction of rhodamine and a fluorescence resonance energy transfer process. Addition of the strong chelating agent ethylenediaminetetraacetic acid disodium salt (EDTA) partly released the cation from the complex with Sn(4+) and restored the yellow fluorescence. In addition, the compound can be used as a fluorescent probe for Sn(4+) in biological systems and m

Fluorescent probe studies of the interactions of 1-alkyl-2-pyrrolidones with stratum corneum lipid liposomes.

Previously, the effects of a series of 1-alkyl-2-pyrrolidones (APs; C2-C8) on the lipoidal pathway of hairless mouse skin (HMS) were studied with a parallel pathway skin model. At their isoenhancement concentrations, these 1-alkyl-2-pyrrolidones induce the same transport enhancement (isoenhancement factor, EHMS) on the lipoidal pathway of the stratum comeum for the probe permeants studied. In the present study, the fluidizing effects of APs upon the stratum comeum lipid liposome (SCLL) bilayer were investigated under these isoenhancement conditions using steady state anisotropy and fluorescenc

Rapid detection of aflatoxin B1 by dummy template molecularly imprinted polymer capped CdTe quantum dots.

A novel and sensitive fluorescent sensor was synthesized for the rapid and specific recognition of aflatoxin B1 (AFB1) by our combining molecular imprinting techniques with quantum dot technology. Molecularly imprinted polymers coated CdTe quantum dots (MIP@CdTe QDs) were prepared through the Stöber method with 5,7-dimethoxycoumarin as a dummy template. 3-Aminopropyltriethoxysilane was selected as the functional monomer, and tetraethyl orthosilicate was used as the cross-linking agent. The best molar ratio of 5,7-dimethoxycoumarin to functional monomer to cross-linker was 4:20:15. The MIP@CdTe

Liposomal vasoactive intestinal peptide for lung application: protection from proteolytic degradation.

Inhalative administration of vasoactive intestinal peptide (VIP) is a promising approach for the treatment of severe lung diseases. However, the clinical use of VIP is limited by the fact that the peptide is prone to rapid degradation mechanisms and proteolytic digestion. Accordingly, VIP exhibits a very short period of activity in the lung. To overcome this problem, we have designed a liposomal drug delivery system for VIP and characterized it in terms of its potential to protect VIP from enzymatic cleavage. The proteolytic conditions of the lung, the target site of aerosolic administered VIP

A mass spectrometry-based assay for improved quantitative measurements of efflux pump inhibition.

Bacterial efflux pumps are active transport proteins responsible for resistance to selected biocides and antibiotics. It has been shown that production of efflux pumps is up-regulated in a number of highly pathogenic bacteria, including methicillin resistant Staphylococcus aureus. Thus, the identification of new bacterial efflux pump inhibitors is a topic of great interest. Existing assays to evaluate efflux pump inhibitory activity rely on fluorescence by an efflux pump substrate. When employing these assays to evaluate efflux pump inhibitory activity of plant extracts and some purified compo

Development of an automated assay for accelerated in vitro detection of DNA adduct-inducing and crosslinking agents.

Current techniques for the identification of DNA adduct-inducing and DNA interstrand crosslinking agents include electrophoretic crosslinking assays, electrophoretic gel shift assays, DNA and RNA stop assays, mass spectrometry-based methods and 32P-post-labelling. While these assays provide considerable insight into the site and stability of the interaction, they are relatively expensive, time-consuming and sometimes rely on the use of radioactively-labelled components, and thus are ill-suited to screening large numbers of compounds. A novel medium throughput assay was developed to overcome th

Dissecting the glycoprofile of the human cytomegalovirus protein gB by comprehensive mass spectrometry analysis.

Glycosylation is the most critical post-translational modification occurring on proteins and its specific pattern depends both on the cells used for their production, such as mammalian cells, insect cells and yeast, and the expression conditions (e.g., mannosidase inhibitors, gene deletion). The aim of this study is to characterize the glycans of recombinant glycoprotein B (gB) from Human Cytomegalovirus (HCMV), expressed in CHO cells, by using different hyphenated mass spectrometry techniques in order to enrich the knowledge about glycosylation pattern changes in the protein production proces

Synthesis of a cyanine-based fluorescent probe and its dual-channel recognition of TNP and PYX in aqueous solution.

