Endogenous acetylcholine concentrations in rat iris-ciliary body.

The technique of microwave irradiation, whereby enzymes are inactivated within milliseconds, was employed to accurately determine the in vivo acetylcholine (ACh) concentration in the iris-ciliary body (ICB). Rats were sacrificed by microwave irradiation at 9.5 kW at 2,450 MHz for 0.9 seconds. The eyes were enucleated, the ICBs isolated and 10 ICBs from 10 eyes were treated as one sample. ACh levels were determined using high performance liquid chromatography in an immobilized-enzyme reactor. Endogenous ACh levels in the rat ICB were 58.8 +/- 5.0 nmoles (8.58 +/- 0.73 micrograms) per gram tissu

Autoimmune human T lymphocytes specific for acetylcholine receptor.

Myasthenia gravis is one of the best characterized human autoimmune disorders. Circulating autoantibodies to the nicotinic acetylcholine receptor (AChR) at the neuromuscular junction play a prominent part in the effector phase, that is, the immunoregulation. Indirect evidence, such as thymic abnormalities and the association with certain histocompatibility antigens (for example HLA-B8,-DR3) suggests a defect of immunoregulation at the level of thymus-dependent (T) lymphocytes. We report here on the isolation of autoreactive T cells from six patients with myasthenia gravis. From one of these pa

EFFECTS OF CENTRAL DEPRESSANT DRUGS UPON ACETYLCHOLINE RELEASE.

Several central depressant and other drugs have been examined for their effects upon acetylcholine release from the stimulated, perfused cat superior cervical ganglion and rat isolated phrenic nerve-diaphragm preparations. The acetylcholine released was assayed biologically. Amylobarbitone sodium, chloral hydrate, trichloroethanol, methylpentynol, methylpentynol carbamate, paraldehyde, procaine hydrochloride and troxidone reduced the presynaptic release of acetylcholine from the ganglion. They also exhibited a postsynaptic blocking action, this component of depressant activity being particular

Ikarisoside A inhibits acetylcholine-induced catecholamine secretion and synthesis by suppressing nicotinic acetylcholine receptor-ion channels in cultured bovine adrenal medullary cells.

Ikarisoside A is a natural flavonol glycoside derived from plants of the genus Epimedium, which have been used in Traditional Chinese Medicine as tonics, antirheumatics, and aphrodisiacs. Here, we report the effects of ikarisoside A and three other flavonol glycosides on catecholamine secretion and synthesis in cultured bovine adrenal medullary cells. We found that ikarisoside A (1-100 μM), but not icariin, epimedin C, or epimedoside A, concentration-dependently inhibited the secretion of catecholamines induced by acetylcholine, a physiological secretagogue and agonist of nicotinic acetylcholi

T-cell recognition of acetylcholine receptor provides a reliable means for monitoring autoimmunity to acetylcholine receptor in antibody-negative myasthenia gravis patients.

Myasthenia gravis (MG) is an autoimmune disease usually associated with autoantibodies (auto-Abs) against nicotinic acetylcholine receptor (AChR). Some MG patients appear negative for anti-AChR Abs (seronegative), and a fraction of these have auto-Abs against muscle-specific kinase. The remaining patients, although displaying MG symptoms, show no detectable auto-Abs. We describe here a possible association of a rare human leukocyte antigen (HLA)-DQ type and AChR Ab-negative MG. We also found that the majority of seronegative patients exhibit an anti-AChR autoimmune T lymphocyte response. We in

Crosslinking of proteins in acetylcholine receptor-rich membranes from Torpedo californica: relation of 43-kD protein and Torpedo dystrophin to acetylcholine receptor.

We examined the spatial relation of 43-kD protein and Torpedo dystrophin, which are cytoplasmic peripheral membrane proteins in the nicotinic acetylcholine receptor (AChR)-rich membranes, to AChR. We used three kinds of the heterobifunctional crosslinking reagents to crosslink proteins in the AChR-rich membranes. Products crosslinked by SMPB (14.5 A span) including 43-kD protein and Torpedo dystrophin appeared at the tops of the stacking gels at the concentrations of 8.89 x 10(-5)M to 8.89 x 10(-3)M SMPB. High molecular weight materials (crosslinked products) increased with increasing concentr

Tetrandrine Alleviates Allergic Rhinitis via REV-ERBα-Dependent Acetylcholine.

Allergic rhinitis is a prevalent chronic inflammatory disorder of the nasal mucosa, imposing substantial medical and socioeconomic burdens. Thus, there is an urgent need to develop novel agents capable of both symptom relief and immune modulation. This study elucidates the mechanism by which tetrandrine alleviates allergic rhinitis through REV-ERBα activation and cholinergic pathway suppression. In ovalbumin-induced allergic rhinitis mice, intranasal tetrandrine significantly reduced sneezing frequency and nasal scratching versus controls, while decreasing serum histamine and substance P level

Methods for analyzing acetylcholine-induced vasodilation in human skin.

Intradermal microdialysis, combined with laser-Doppler flowmetry and local heating, is commonly used to investigate mechanisms of endothelial function and dysfunction in the cutaneous microcirculation. Local perfusion of acetylcholine (ACh), with and without NG-nitro-l-arginine methyl ester [l-NAME; a nonselective nitric oxide (NO) synthase inhibitor], allows for direct assessment of pharmacologically induced endothelium- and NO-dependent vasodilation. However, variability in the analysis and reporting of cutaneous vascular conductance (CVC) responses to in vivo cholinergic stimulation limits

Non-neuronal acetylcholine: from cellular signaling to clinical implications.

