OphthalMimic: A new alternative apparatus without animal tissue for the evaluation of topical ophthalmic drug products.

The necessity of animal-free performance tests for novel ophthalmic formulation screening is challenging. For this, we developed and validated a new device to simulate the dynamics and physical-chemical barriers of the eye for in vitro performance tests of topic ophthalmic formulations. The OphthalMimic is a 3D-printed device with an artificial lacrimal flow, a cul-de-sac area, a support base, and a simulated cornea comprised of a polymeric membrane containing poly-vinyl alcohol 10 % (w/v), gelatin 2.5 % (w/v), and different proportions of mucin and poloxamer, i.e., 1:1 (M1), 1:2 (M2), and 2:1

Development of ophthalmic nanoemulsions of β-caryophyllene for the treatment of Acanthamoeba keratitis.

Although rare, amoebic keratitis (AK) is a disease caused by Acanthamoeba spp. that can lead to blindness. The drugs currently available for its treatment are very toxic, which has motivated the investigation for more effective and safe therapeutic options. In this study, the in vitro activity of ß-caryophyllene (BCP) was exploited taking into account its action against other protozoans as well as its well-known healing and anti-inflammatory properties (aspects relevant for the AK pathogenesis). On the other hand, high volatilization and oxidation phenomena are found for this compound, which l

An insight on ophthalmic drug delivery systems: Focus on polymeric biomaterials-based carriers.

Presently, different types of eye diseases, such as glaucoma, myopia, infection, and dry eyes are treated with topical eye drops. However, due to ocular surface barriers, eye drops require multiple administrations, which may cause several risks, thereby necessitating additional strategies. Some of the key characteristics of an ideal ocular drug delivery system are as follows: (a) good penetration into cornea, (b) high drug retention in the ocular tissues, (c) targetability to the desired regions of the eye, and (d) good bioavailability. It is worthy to note that the corneal epithelial tight ju

London 2012: prescribing for athletes in ophthalmology.

AIMS: Prescribing for athletes requires an up-to-date knowledge of the World Anti-Doping Agency's list of prohibited substances. As the London 2012 Olympic Games attract athletes from around the world, we review the current guidelines with respect to all medications licensed for ophthalmic use in the United Kingdom. We describe the process that an ophthalmologist can use to check for permissible medications and also highlight treatments that are contraindicated. METHODS: We systematically reviewed all 77 drugs listed in Section 11 of the British National Formulary (Issue 63) for use in the tr

Exosomes in the treatment of age-related ophthalmic diseases: an updated review.

Exosomes are nanoscale extracellular vesicles of endosomal origin that mediate intercellular communication by transferring bioactive cargo, including proteins, lipids, messenger RNAs, microRNAs, and other regulatory molecules. Through this process, they can modulate the biological functions of recipient cells. Age-related ophthalmic diseases, including age-related macular degeneration (AMD), dry eye disease (DED), and age-related cataract (ARC), are degenerative ocular conditions whose incidence increases with age and are associated with chronic inflammation, oxidative stress, dysregulated ang

Cardiac safety of ophthalmic timolol.

It is generally believed that topical administration of eye drops safeguards against harmful systemic effects. However, about 80% of the drug in the ophthalmic products is systemically absorbed and the first-pass metabolism is avoided. Ophthalmic timolol is widely prescribed in the treatment of glaucoma either alone or in the combination eye drop products, many of which have been launched fairly recently. Ophthalmic timolol may cause serious adverse effects such as symptomatic bradycardia, various conduction disorders in the heart, orthostatic hypotension, syncope and falls. Areas covered: In

Selection Criteria for Oils, Surfactants, and Co-Surfactants in Ocular Nanoemulsion Formulation: A Mini Review.

The ocular nanoemulsions (NE) are biphasic systems mainly composed of oil and water emulsified by surfactants/cosurfactants. The extensive surface area of ocular NE enhances corneal contact, leading to improved drug penetration and making it a preferable delivery system. They can also increase the solubility of drugs across the ocular barrier with improved residence time. Oils, surfactants, and co-surfactants used in formulating ocular NEs present a significant challenge in developing safe, stable, less irritant, more permeable, improved residence time, and highly bioavailable products. Autologous serum eye drops for ocular surface disorders.

PURPOSE OF REVIEW: Ocular surface disease is a prevalent, diverse group of conditions that cause patient discomfort and decreased visual acuity and present considerable expense to both patients and healthcare systems. Autologous serum eye drops are a topical treatment modality derived from the patient's own blood. Use of serum eye drops for ocular surface disease has been promising due to biochemical similarities to endogenous tears. RECENT FINDINGS: Use of serum eye drops for moderate to severe ocular surface diseases such as dry eye, corneal epithelial defects, and inflammatory conditions h