A fluorescent probe with excellent biocompatibility and water solubility was constructed, which can achieve highly selective detection of trace 2,4,6-trinitrophenol (TNP) and 2,6-bis(picrylamino)-3,5-dinitropyridine (PYX). The probe exhibits a purple color and red fluorescence in aqueous solution, and can specifically and rapidly recognize TNP and PYX through a dual-channel mode of naked-eye color change and fluorescence quenching, showing good detection sensitivity. Spectroscopic test results indicated that the limits of detection (LOD) of TNP and PYX measured by UV-vis absorption spectroscop

A new quinolinium-based colorimetric and NIR fluorescence dual response probe for bisulfite recognition and its application in real-world samples and cell imaging.

Sulfur dioxide (SO₂) has long been recognized as an environmental pollutant, and excessive intake of SO₂ and its derivatives can cause adverse effects on human health. In this work, three near-infrared (NIR) fluorescence turn-on probes (QT-BT, QT-BI and QT-PT) for HSO₃- detection were designed and synthesized. Among the probes, QT-PT exhibited superior overall HSO₃- recognition performance. In PBS:DMF (7:3, v/v, pH = 7.4) buffer solution, QT-PT enabled dual-channel recognition of HSO₃- through a distinct color change (from Light yellow to pink) and a remarkable fluorescence turn-on response (6

Photoactivated fluorescent nanozymes from mulberry leaf-derived carbon dots for dual-mode antioxidant determination and biothiol contribution estimation.

Contribution-resolved antioxidant determination in complex food matrices remains challenging because the total antioxidant response usually arises from multiple overlapping reducing species. Herein, red-emissive carbon dots (CDs) were prepared from mulberry leaves via a solvothermal method and developed as highly efficient photoactivated fluorescent nanozymes for dual-mode antioxidant determination. The CDs exhibited a large Stokes shift (Δλ = 272 nm) and were stably dispersed in aqueous media with the assistance of Tween-20. Under 365 nm irradiation, the CDs showed oxidase-like activity towar

A selective near-infrared fluorescent probe for detecting hydrogen peroxide in Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with growing incidence among younger populations, highlighting the urgent need for early and accurate detection of AD-related biomarkers. Excessive production of reactive oxygen species (ROS), particularly hydrogen peroxide (H2O2), induces oxidative stress, leading to neuronal damage and death, which implicates H2O2 as a potential biomarker for AD. In this study, leveraging the advantages of fluorescence imaging, we developed a novel near-infrared (NIR) fluorescent probe, LDMD-B, capable of selectively detecting H2O2 through

Construction of an ion-imprinted ratiometric fluorescence sensor integrating zirconium-based metal-organic framework and carbon dots for sensitive detection of copper ion.

The pollution of Cu2+ has seriously endangered human health and water ecological environment. It is crucial to explore a rapid and sensitive fluorescence detection method for monitoring Cu2+. In present work, an ion-imprinted functionalized zirconium-based metal-organic framework (NH2-UiO-66@IIPs) was innovatively designed and prepared, and yellow fluorescent carbon dots (CDs) were fabricated with o-phenylenediamine and tetraphenylethylene via a hydrothermal method. The experimental results demonstrated that Cu2+ could quench the fluorescence intensity of NH2-UiO-66@IIPs, and the fluorescence

Selective ratiometric fluorescent detection of chlortetracycline in environmental water based on hydrolysis-induced emission.

For specific determination of chlortetracycline (CTC) among tetracycline antibiotics (TCs), the current study focused on the hydrolysis effect of TCs. In water, CTC was hydrolyzed into isochlortetracycline (iso-CTC) along with blue fluorescence, but other TCs remained stable without blue fluorescence. CTC was thus identified within TCs based on hydrolysis-induced emission. To improve signal stability, Mn-doped ZnS (Mn:ZnS) quantum dots (QDs) that exhibited red luminescence were introduced to achieve ratiometric sensing. After adding CTC to Mn:ZnS QDs, the red emission of Mn:ZnS QDs was decreas

A versatile sensor capable of ratiometric fluorescence detection of trace water and turn-on detection of Cu2+ modulating the binding interaction of a Cu(II) complex with BSA and DNA complemented by docking studies.