For over a century, acetylcholine (ACh) was viewed primarily as a wired neurotransmitter of cholinergic nerves. However, accumulating evidence supports the view of the non-neuronal cholinergic system (NNCS) as an evolutionarily ancient autocrine and paracrine network operating across peripheral tissues. Rather than an extension of neuronal signaling, non-neuronal ACh can be viewed as a tissue-embedded chemical communication system positioned at the interface of cellular homeostasis and systemic neural reflexes. This review frames the NNCS as a network of tissue-resident ACh-producing cells tha

Drug Discovery Targeting Nicotinic Acetylcholine Receptors for Hearing Loss.

Hearing loss is detrimental to human health, and currently, more than 1.5 billion people are affected by hearing loss. Active military personnel and construction workers are examples of individuals in the workplace who are exposed to loud noise and are at serious risk of hearing loss. While there is currently no therapy for hearing loss, evidence supports investigating the enhancement of the Medial Olivocochlear (MOC) system, an efferent pathway for hearing that serves as a gain-control for hearing loss protection. Selectively modulating the α9α10 nicotinic acetylcholine receptor (nAChRs) foun

Muscarinic Acetylcholine Receptor Activators as Transformative Therapeutics for Schizophrenia.

While traditional antipsychotic drugs provide symptomatic relief for positive symptoms in patients with schizophrenia, many patients are refractory to traditional antipsychotics. Furthermore, these medicines are ineffective in treating negative and cognitive symptoms and have serious adverse effects that limit their utility. Traditional antipsychotics act as antagonists or partial agonists of D2 dopamine receptors, an action that is key to their antipsychotic efficacy and adverse effects. Muscarinic acetylcholine receptor (mAChR) agonists have now emerged as the first truly novel treatments fo

α4 Nicotinic Acetylcholine Receptors in Lipopolysaccharide-Related Lung Inflammation.

Sepsis remains an important healthcare challenge. The lungs are often affected in sepsis, resulting in acute lung injury characterized by inflammation. Mechanisms involving lipopolysaccharide (LPS) stimulation of toll-like receptor (TLR) signaling with induction of proinflammatory pathways have been implicated in this process. To date, however, studies targeting these pathways have failed to improve outcomes. We have found that LPS may also promote lung injury through the activation of α4 nicotinic acetylcholine receptors (α4 nAChRs) in immune cells. We observed increased expression of α4 nACh

Corneal acetylcholine regulates sensory nerve activity via nicotinic receptors.

PURPOSE: Sensory nerve terminals are highly distributed in the cornea, and regulate ocular surface sensation and homeostasis in response to various endogenous and exogenous stimuli. However, little is known about mediators regulating the physiological and pathophysiological activities of corneal sensory nerves. The aim of this study was to investigate the presence of cholinergic regulation in sensory nerves in the cornea. METHODS: Localization of choline acetyltransferase (ChAT) and vesicular acetylcholine transporter (vAChT) was evaluated using western blotting and immunohistochemical analys

Muscarinic Acetylcholine Receptor Agonists as Novel Treatments for Schizophrenia.

Schizophrenia remains a challenging disease to treat effectively with current antipsychotic medications due to their limited efficacy across the entire spectrum of core symptoms as well as their often burdensome side-effect profiles and poor tolerability. An unmet need remains for novel, mechanistically unique, and better tolerated therapeutic agents for treating schizophrenia, especially those that treat not only positive symptoms but also the negative and cognitive symptoms of the disease. Almost 25 years ago, the muscarinic acetylcholine receptor (mAChR) agonist xanomeline was reported to r

Acetylcholine receptor subunit expression in Huntington's disease mouse muscle.

Huntington's disease (HD) causes neurological impairments, as well as muscle dysfunction, including smaller neuromuscular junctions (NMJs). This study assessed the expression levels of the subunits of the nicotinic acetylcholine receptor (nAChR) in muscles of the R6/2 mouse model of HD. Based on our previous findings of reduced NMJ size in R6/2 mice, it was hypothesized that muscles from R6/2 mice would also show an altered expression pattern of nAChR subunits compared to wild-type (WT) mice. Therefore, the mRNA levels of nAChR subunits were quantified in R6/2 and WT mouse muscles using qRT-PC

CAKUT and Autonomic Dysfunction Caused by Acetylcholine Receptor Mutations.

Congenital anomalies of the kidney and urinary tract (CAKUT) are the most common cause of chronic kidney disease in the first three decades of life, and in utero obstruction to urine flow is a frequent cause of secondary upper urinary tract malformations. Here, using whole-exome sequencing, we identified three different biallelic mutations in CHRNA3, which encodes the α3 subunit of the nicotinic acetylcholine receptor, in five affected individuals from three unrelated families with functional lower urinary tract obstruction and secondary CAKUT. Four individuals from two families have additiona

Detection of Acetylcholine Receptor Blocking Antibodies by Flow Cytometry.

OBJECTIVES: Detection of acetylcholine receptor (AChR) blocking antibodies through the use of a radiolabel has become standard procedure in most laboratories. Known drawbacks associated with radioassay, including cost of radioisotopes, hazards to laboratory professionals, and manufacture and disposal of radioactive materials, have prompted investigation into replacement assays. We describe here a high-throughput immunofluorescent flow cytometric assay designed for the detection of AChR blocking antibodies. METHODS: In total, 323 serum samples were tested on both the AChR blocking radioassay a