A fluorescent molecule, pyridine-coupled bis-anthracene (PBA), has been developed for the selective fluorescence turn-on detection of Cu2+. Interestingly, the ligand PBA also exhibited a red-shifted ratiometric fluorescence response in the presence of water. Thus, a ratiometric water sensor has been utilized as a selective fluorescence turn-on sensor for Cu2+, achieving a 10-fold enhancement in the fluorescence and quantum yield at 446 nm, with a lower detection limit of 0.358 μM and a binding constant of 1.3 × 106 M-1. For practical applications, sensor PBA can be used to detect Cu2+ in vario

Detection of silver through amplified quenching of fluorescence from polyvinyl pyrrolidone-stabilized copper nanoclusters.

Silver ion detection with ultra-high sensitivity was established. We synthesized copper nanoclusters (CuNCs) with blue fluorescence through a one-pot process. Instead of a direct quencher toward the CuNCs, silver ions activated the strong oxidation from persulfate and subsequently converted divalent manganese ion into manganese dioxide (MnO2). The surface charges of MnO2 and the CuNCs brought them together and quenched the fluorescence from the latter. Due to silver ions' role as the catalyst in the process, it cycled and even a small amount leads to a significant fluorescence change. This sig

Dimethylamino naphthalene-based fluorescent probes for hydrogen sulfide detection and living cell imaging.

Hydrogen sulfide shows great importance in various physiological and biochemical processes. The development of fluorescence probes for facile and efficient detection of H2S has attracted increasing attention of researchers. Herein, we synthesized two fluorescence probes based on simple naphthalene structure for detection of H2S. Upon reaction with H2S, the probe DN-DM exhibited a red fluorescence emission with large Stokes shift. The probe showed high sensitivity, pH insensitivity and good selectivity for H2S over other analytes including common biothiols. The detection mechanism was based on

Dihydroxy-Substituted Coumarins as Fluorescent Probes for Nanomolar-Level Detection of the 4-Amino-TEMPO Spin Label.

This paper reports on dihydroxycoumarins as fluorescent probes suitable for the detection and determination of the nitroxide radical, namely 4-amino-TEMPO. Since 4-amino-TEMPO is used as a spin label for the detection of various radicals and damage caused by these species, its determination under physiological conditions might help us to understand the mechanism of the oxidative stress. Among different coumarins studied, only dihydroxy-substituted derivatives show high sensitivity, specificity, and selectivity for the nitroxide radical. In this assay, dihydroxy-substituted coumarins under the

Synthesis and evaluation of a novel 'off-on' chemical sensor based on rhodamine B and the 2,5-pyrrolidinedione moiety for selective discrimination of glutathione and its bioimaging in living cells.

A new "turn-on" fluorescent probe, RDMBM, based on the rhodamine B dye and the 2,5-pyrrolidinedione moiety was synthesized and characterized. Its sensing behavior toward various amino acids was evaluated via UV-vis and fluorescence spectroscopic techniques. The observed spectral changes showed that RDMBM displays high selectivity and sensitivity toward GSH in MeOH/H2O (1:2, v/v, pH 7.40, Tris-HCl buffer, 1 mM) solution and that it undergoes 1:1 covalent binding with GSH. More importantly, the hydrogenation and ring-opening of the nitrogen atom in the spirane structure of rhodamine B derivative

Development of large Stokes shift, near-infrared fluorescence probe for rapid and bioorthogonal imaging of nitroxyl (HNO) in living cells.

Nitroxyl (HNO), as an electron reduced and protonated form of nitric oxide, is emerging as a potential diagnostic and therapeutic biomarker. It is still of great interest to develop probes of desirable properties to study its biological functions. Here we develop a near infrared fluorescence probe for detecting and visualizing exogenous and endogenous HNO in living cells. The probe is designed by coupling a HNO-responsive moiety, diphenylphosphinobenzoyl group, with a near infrared fluorophore with large of Stokes shift via an ester linker.
